Verfahren zur Herstellung von Retinin
Zur Überführung von Vitamin A in Retinin (Vitamin-A-aldehyd) ist bisher eine einzige Methode bekannt; sie besteht in der Oxydation des Vitamins A mittels aktivem Mangandioxyd in einem indifferenten Lösungsmittel (zum Beispiel Petroläther). Diese Oxydation verläuft langsam und dauert 6-8 Tage. Durch Adsorption des Oxydationsproduktes am Manganoxydniederschlag treten dabei erhebliche Verluste an Retinin ein.
Es wurde nun ein neues Verfahren zur Herstellung von Retinin durch Oxydation von Vitamin A gefunden, welches glatt verläuft und eine gute Ausbeute an reinem Retinin ergibt. Dieses neue Verfahren besteht darin, dass man eine Lösung von Vitamin A in einem oxydationsunempfindlichen, organischen Lösungsmittel, vorzugsweise Eisessig, in Gegenwart eines Platinkatalysators bei Raumtemperatur mit Sauerstoff schüttelt. Innerhalb weniger Stunden wird die zur Oxydation des Vitamin-A-Alkohols zum Aldehyd notwendige Menge Sauerstoff aufgenommen und die Sauerstoffaufnahme hört auf. Aus der Lösung kann der gebildete Aldehyd nach Abtrennen des Katalysators leicht, zum Beispiel durch chromatographisches Reinigen des Eindampfrückstandes, isoliert werden.
Beispiel
406 mg Platinoxyd werden in einer Schüttelente mit wenig Eisessig übergossen und mit Wasserstoff aushydriert (etwa eine Stunde). Hierauf evakuiert man den Wasserstoff und ersetzt ihn durch vorsichtiges Einleiten von Luft. Durch wiederholtes Evakuieren und neues Einströmen von Luft wird die Apparatur von Wasserstoff vollkommen befreit und hierauf mit reinem Sauerstoff gefüllt. Man gibt eine Lösung von 1 g Vitamin-A-Alkohol in wenig Eisessig zu und beginnt zu schütteln, wodurch die Sauerstoffaufnahme einsetzt. Nach 6l/2 Stunden ist fast genau ein halbes Mol Sauerstoff pro Mol Vitamin A aufgenommen und die Oxydation kommt zum Stillstand.
Man entfernt den Sauerstoff im Vakuum, füllt die Apparatur mit Luft, filtriert das Platin ab, dampft im Vakuum ein, löst den Rückstand in Petroläther und chromatographiert an einer Aluminiumoxydsäule (nach Brockmann, durch Zugabe von 10 0/o Wasser ser abgeschwächt).
Die gelbe Hauptzone des Chromatogramms ergibt nach dem Eluieren ein bräunlichgelbes Ö1, welches in wenig Hexan (oder Petroläther) gelöst und in einer Kältemischung von Aceton/Kohlendioxydschnee zur Kristallisation gebracht wird. Nach 1-2 Tagen hat sich das Retinin kristallisiert ausgeschieden. Obwohl der Schmelzpunkt dieser Kristalle noch zu tief liegt, zeigen sie schon das richtige Absorptionsspektrum des Retinins (Absorptionsmaximum bei 368, 5m, ElC% 1672, in Petroläther vom Siedepukt 30-60 ). Durch zwei weitere Kristallisationen steigt der Schmelzpunkt der Retininkristalle auf 61-62.
Ausbeute an reinem Retinin: 200 mg. Aus den Mutterlaugen der Kristallisation kann man weitere Mengen des Retinins in Form seines 2,4- Dinitrophenylhydra- zons gewinnen. Dieses Dinitrophenylhydrazon schmilzt nach dem Umkristallisieren aus einem Aceton/Alkohol-Gemisch bei 206-207 .

Verfahren zur Herstellung von Retinin durch Oxydation von Vitamin A, dadurch gekennzeichnet, dass man eine Lösung von Vitamin A in einem oxydationsunempfindlichen, organischen Lösungsmittel in Gegenwart eines Platinkatalysators bei Raumtemperatur mit Sauerstoff schüttelt.

Process for the production of retinin
A single method is currently known for converting vitamin A into retinin (vitamin A aldehyde); it consists in the oxidation of vitamin A by means of active manganese dioxide in an inert solvent (for example petroleum ether). This oxidation is slow and lasts 6-8 days. Due to the adsorption of the oxidation product on the manganese oxide precipitate, considerable losses of retinin occur.
A new process has now been found for the production of retinin by oxidation of vitamin A, which proceeds smoothly and gives a good yield of pure retinin. This new process consists in shaking a solution of vitamin A in an oxidation-resistant, organic solvent, preferably glacial acetic acid, in the presence of a platinum catalyst at room temperature with oxygen. Within a few hours, the amount of oxygen required to oxidize the vitamin A alcohol to the aldehyde is absorbed and the oxygen absorption ceases. The aldehyde formed can easily be isolated from the solution after the catalyst has been separated off, for example by chromatographic purification of the evaporation residue.

406 mg of platinum oxide are poured with a little glacial acetic acid in a shaking duck and hydrogenated with hydrogen (about an hour). The hydrogen is then evacuated and replaced by carefully introducing air. The apparatus is completely freed of hydrogen by repeated evacuation and fresh influx of air and then filled with pure oxygen. A solution of 1 g of vitamin A alcohol in a little glacial acetic acid is added and shaking begins, causing oxygen uptake to begin. After 6 1/2 hours, almost exactly half a mole of oxygen per mole of vitamin A has been absorbed and the oxidation comes to a standstill.
The oxygen is removed in vacuo, the apparatus is filled with air, the platinum is filtered off, evaporated in vacuo, the residue is dissolved in petroleum ether and chromatographed on an aluminum oxide column (according to Brockmann, weakened by adding 10 0 / o water).
After elution, the yellow main zone of the chromatogram gives a brownish-yellow oil which is dissolved in a little hexane (or petroleum ether) and crystallized in a cold mixture of acetone / carbon dioxide snow. After 1-2 days, the retinin precipitated in crystallized form. Although the melting point of these crystals is still too low, they already show the correct absorption spectrum of retinin (absorption maximum at 368.5m, ElC% 1672, in petroleum ether with a boiling point of 30-60). Two further crystallizations increase the melting point of the retinin crystals to 61-62.
Yield of pure retinin: 200 mg. Further amounts of retinin in the form of its 2,4-dinitrophenylhydrazone can be obtained from the mother liquors from the crystallization. This dinitrophenylhydrazone melts at 206-207 after recrystallization from an acetone / alcohol mixture.
