Diese Verbindung zeigt eine hervorragende di- uretische und natriuretische Wirkung. Dies geht aus Vergleichsversuchen mit dem im Handel erhältlichen Chlorothiazid (6-Chlor-7-sulfamyl-1,2,4-benzothiadi- azin-1,1-dioxyd) hervor. Akute Versuche von sechs stündiger Dauer am Hund zeigen die nachstehenden Ergebnisse, wobei der neuen Verbindung die Be zeichnung Su-5879 zugeordnet wurde.

Aus dieser Tabelle ergibt sich, dass die diuretische Wirkung der neuen Verbindung am Hund we sentlich besser ist als diejenige von Chlorothiazid. Bei einem weiteren Vergleich der diuretischen Wirkung der neuen Verbindung mit Chlorothiazid erhielten Ratten, denen seit 18 Stunden die Nahrung entzogen war, 0,2o/oige Natriumchloridlösung in einer Menge, die<B>5019</B> ihres Körpergewichtes ent sprach. Unmittelbar anschliessend wurde ihnen je eine dieser Verbindungen, gelöst in 0,1 ems verdünn ter Natronlauge, oral gegeben.
Der spontan entleerte Urin wurde in graduierten Zylindern während der nächsten drei Stunden gesammelt, sein Volumen ge messen und in Prozenten der Flüssigkeitszufuhr um gerechnet. Die Konzentrationen von Natrium und Kalium wurden flammenphotometrisch bestimmt und die Gesamtausscheidung dieser Elektrolyse während der dreistündigen Versuchsperiode berechnet. Die in der folgenden Tabelle zusammengefassten Werte sind Durchschnittswerte von 8-12 Ratten pro Gruppe.

Die neue Verbindung besitzt auch bei Anwen dung am Menschen eine starke diuretische und natri- uretische Wirkung, während die Kaliumausscheidung nicht in gleichem Masse gefördert wird.
Bei Patien ten mit Flüssigkeits- und Kochsalzretention infolge Erkrankung des Herzens, der Leber (Cirrhose) oder der Nieren (Nephrose) bewirkt die Verbindung eine therapeutisch erwünschte Förderung der Flüssig- keits- und Kochsalzausscheidung, ohne dass gleich zeitig Gefahr einer Kaliumverarmung besteht. Die für eine derartige Wirkung erforderlichen Dosen bei peroraler Applikation liegen durchschnittlich zwi schen 25 und 100 mg pro Tag.
Die neue Verbin dung ist auch in der Lage, den arteriellen Hoch druck zu senken, vor allem, wenn sie gleichzeitig mit einem andern blutdrucksenkenden Mittel gegeben wird.
Die neue Verbindung soll als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden. Das erfindungsgemässe Verfahren zur Herstel lung der neuen Verbindung ist dadurch gekennzeich net, dass man 5-Chlor-2,4-disulfamyl-anilin oder ein Salz davon mit Formaldehyd oder einem funk tionellen Derivat desselben umsetzt. Vorzugsweise nimmt man die Reaktion in Gegenwart einer Säure, wie einer Mineralsäure, beispielsweise einer Halogen wasserstoffsäure, z. B. Salzsäure oder Bromwasser stoffsäure, oder Schwefelsäure, gegebenenfalls in wasserfreier Form, vor. Der Formaldehyd kann auch in Form eines seiner Polymeren, z.
B. als Paraform- aldehyd, Trioxan oder Hexamethylentriamin einge- setzt werden. Als funktionelle Derivate können Ace tale, z. B. Dimethoxymethan oder Diäthoxymethan, verwendet werden. Die Reaktion wird in erster Linie mit ungefähr äquivalenten Mengen der Reaktions komponenten durchgeführt. Durch höhere Mengen an Aldehyd werden die Ausbeuten infolge einer Re aktion mit der Sulfamylgruppe in 4-Stellung ver mindert. Die Reaktion lässt sich in Abwesenheit oder vorzugsweise in Gegenwart von Lösungsmitteln, wie eines Äthers, z.
B. p-Dioxan oder Diäthylenglykol- dimethyläther, oder eines Formamids, z. B. Dimethyl- formamid, bei Raum- oder erhöhter Temperatur und bei normalem oder erhöhtem Druck oder in Gegenwart eines inerten Gases, wie Stickstoff, durch führen.
Je nach den Reaktionsbedingungen erhält man die neue Verbindung in freier Form oder in Form ihrer Salze. Erhaltene Metallsalze können zum Bei spiel durch Reaktion mit wässrigen sauren Mitteln, wie Mineralsäure, z. B. Halogenwasserstoffsäure, beispielsweise Salzsäure, oder Schwefelsäure, in die freie Verbindung übergeführt werden.
Diese wie derum lässt sich in die Metallsalze, wie Alkalimetall- salze, überführen durch Behandeln zum Beispiel mit einem Metallhydroxyd, wie Natrium- oder Kalium hydroxyd, in einem Lösungsmittel, wie einem Alka- nol, z. B.
Methanol oder Äthanol, oder Wasser und anschliessendem Abdampfen des Lösungsmittels, oder durch Reagierenlassen der freien Verbindung in einem Äther, wie p-Dioxan oder Diäthylenglykol- dimethyläther, mit einem Alkalimetallhydrid oder -amid, z. B. Natrium- oder Kaliumhydrid oder -ainid.
In den nachfolgenden Beispielen sind die Tem peraturen in Celsiusgraden angegeben. <I>Beispiel l</I> Man erhitzt eine Mischung von 2,9 g 5-Chlor- 2,4-dis,tilfamyl-anilin in 15 cm3 wasserfreiem Di- äthylenglykol-dimethyläther, 0,5 cm3 Essigsäureäthyl- ester, enthaltend 109,5 g Chlorwasserstoffsäure pro 1000 cm3, und 0,33 g Paraformaldehyd auf 80 bis 90 und hält eine Stunde bei dieser Temperatur.
Man kühlt auf Raumtemperatur ab, engt die Reak tionsmischung unter vermindertem Druck auf ein Drittel ihres Volumens ein und verdünnt mit Was ser. Das auskristallisierte Produkt wird abfiltriert und aus Wasser umkristallisiert. Man erhält so das 6 - Chlor - 7 - sulfamyl - 3,4 - dihydro-1,2,4-benzothiadi- azin-l,l-dioxyd, das bei 226-268 schmilzt.
Ersetzt man in obigem Beispiel den Paraform- aldehyd durch 0,84 g Dimethoxymethan und ver fährt im übrigen wie oben beschrieben, so erhält man die nämliche Verbindung.
<I>Beispiel 2</I> Eine Mischung von 2,9 g 5-Chlor-2,4-disulfamyl- anilin in 15 cm3 Diäthylenglykol - dimethyläther, 0,8 cm3 einer 37o/oigen wässrigen Formaldehyd lösung (0,3g Formaldehyd) und 0,5 cm3 einer ge sättigten Salzsäure-Essigsäureäthylesterlösung wird eine Stunde auf 80-90 erhitzt.
Nach dem Eindamp fen unter vermindertem Druck und Zufügen von Wasser kristallisiert das 6-Chlor-7-sulfamyl-3,4-di- hydro-1,2,4-benzothiazin-1,1-dioxyd. Es wird aus Wasser umkristallisiert und schmilzt bei 271-275 . <I>Beispiel 3</I> Eine Mischung von 2,9 g 5-Chlor-2,4-disulf- amylanilin, 0,3 g Paraformaldehyd und 15 cm3 Di- äthylenglykol-dimethyläther wird eine Stunde auf 80 bis 90 erhitzt.
Man engt die Reaktionsmischung unter vermindertem Druck ein, fügt Wasser zu, wor auf das 6-Chlor-7-sulfamyl-3,4-dihydro-1,2,4-benzo- thiadiazin-1,1-dioxyd langsam kristallisiert. Es wird abfiltriert und aus Wasser umkristallisiert. Eine Mischprobe mit dem nach Beispiel 1 erhaltenen Produkt beginnt bei 263 zu schmelzen.
<I>Beispiel 4</I> Eine Mischung von 5,9 g 5-Chlor-2,4-disulf- amylanilin in 40 cm3 Äthanol (90o/oig), enthaltend 1 Tablettchen festes. Natriumhydroxyd, wird mit 1,6 cm3 wässrigem Formaldehyd (37 %ig) behandelt und eine Stunde auf dem Dampfbad erhitzt.
Nach dem Kühlen und Neutralisieren mit 2-n. wässriger Salzsäure kristallisiert das 6-Chlor-7-sulfamyl-3,4- dihydro-1,2,4-benzothiadiazin-1,1-dioxyd aus. Es wird aus Wasser umkristallisiert und schmilzt bei 272-275 .
Man engt die Mutterlaugen ein und fügt Wasser zu, worauf ein zweiter Anteil 6-Chlor-7-sulfamyl- 3,4-dihydro-1,2,4-benzothiadiazin-1,1-dioxyd erhal ten wird. Nach dem Umkristallisieren aus Wasser schmilzt es bei 272-275 . Eine Mischschmelz punktprobe mit dem nach Beispiel 1 erhaltenen Pro dukt zeigt keine Depression. An Stelle von Äthanol können auch andere nie dere Alkanole, wie Methanol, Propanol, Isopropanol oder n-Butanol, als Lösungsmittel verwendet werden.
Auch andere Alkalimetallhydroxyde, wie Lithium oder Kaliumhydroxyd, oder andere basische Kon densationsmittel lassen sich verwenden.
Das Natriumsalz dieser Verbindung erhält man durch Eindampfen einer 6-Chlor-7-sulfamyl-3,4-di- hydro-1,2,4-benzothiadiazin-1,1-dioxyd und eine äquimolare Menge Natriumhydroxyd enthaltenden wässrigen Lösung.

This table shows that the diuretic effect of the new compound on dogs is significantly better than that of chlorothiazide. In a further comparison of the diuretic effect of the new compound with chlorothiazide, rats that had been deprived of food for 18 hours were given 0.2% sodium chloride solution in an amount corresponding to 5019 of their body weight. Immediately afterwards, each of these compounds was given orally, dissolved in 0.1 ems dilute sodium hydroxide solution.
The spontaneously emptied urine was collected in graduated cylinders over the next three hours, its volume measured and converted as a percentage of the fluid intake. The concentrations of sodium and potassium were determined by flame photometry and the total excretion of this electrolysis calculated during the three hour test period. The values summarized in the following table are average values from 8-12 rats per group.

The new compound has a strong diuretic and natriuretic effect even when used on humans, while potassium excretion is not promoted to the same extent.
In patients with fluid and salt retention as a result of diseases of the heart, liver (cirrhosis) or kidneys (nephrosis), the compound promotes the excretion of fluids and salt, which is therapeutically desirable, without the risk of potassium depletion. The doses required for such an effect for oral administration are on average between 25 and 100 mg per day.
The new compound is also capable of lowering high arterial pressure, especially when given at the same time as another antihypertensive agent.
The new compound is intended to be used as a remedy in the form of pharmaceutical preparations. The process according to the invention for preparing the new compound is characterized in that 5-chloro-2,4-disulfamyl-aniline or a salt thereof is reacted with formaldehyde or a functional derivative thereof. Preferably, the reaction is carried out in the presence of an acid such as a mineral acid, for example a hydrohalic acid, e.g. B. hydrochloric acid or hydrobromic acid, or sulfuric acid, optionally in anhydrous form, before. The formaldehyde can also be in the form of one of its polymers, e.g.
B. can be used as paraformaldehyde, trioxane or hexamethylene triamine. As functional derivatives Ace can tal, z. B. dimethoxymethane or diethoxymethane can be used. The reaction is carried out primarily with approximately equivalent amounts of the reaction components. Higher amounts of aldehyde reduce the yields as a result of a reaction with the sulfamyl group in the 4-position. The reaction can be carried out in the absence or, preferably, in the presence of solvents such as an ether, e.g.
B. p-dioxane or diethylene glycol dimethyl ether, or a formamide, z. B. dimethyl formamide, at room or elevated temperature and at normal or elevated pressure or in the presence of an inert gas such as nitrogen, through.
Depending on the reaction conditions, the new compound is obtained in free form or in the form of its salts. Metal salts obtained can, for example, by reaction with aqueous acidic agents, such as mineral acid, e.g. B. hydrohalic acid, for example hydrochloric acid, or sulfuric acid, can be converted into the free compound.
This in turn can be converted into metal salts, such as alkali metal salts, by treatment, for example, with a metal hydroxide, such as sodium or potassium hydroxide, in a solvent such as an alkanol, e.g. B.
Methanol or ethanol, or water and subsequent evaporation of the solvent, or by allowing the free compound to react in an ether such as p-dioxane or diethylene glycol dimethyl ether with an alkali metal hydride or amide, e.g. B. sodium or potassium hydride or ainide.
In the following examples, the temperatures are given in degrees Celsius. <I> Example 1 </I> A mixture of 2.9 g of 5-chloro-2,4-dis, tilfamyl-aniline in 15 cm3 of anhydrous diethylene glycol dimethyl ether containing 0.5 cm3 of ethyl acetate is heated 109.5 g of hydrochloric acid per 1000 cm3, and 0.33 g of paraformaldehyde to 80 to 90 and held for one hour at this temperature.
The mixture is cooled to room temperature, the reaction mixture is concentrated under reduced pressure to one third of its volume and diluted with water. The product which has crystallized out is filtered off and recrystallized from water. This gives 6 - chloro - 7 - sulfamyl - 3,4 - dihydro-1,2,4-benzothiadiazine-1,1-dioxide, which melts at 226-268.
If the paraformaldehyde in the above example is replaced by 0.84 g of dimethoxymethane and the rest of the procedure is as described above, the same compound is obtained.
<I> Example 2 </I> A mixture of 2.9 g of 5-chloro-2,4-disulfamylaniline in 15 cm3 of diethylene glycol dimethyl ether, 0.8 cm3 of a 37% aqueous formaldehyde solution (0.3 g of formaldehyde ) and 0.5 cm3 of a saturated hydrochloric acid-ethyl acetate solution is heated to 80-90 for one hour.
After evaporation under reduced pressure and addition of water, the 6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiazine-1,1-dioxide crystallizes. It is filtered off and recrystallized from water. A mixed sample with the product obtained according to Example 1 begins to melt at 263.
<I> Example 4 </I> A mixture of 5.9 g of 5-chloro-2,4-disulphamylaniline in 40 cm3 of ethanol (90%), containing 1 solid tablet. Sodium hydroxide is treated with 1.6 cm3 of aqueous formaldehyde (37%) and heated on the steam bath for one hour.
After cooling and neutralizing with 2-n. 6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine-1,1-dioxide crystallizes out from aqueous hydrochloric acid. It is recrystallized from water and melts at 272-275.
The mother liquors are concentrated and water is added, whereupon a second portion of 6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine-1,1-dioxide is obtained. After recrystallizing from water, it melts at 272-275. A mixed melt point sample with the product obtained according to Example 1 shows no depression. Instead of ethanol, other lower alkanols, such as methanol, propanol, isopropanol or n-butanol, can also be used as solvents.
Other alkali metal hydroxides, such as lithium or potassium hydroxide, or other basic condensation agents can also be used.
The sodium salt of this compound is obtained by evaporating an aqueous solution containing 6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine-1,1-dioxide and an equimolar amount of sodium hydroxide.
