in welcher R, Wasserstoff, eine Alkylgruppe oder eine allenfalls monosubstituierte Phenylgruppe,
R Wasserstoff oder eine allenfalls monosubstituierte Aralkylgruppe und X eine hydrogenolytisch leicht abspaltbare Gruppe darstellen.
Die neuen Derivate sind kristallisierte Substan zen, die sich in Wasser nicht, in organischen Lö sungsmitteln mehr oder weniger leicht lösen. Sie stel len wertvolle Zwischenprodukte für die Synthese von antibiotisch wirksamen Stoffen dar: So kann man sie z. B. durch Veresterung mit einer Säure, reduktive Aufspaltung der erhaltenen Ester und Cyclisierung der gebildeten Hydroxamsäuren mittels basischer Mittel in antibiotisch wirksame Aminoisoxazolidon- verbindungen überführen.
Besonders geeignet sind dazu Verbindungen der Formel I, in welcher R1 Wasserstoff, R eine Benzylgruppe und X eine Carbobenzyloxy- oder Carballyloxygruppe darstellt, denn sie lassen sich auf dem angegebenen Synthese weg in das sogenannte Cycloserin , ein vor kurzem bekanntgewordenes Antibioticum, überführen.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Derivate von a-Amino-ss-hydroxy-carbon- säuren der Formel 1 ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

mit Hydroxylamin bzw. einem allenfalls monosubsti tuierten Aralkyläther von Hydroxylamin der Formel H2N-OR in Gegenwart eines wasserabspaltenden Mittels umsetzt.
Besonders geeignete Ausgangsverbindungen sind solche der Formel 1I, in welcher Ri Wasserstoff und X das Carbobenzyloxyradikal darstellt. Wertvoll sind auch Ausgangssäuren der Formel II, worin R, Me- thyl oder Phenyl und X das Carbobenzyloxyradikal bedeuten. Als hydrogenolytisch leicht abspaltbare Gruppe kann X neben der Carbobenzyloxygruppe mit Vorteil auch eine Carballyloxygruppe darstellen.
Die Umsetzung der Säure<B>11</B> mit dem Hydroxyl- amin bzw. den O-Äthern des letzteren ist leicht exo- therm. Es ist deshalb zweckmässig, das Reaktionsge misch zu kühlen. Als wasserabspaltende Mittel ver wendet man mit Vorteil N,N'-disubstituierte Carbo- diimide in ungefähr molaren Mengen. Besonders ge eignet ist das N,N'-Dicyclohexyl-carbodiimid. Die Reaktion wird am besten in einem indifferenten Lö sungsmittel, wie z.
B. Tetrahydrofuran, Dimethyl- formamid oder Dioxan, ausgeführt.
Eine besonders zweckmässige Ausführungsform des erfindungsgemässen Verfahrens besteht darin, dass man die Säure der Formel 1I mit dem Hydroxyl- amin bzw. dessen O-Äther in Tetrahydrofuran löst und dieses Gemisch unter mässiger Kühlung mit N,N'-Dicyclohexyl-carbodiimid versetzt. Nach Abfil- trieren des gebildeten N,N'-Dicyclohexyl-harnstoffes kann aus dem Filtrat das Reaktionsprodukt nach Abdampfen des Lösungsmittels sofort rein erhalten werden.
<I>Beispiel 1</I> 41 Gewichtsteile DL-1-Carbobenzyloxyamino-2- hydroxypropionsäure werden mit 21 Gewichtsteilen O-Benzyl-hydroxylamin in 300 Raumteilen Tetra- hydrofuran gelöst. Unter Eiskühlung und Rühren lässt man eine Lösung von 36 Gewichtsteilen N,N'- Dicyclohexyl-carbodiimid in 50 Raumteilen Tetra- hydrofuran innert 10-15 Minuten zutropfen. Man rührt 3 Stunden ohne Kühlung weiter, saugt vom gebildeten N,N'-Dicyclohexyl-harnstoff ab und dampft das Lösungsmittel im Vakuum ab.
Das Reak tionsprodukt bleibt als kristallisierter Rückstand zu rück. Nach Umlösen aus Methanol schmilzt der er haltene 1-Carbobenzyloxyamino-2-hydroxy-propion- hydroxamsäure-O-benzyläther bei 107-109 .
<I>Beispiel 2</I> 46,1 Gewichtsteile ölige DL-1-Carbobenzoxy- amino-2-hydroxy-buttersäure (hergestellt durch Car bobenzoxylierung der 1-Amino-2-hydroxy-butter- säure [Threonin] in sodaalkalischer Lösung;
zähes, hellgelbes Öl, das nicht kristallisiert) und 22,4 Ge wichtsteile O-Benzyl-hydroxylamin werden in 150 Raumteilen Tetrahydrofuran gelöst und unter Eis kühlung und Rühren 37,7 Gewichtsteile N,N'-Di- cyclohexyl-carbodiimid, in wenig Tetrahydrofuran gelöst, zugetropft. Nach beendetem Zutropfen rührt man noch 2-3 Stunden bei Zimmertemperatur wei ter, nutscht vom gebildeten N,
N'-Dicyclohexyl-harn- stoff ab und dampft das Filtrat im Vakuum zur Trockne ein. Das zurückbleibende Öl wird in 200 bis 300 Raumteilen Äther gelöst und in Eiswasser gekühlt. Der gebildete DL-1-Carbobenzoxyamino-2- hydroxy - buttersäure - hydroxamsäure- O -benzyläther beginnt sich bald als gallertige Masse abzuscheiden.
Man lässt über Nacht im Eisschrank stehen, nutscht ab und reinigt durch Umkristallisieren aus 250 Raum teilen Toluol. Schmelzpunkt 92-95 . <I>Beispiel 3</I> 50 Gewichtsteile 1-Carbobenzyloxyamino-2-hy- droxy-2-phenyl-propionsäure (hergestellt durch Car- bobenzoxylierung der 1-Amino-2-hydroxy-2-phenyl- propionsäure [ss-Phenyl-serin] in sodaalkalischer Lö sung;
Schmelzpunkt nach Umkristallisieren aus Te- trachlorkohlenstoff 94-96 ) und 19,5 Gewichtsteile O-Benzyl-hydroxylamin werden in 300 Raumteilen Tetrahydrofuran gelöst und unter Eiskühlung und Rühren 32,7 Gewichtsteile N,N'-Cyclohexyl-carbo- diimid, mit wenig Tetrahydrofuran vermischt, zuge- tropft. Nach beendetem Zutropfen rührt man noch 2-3 Stunden bei Zimmertemperatur weiter,
nutscht vom gebildeten N,N'-Dicyclohexyl-harnstoff ab und dampft das Filtrat im Vakuum zur Trockne ein. Der zurückbleibende kristallisierte Rückstand wird mit Äther angeteigt, genutscht und reichlich mit Äther und Petroläther gewaschen. Nach Umkristallisieren aus Toluol schmilzt der 1-Carbobenzyloxyamino-2- hydroxy-2-phenyl-propion-hydroxamsäure -O -benzyl- äther bei 160-163 .

R is hydrogen or a monosubstituted aralkyl group, if any, and X is a group which can easily be split off by hydrogenolysis.
The new derivatives are crystallized substances that do not dissolve in water, and more or less easily in organic solvents. You stel len valuable intermediates for the synthesis of antibiotic substances: So you can z. B. by esterification with an acid, reductive cleavage of the esters obtained and cyclization of the hydroxamic acids formed by means of basic agents in antibiotic Aminoisoxazolidon- compounds.
Particularly suitable for this purpose are compounds of the formula I in which R1 is hydrogen, R is a benzyl group and X is a carbobenzyloxy or carballyloxy group, because they can be converted into the so-called cycloserine, a recently known antibiotic, by the synthesis given.
The process according to the invention for the preparation of the new derivatives of α-amino-β-hydroxy-carboxylic acids of the formula 1 is characterized in that a compound of the formula

with hydroxylamine or a possibly monosubstituted aralkyl ether of hydroxylamine of the formula H2N-OR in the presence of a water-releasing agent.
Particularly suitable starting compounds are those of the formula 1I in which Ri is hydrogen and X is the carbobenzyloxy radical. Starting acids of the formula II in which R is methyl or phenyl and X is the carbobenzyloxy radical are also valuable. As a group which can easily be split off by hydrogenolysis, X can advantageously also represent a carballyloxy group in addition to the carbobenzyloxy group.
The reaction of the acid 11 with the hydroxylamine or the O-ethers of the latter is slightly exothermic. It is therefore advisable to cool the reaction mixture. N, N'-disubstituted carbodiimides in approximately molar quantities are advantageously used as dehydrating agents. N, N'-dicyclohexylcarbodiimide is particularly suitable. The reaction is best in an indifferent Lö solvent such.

While cooling with ice and stirring, a solution of 36 parts by weight of N, N'-dicyclohexylcarbodiimide in 50 parts by volume of tetrahydrofuran is added dropwise over a period of 10-15 minutes. The mixture is stirred for a further 3 hours without cooling, the N, N'-dicyclohexylurea formed is filtered off with suction and the solvent is evaporated off in vacuo.
The reaction product remains as a crystallized residue. After dissolving from methanol, the 1-carbobenzyloxyamino-2-hydroxy-propionic hydroxamic acid-O-benzyl ether obtained melts at 107-109.
<I> Example 2 </I> 46.1 parts by weight of oily DL-1-carbobenzoxy-amino-2-hydroxy-butyric acid (prepared by carbobenzoxylation of 1-amino-2-hydroxy-butyric acid [threonine] in a soda-alkaline solution ;
viscous, light yellow oil that does not crystallize) and 22.4 parts by weight of O-benzyl-hydroxylamine are dissolved in 150 parts by volume of tetrahydrofuran and, while cooling with ice and stirring, 37.7 parts by weight of N, N'-dicyclohexyl-carbodiimide in a little tetrahydrofuran dissolved, added dropwise. After the end of the dropwise addition, the mixture is stirred for another 2-3 hours at room temperature, the N formed is filtered off with suction.
N'-dicyclohexyl urea and the filtrate evaporated to dryness in vacuo. The remaining oil is dissolved in 200 to 300 parts by volume of ether and cooled in ice water. The DL-1-carbobenzoxyamino-2-hydroxy-butyric acid-hydroxamic acid-O -benzyl ether formed soon begins to separate out as a gelatinous mass.
It is left to stand in the refrigerator overnight, suction filtered and purified by recrystallization from 250 parts of toluene. Melting point 92-95. <I> Example 3 </I> 50 parts by weight of 1-carbobenzyloxyamino-2-hydroxy-2-phenylpropionic acid (prepared by carbobenzoxylation of 1-amino-2-hydroxy-2-phenylpropionic acid [ss-phenyl -serine] in a soda-alkaline solution;
Melting point after recrystallization from carbon tetrachloride 94-96) and 19.5 parts by weight of O-benzyl-hydroxylamine are dissolved in 300 parts by volume of tetrahydrofuran and, with ice-cooling and stirring, 32.7 parts by weight of N, N'-cyclohexyl-carbodiimide with a little tetrahydrofuran mixed, added dropwise. When the dropping is complete, the mixture is stirred for another 2-3 hours at room temperature,
The N, N'-dicyclohexylurea formed is filtered off with suction and the filtrate is evaporated to dryness in vacuo. The remaining crystallized residue is made into a paste with ether, suction filtered and washed copiously with ether and petroleum ether. After recrystallization from toluene, the 1-carbobenzyloxyamino-2-hydroxy-2-phenyl-propion-hydroxamic acid -O -benzyl- ether melts at 160-163.
