Verfahren zur Herstellung von substituierten 2-Oxo-benzo-chinolizinen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von substituierten 2-Oxo-berrzo-chinolizinen der Formel

in welcher R3 einen Alkyl-, Alkenyl- oder Aralky liest, der Äther-Sauerstoff bzw.
ey clisch gebundenen Sauerstoff enthalten kann, und R1 und R., Wasserstoff oder Alkoxygruppen oder zusammen einen Alkvlendioxvrest bedeuten, und von im isocyclischen Ring partiell lry drier- ten Derivaten davon.
Das erfindungsgemässe Verfahren ist da- durch ;gekennzeichnet, dass man ein 1-Ca.rb- all;oxymethyl-1,2,3,4-tetrahy droisochinolin, des sen isoeyclischer Ring partiell hydriert und in der --enannten Weise substituiert sein kann, mit Formaldelry d oder einer formaldehyd- abgebenden Verbindung und einer 1Ialonsäure der Formel R3 CH (COOH) 1,
oder einem Di- ester davon umsetzt, wobei die entsprechende 11j-droisochinolyl-(N)-methylmalonsäure oder deren Diester entsteht, im ersteren Fall die freien Carboxylgruppen verestert,
den erhal tenen Tricarbonsäureester mittels eines alkali- sehen Kondensationsmittels durch intramole- kulare Kondensation cyclisiert und den gebil deten tricyclischen Ketodicarbonsäureester verseift und decarboxy liert.
Die als Ausgangsmaterial benötigten 1- Carbalkoxymethy 1-1,2,3,4-tetrahydroisochino- line, deren isocyclischer Ring partiell hydriert und in der genannten Weise substituiert sein kann, lassen sich nach bekannten Verfahren (z.
B. gemäss J. Chem. Soc. London 1951, Seite 3472, und 1953, Seite 2465) durch Um setzung der in (-Stellung entsprechend sub stituierten Äthylamine mit Malonsäurehalb- esterchlorid, Cyclisierung der gebildeten Halb- esteramide und nachfolgende katalytische Hy drierung gewinnen.
In der ersten Reaktionsstufe (14Zannich- Reaktion) kann man das substituierte Ilydro- isochinolin und den Formaldehyd mit der freien monosubstituierten Malonsäure zur Re aktion bringen.
Dabei lässt man die Reak tionspartner mit Vorteil in wässriger Lösung bei Raumtemperatur während längerer Zeit aufeinander einwirken; das auskristallisierende Kondensationsprodukt, das eine freie Iso- chinolinyl-methylmalonsäure darstellt und zer- setzlich ist, wird anschliessend verestert, wo durch es in einen Tricarbonsäureester über geht. Diese Veresterung geschieht zweckmässig mit Diazomethan in ätherischer Lösung oder mit einem Dialkylsulfat in Gegenwart von Alkalicarbonat, z.
B. mit Dimethylsulfat in wässriger Sodalösung.
Der gleiche Tricarbonsäureester kann auch direkt so gewonnen werden, dass man für die Mannich-Kondensation als Reaktionspartner anstelle der substituierten freien Malonsäure einen entsprechenden Malonsäurediester, ins besondere den Dimethylester, verwendet. Im letzteren Falle empfiehlt es sich, die Reaktions partner in einem inerten Lösungsmittel, z. B. in einem niederen Alkohol, miteinander zu er wärmen.
Die gewonnenen Tricarbonsäureester wer den nun nach Dieckmann mittels eines alkali schen Kondensationsmittels cyclisiert. Als alkalische Kondensationsmittel kann man z. B. Alkalialkoholate verwenden und die Cyclisie- rung v orteilhafterweise durch Erwärmen in einem inerten Lösungsmittel, wie z. B. Benzol oder Toluol, zustandebringen.
Die gebildeten Chinolizine, die in 1- und '-Stellung veresterte Carboxy lgruppen tragen, werden sodann zwecks Abspaltung dieser Gruppen verseift und decarboxyliert. Dies ge schieht mit Vorteil durch Erwärmen mit ver dünnten Mineralsäuren, z. B. mittels Salz säure. Aus den erhaltenen mineralsauren Lö sungen kann man die freien Basen durch Ver setzen mit Alkali ausscheiden.
Die gebildeten Verbindungen der ange gebenen Formel, welche, wie gesagt, im iso- cyclischen Ring partiell hydriert sein können, sind kristallin und in Wasser löslich. Mit den gebräuchlichen anorganischen und organischen Säuren, z. B. mit Salzsäure, Schwefelsäure, Phosphorsäure, Weinsäure, Essigsäure, Citro- nensäure usw., bilden sie in Wasser lösliche Salze.
Sowohl die Basen als auch die Salze zeichnen sich durch blutdrucksenkende, seda tive , analgetische und antipyretische Wirkun gen aus und können deshalb als Heilmittel oder als Zwischenprodukte zur Synthese von Heilmitteln verwendet werden. Beispiel <I>1</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 35 Ge wichtsteile Paraformaldehyd und 150 Ge- wichtsteile Mono-methyl-malonsäuredimethyl- ester werden in 1.000 Raumteilen Methanol 24 Stunden unter Rückfloss gekocht.
Das 1 Carbäthoxy methy 1- 2 - (2',2- dicarbomethoxy-n- propy l) - 6,7 - dimethoxy -1,2, 3,4 - tet.rahydroiso- chinolin kristallisiert beim Abkühlen und schmilzt nach dem Umlösen aus Methanoi- Wasser bei 89-91 .
28 Gewiehtsteile -Natrium werden in 500 Raumteilen Methanol gelöst und die Lösung zur Trockne eingeengt. -Nun werden 3500 Raumteile Toluol und 440 Gewichtsteile 1- Carbä.tlioxy methy 1-2- (2',2' - dicarbomethoxv-n- propy 1) -6,7-dimethoxy -1,2,3,
4 - tetrahy droiso- ehinolin vom Schmelzpunkt 89-91 zugegeben und die Mischung unter Rühren erwärmt. Der bei der Kondensation gebildete Alkohol wird kontinuierlich abdestilliert und die Destilla tion unterbrochen, sobald der Siedepunkt des Toluols erreicht ist. Die -Mischung wird an schliessend 2 Stunden unter Rückfloss erhitzt, dann zur Trockne eingeengt.
Der Rückstand wird in 6000 Raumteilen 3n-Salzsäure gelöst und während 16 Stunden auf 120 Badtem- peratur erhitzt, wobei Verseifung, und De- carboxylierung stattfindet.
Nach dem Ab kühlen wird mit Ammoniak alkalisch ge stellt und das ausgefallene 2-Oxo-3-methy 1-9,10- dimetlioxy-1,2,3,4,6, 7 -hexahvdro-benzo [al chi- nolizin ausMethanol-Wasserumgelöst. Schmelz punkt 138-140 . Hydrochlorid: Schmelzpunkt 204-205 .
<I>Beispiel 2</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-t.etrahy droisochinolin, 35 Ge- wiehtsteile Paraformaldehvd und 160 Ge- wiehtsteile Mono-äthvl-nralonsäuredimeth5-1- ester werden in 1000 Raumteilen 'Methanol suspendiert und 24 Stunden unter Rückfloss gekocht.
Das beim Abkühlen kristallisierende 1-Carbätlioxy methyl - 2 - (2',2'- dica.rbomethoxy- n-buty 1)- 6,7 - dimethoxy-1,2,3,4-tetrahy droiso- chinolin schmilzt nach dem Umlösen aus Me thanol bei 118-120 .
28 Gewichtsteile -Natrium werden in 650 Raumteilen absolutem Äthanol gelöst, zur Trockne eingeengt und mit 3600 Raumteilen Toluol und 451 Gewiehtsteilen 1-Carbäthoxy- methyl- 2 - (2',2'- dicarbomethoxy-n-butyl)- 6,7- dimethoxy-1,2,3,4-tetrahydroisoehinolin ver setzt. Unter Erwärmen wird der gebildete Alkohol entfernt, bis der Siedepunkt des Toluols erreicht ist.
Dann wird noch 2 Stun den unter Rückfluss gekocht und hierauf zur Trockne eingeengt. Der Rückstand wird in 5200 Raumteilen 3n-Salzsäure gelöst und zur Verseifung und Decarboxylierung während 14 Stunden auf 120 Badtemperatur erhitzt. Nach dem Erkalten wird mit Äther gewaschen, mit. Kohle entfärbt und nach dem Alkalisch stellen in Äther aufgenommen.
Das 2-Oxo-3- Rliyl - 9,10-dimethoxy-1,2,3,4,6,7-liexahydro- benzo[a]ehinolizin schmilzt nach dem Uinlösen aus Di-n-butyläther bei 110-112 .
Hydroelilorid: Schmelzpunkt 198-200 ; Hydrobromid: Schmelzpunkt 218 .
<I>Beispiel 3</I> 279 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin und 145 Gewiehtsteile Mono-äthylmalonsäure wer den in 600 Raumteilen Wasser gelöst, mit 110 Gewiehtsteilen einer 30 loigen wässrigen Form aldehydlösung versetzt und 2 Stunden bei Zimmertemperatur stehengelassen.
Das aus kristallisierte 1-Carbäthoxymetb.yl-2-(2',2'-di- earboxy - n - butyl) -6,7-dimethoxy-1,2,3,4-tetra- liydi-oisoehinolin wird filtriert, mit wenig Eis wasser gewaschen und im Vakuumexsiccator über Pbosphorpentoxyd getrocknet. Schmelz punkt 66-68 unter Zersetzung.
425 (Tewichtsteile 1-CarbäthoxymethSrl-2- (2',2'-diearboxy-n-blityl) -6,7-dimethoxy-1,2,3,4- teti@ahvdroisochinolin werden in 5000 R.aum- teilen.Äther suspendiert und mit 8000 Raum teilen einer aus 350 Gewichtsteilen Nitroso- niethvlliarnstoff bereiteten Diazomethanlösung in Äther versetzt. Nach der Zugabe wird wäh rend 15 Minuten gerührt, filtriert und zur Trockne eingeengt.
Der Rückstand liefert nach dem Aufnehmen in Methanol das kri stallisierte 1-Carbäthaxymethyl-2-(2',2'-dicarbo- in ethoxy-n - butyl) -6,7-dimethoxy-1,2,3,4-tetra- hydroisochinolin vom Schmelzpunkt 118-120 , welches gemäss Beispiel 2 weiterverarbeitet werden kann.
<I>Beispiel 4</I> 425 Gewichtsteile 1-Carbäthoxymethyl-2 (2',2'-dicarboxy-n-butyl)-6,7-dimethoxy-1,2,3,4- tetrahydroisochinolin (gemäss Beispiel 3 ge wonnen) werden in 2000 Raumteilen Aceton gelöst und mit 420 Gewichtsteilen trockenem Kaliumcarbonat versetzt; man lässt sodann 260 Gewichtsteile Dimethylsulfat zutropfen, wobei die Temperatur um etwa 10 steigt. Nach dem Filtrieren wird mit Aceton ausgewaschen.
die vereinigten Acetonlösungen eingeengt und der Rückstand aus Methanol ki--istallisiert. Das dabei gewonnene 1-Carbäthoxymethyl-2-(2',2' dicarbomethoxy-n-butyl)-6,7-dimethoxy-1,2,3,4- tetrahydroisochinolin schmilzt bei 118-120 ; es kann gemäss Beispiel 2 weiterverarbeitet werden.
Beispiel <I>5</I> 270 Gewichtsteile 1-Carbomethoxymethyl- 6,7 - dimethoxy -1,2,3,4 - tetrahydroisochinolin (Schmelzpunkt 84-86 ), 35 Gewichtsteile Paraformaldehyd und 160 Gewichtsteile Mono äthyl-malonsäuredimethylester werden in 1000 Raumteilen Methanol suspendiert und 24 Stunden unter Rückfluss gekocht.
Das beim Abkühlen kristallisierende 1-Carbomethoxy methyl=2-(2',2'-dicarbomethoxy-n-biityl)=6,7-di- methoxy-1,2,3,4-tetrahydroisochinolin schmilzt nach dem Umlösen aus Methanol-Wasser bei 125-126 . Dieser Triester kann gemäss Bei spiel 2 in 2-Oxo-3-äthyl-9,10-dimethoxy-1,2,3,4, 6,7-hexahydro-benzo [a] chinolizin übergeführt werden.
Beispiel <I>6</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6, 7- dimethoxy-1,2,3,4 - tetrahydroisochinolin und 160 Gewichtsteile Mono-isopropy lmalonsäure werden in 600 Raumteilen Wasser gelöst, mit. 110 Raumteilen einer 30%igen wässrigen Form aldehydlösung versetzt und 48 Stunden bei Raumtemperatur stehengelassen.
Das auskri stallisierte 1-Carbäthoxymethyl-2-(2',2'-dicarb- oxy- 3'-methyl-n-biit3#l)-6,7-dimethoxy-1,2,3,4- tetrahydroisochinolin schmilzt nach dem Um lösen aus Dioxan-Äther bei 89 unter Zerset zung.
Der entsprechende, analog Beispiel 3 daraus hergestellte Triester schmilzt nach dem Umlösen aus Essigester-Petroläther bei 64 bis 65 und liefert nach Dieckmann-Kondensation, Verseifung und Decarboxylierung entspre chend den Angaben in Beispiel 2 das 2-Oxo-3- isopropyl - 9,10 - dimethoxy -1,2,3,4,6,7 - hexa- hydro-benzo [a] chinolizin vom Schmelzpunkt 123-124 . Hydrobromid: Schmelzpunkt 222 bis 223 .
<I>Beispiel 7</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 175 Gewichtsteile Mono-allylmalonsäure-dimethyl- ester und 33 Gewichtsteile Pa.raformaldehyd werden in<B>1,000</B> Raumteilen Methanol während 24 Stunden unter R.ückfluss erwärmt.
Das beim Erkalten auskristallisierende 1-Carbo- methoxvuuethyl-2- (2',2'-dicarbomethoxy-n-pen- ten- [4' ] -y1) - 6,7 - dimethoxy-1,2,3,4-tetrahydro- isochinolin (beim Kochen in Methanol ist eine Umesterung erfolgt) schmilzt bei 93-95 und liefert nach Dieckmann-Kondensation, Versei fung und Decarboxylierung entsprechend den Angaben in Beispiel 1 das 2-Oxo-3-allyl-9,10- dimethoxy-1,2,3,4,6,
7 - hexahydro-benzo [a] chi- nolizin vom Schmelzpunkt 116-117 . Hydro- bromid: Schmelzpunkt 204-205 .
<I>Beispiel 8</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 35 Ge wichtsteile Paraformaldehyd und 180 Ge wichtsteile Mono-n-butyl-malonsäuredimethyl- ester werden in 1000 Raumteilen Methanol 24 Stunden unter Rückfloss erwärmt.
Die basischen Anteile werden mit 3n-Salzsäure aus der Reaktionsmischung extrahiert; nach Ver setzen des sauren Auszuges mit Ammoniak lösung und Aufnehmen in Chloroform erhält man das kristallisierte 1-Carbäthoxymethyl=2 (2',2'-dicarbomethoxy-n-hexyl)-6,7-dimethoxy- 1,2,3,4 - tetrahydroisochinolin. Schmelzpunkt nach dem Umlösen aus Methanol-Wasser 79 bis 81 . Es kann nach Beispiel 9 weiterverarbeitet werden.
Beispiel <I>9</I> Bei der Umsetzung von 280 Gewichtsteilen 1-Carbäthoxymethyl - 6,7-(dimethoxy-1,2,3,4- tetrahydroisochinolin,175 Gewichtsteilen Mono- n-butyl-malonsäure und 220 Raumteilen einer 30%igen wässrigen Formaldehydlösung nach den Angaben von Beispiel 3 und Ausscheiden des Kondensationsproduktes mittels Methylen- ehlorid erhält man 1-Carbäthoxymeth-,#-1-2 (2',2'-dicarboxy-n-hexyl)-6,7-dimethoxy-1,2,3,
4- tetrahydroisochinolin, welches durch Vereste- rung mit einer ätherischen Diazomethanlösung entsprechend den Angaben von Beispiel 3 in das schon in Beispiel 8 beschriebene 1-Carb äthoxymethyl-2-(2',2'-dicarbomethoxy-n-hexyl)- 6,7 - dimethoxy - 1,2,3,4 - tetrahy droisochinolin vom Schmelzpunkt 79-81 übergeführt wer den kann.
Aus letzterem entsteht. nach Deck- inann-Kondensation, Verseifung und De- carboxylierung entsprechend den Angaben in Beispiel 1 das 2-Oxo-3-n-butyl-9,10-dimetlr- oxy-1,2,3,4,6,7-hexahydro-benzo [a] chinolizin. Schmelzpunkt nach Umlösen aus n-Butanol 112 bis 114 . Hydrochlorid: Schmelzpunkt<B>188</B> bis 189 .
Beispiel. <I>10</I> 280 Gewichtsteile 1-Carbä.thoxymethyl-6, 7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 35 Ge wiehtsteile Paraformaldehyd und 220 Ge wichtsteile Mono-benzyl-malonsäuredimethyl- ester werden in 1000 Raumteilen lIethanol 24 Stunden unter Rückfloss gekocht. Der nach dem Einengen zur Trockne erhaltene Rück stand wird in 3n-Salzsäure gelöst und die wä.ssrige Lösung mit Äther gewaschen.
Nach Versetzen der wässrigen Lösung mit Ammo- niaklösung werden die ausgeschiedenen Basen in Äther aufgenommen und nach dem Ein engen aus Äther-Petroläther kristallisiert. Das gewonnene 1-Carbäthoxymethyl-2- (2',2'-di- carbomethoxy -3'-phenyl-n-propyl)-6,7-dimeth- oxy-1,2,3,4-tetra.hy droisochinolin schmilzt bei 97-98 und liefert nach Dieckmann-Konden- sation,
Verseifung und Decarboxylierung ent sprechend den Angaben in Beispiel 1 das 2- Oxo - 3 -benzyl-9,10-dimethoxy-1,2,3,4,6,7-hexa- hydro-benzo [a] ehinolizin. Schmelzpunkt nach Umlösen aus Methanol-Wasser 139-141 ; Hydrochlorid: Schmelzpunkt 165-167 .
<I>Beispiel 11</I> 209 Gewichtsteile 1-Carbomethoxy-methyl- 1,2,3,4,5,6, 7,8-octahydroisochinolin (Siedepunkt 1.05-110 ;0,1 mm, nach bekanntem Verfahren aus dem Malonsäurehalbester-cyclohexen-(1)- yl-ätliylamid bereitet), 150 GewichtsteileMono- äthyl-malonsäure-dimethylester und 35 Ge wichtsteile Paraformaldehyd werden in 2000 Raumteilen Methanol 15 Stunden unter Rück fluss gekocht. Der nach dem Einengen er haltene Rückstand wird in 3n-Salzsäure ge löst und mit Äther gewaschen.
Die wässrige Lösung wird durch Zugabe von Sodalösung auf pH 5 gestellt und mit Chloroform mehr mals ausgezogen. Das durch Trocknen und Einengen der Chloroformlösung erhaltene ölige 1-Carbomethoxymethyl-2- (2',2'-dicarbo- methoxy- n - butyl) -1,2,3,4, 5, 6,7, 8-octahydroiso- chinolin liefert nach Dieckmann-Kondensation, Verseifung und Decarboxylierung,
entspre chend den Angaben in Beispiel 1, das 2-Oxo-3- ä.thvl -1,2,3,4,6,7,8,9,10,11-decahydrobenzo [a] - chinolizin. Das Keton kann nach Destillation im Hochvakuum (Siedepunkt 100-110 /0,05 Millimeter) aus Petroläther kristallisiert wer den. Schmelzpunkt 69 . Hydrochlorid: Schmelz punkt 122 .
<I>Beispiel</I> .22 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 150 Gewichtsteile Mono-isobutyl-malonsäure-dime- thyiester und 35 Gewichtsteile Paraformalde- hy d werden in 1000 Raumteilen Methanol während 24 Stunden unter Rückfluss gekocht.
Das beim Abkühlen auskristallisierende 1-Carb- ä,th oxy - methyl - 2 - (2',2'-diearbomet.hoxy-4'-me- thyl-n-penty 1)-6,7-dimethoxy-1,2',3,4-tetraliydro- isoehinolin schmilzt nach dem Umlösen aus Methanol bei 94-96 .
Aus letzterem entsteht nach Dieclzmann-Kondensation, Verseifiing und Decarboxylierung entsprechend den An gaben in Beispiel 1 das 2-Oxo-3-isobutyl-9,10- dimethoxy-1,2,3,4,6, 7 -hexahydro-benzo [a] chi- nolizin. Schmelzpunkt nach Umlösen aus Iso- propyläther 126-128 . Hydrochlorid: Schmelz punkt 196-197 .
Beispiel <I>23</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 35 Ge wichtsteile Paraformaldehyd und 180 Ge wichtsteile Mono-n-ainyl-malonsäure-dimethy 1- ester werden in 1000 Raumteilen Methanol 24 Stunden unter Rückfluss gekocht.
Der nach dem Einengen zur Trockne verbleibende, aus 1-Carbäthoxymethyl- 2-(2',2'-dicarbomethoxy- n-hept.yl)- 6,7-dimethoxy-1,2,3,4-tetrahydroiso- chinolin bestehende Rückstand wird direkt, entsprechend den Angaben in Beispiel 1, einer Dieckmann-Kondensation, Verseifung und De- carboxylierung unterworfen.
Das erhaltene 2- Oxo - 3-n-amyl-9;10-dimethoxy-1,2,3,4,6,.7-hexa- hydro-benzo [a.] chinolizin schmilzt nach dem Trocknen und Umlösen aus Essigester-Petrol- äther bei 121-122 . Hydrochlorid : Schmelz punkt 184--185 .
Beispiel <I>14</I> 280 Gewichtsteile 1-Carbäthoxymethyl-6,7- dimethoxy-1,2,3,4-tetrahydroisochinolin, 35 Ge wichtsteile Paraformaldehyd und 170 Ge wichtsteile Mono-n-hexyl-malonsäure-dimethyl- ester werden in 1000 Raumteilen Methanol 24 Stunden unter Rückfluss gekocht.
Das beim Abkühlen auskristallisierende 1-Carb- äthoxymethyl-2- (2', 2'-dicarbomethoxy-n-octyl)- 6, 7 - dimethoxy - 1,2,3,4 - tetrahydroisochinolin schmilzt nach dem Umlösen aus Methanol bei 91-93 ;
es liefert nach Dieckmann-Konden- sation, Verseifung und Decarboxylierung, ent sprechend den Angaben in Beispiel 1, das 2 Oxo-3-n-hexyl-9,10-dimethoxy-1,2,3,4,6,7-hexa- hy dro-benzo [a] chinolizin, das nach dem Um lösen aus Isopropyläther bei 92-94 schmilzt; Hydrochlorid: Schmelzpunkt 173-175 .
<I>Beispiel 15</I> 263 Gewichtsteile 1-Carbäthoxymethyl-6,7- methylendioxy - 1,2,3,4 - tetrahydroisochinolin (Smp. 55-57 ; nach bekannten Verfahren aus dem Malonsäure - monoäthylester - monohomo- piperonylamid vom Schmelzpunkt 96-98 über das 1- Carbäthoxymethyl-6,7-methylen- dioxy -3,4-dihydroisochinolin vom Schmelz punkt 144-146 bereitet),
35 Gewichtsteile Paraformaldehyd und 160 Gewichtsteile Mono äthyl-malonsäure-dimethylester werden in 1000 Raumteilen Methanol 24 Stunden unter Rück fluss gekocht.
