Verfahren zur Herstellung von /\ 5.16.pregnadienverbindungen Die Herstellung von wertvollen Steroiden durch Totalsynthese, inklusive Bildung des Vierringsy stems und Einführung der angu- lären Methylgruppen in den Stellungen 1.0 und 1 3 sowie das Einfügen gewünschter funk tioneller Substituenten in das Ringsystem bietet bedeutende Schwierigkeiten.
Zusätzlich zli diesen Schwierigkeiten wird das Problem noch durch die '#')tereochemieverhältnisse er- schwert. So können gesättigte Steroide mit einem Minimum von 6 as3 mmetrischen Zen tren in mindestens 64 stereochemischen Modi- Eikationen vorkommen. Trotz dieser Schwie rigkeiten wurden Versuche unternommen, Steroide durch Totalsynthese herzustellen. Anreiz dazu bot die Wichtigkeit und der Wert solcher Steroide, z. B. des Cortisons, sowie die Seltenheit der natürlichen Rohstoffe, die als Ausgangsmaterial für die Herstellung dieser wichtigen Produkte in Frage kommen.

Das 2,4b-Diinethyl-l-(f oxy-äthyl)-2-methallyl- 7-äthylendioxy-1,2,3,4,4a,4b,5,6, 7, 8,10,10a- dodekahydrophenanthren-4-on kann dadurch hergestellt werden, dass 2,4b-Dimetliyl-l- (2-tosyloxy-äthyl) -2- methallyl-7-äthylendioxy-1,2,3,4,4a,4b,5,6,7, 8,10,10a-dodekahydrophenanthr en-4-on der alkalischen Hydrolyse unterworfen wird.

Zur Herbeifiihrung des Ringschlusses kann man zum Beispiel eine Suspension von 2,4b-Dimetllyl-7-äthy lendioxy-2-(ss-keto- propyl) -1-acetaldehydo-1,2,3,4,4a,4b,5,6, 7,8,10,7 0a-dodekahydrophenanthren-4-on in einer wässrigen Lösung von Kalium hydroxyd während 19 Stunden auf ungefähr 1.00 C erhitzen, wobei man A5.16_3_Äthyleil- clioxy-pre,-nadien-11,20-dion erhält.
Dieses Produkt kann aus der alkalischen Lösung durch Extrahieren mit einem wasserLinlös- liehen Lösungsmittel, z. B. Chloroform, und Konzentrieren des Auszuges isoliert werden.
Das 45,16-pregnadien-3,1.1,20-trion mit ge schützter 3-Ketogruppe sowie das daraus Bure h Hydrolyse mit Säure erhältliche d 4,16- Preonadien-3,17,20-trion sind wertvolle Zwi schenprodukte für die Synthese von Corti- sonen. So kann z. B. dl-d 5,16-3_Äthylendiox@-- pregnadien-11,20-dion durch selektive Hydrie- rung; mit.
Wasserstoff in Gegenwart von Raney-Niekel-Katalysator in dl-,95-3-Äthylen- (lioxl--pi-eglien-11,20-dion umgewandelt wer- den. Es kann dabei die Methode verwendet werden, die in der Literatur für die Reduk tion des A5,16_3_Oxy-20-keto-pregnadiens zu -9 5-3-Oxy-20-ketopregnen angegeben ist. Das dl-d,5-3-Äthylendioxy-pregnen-11,20-dion kann nach den in der Literatur beschriebenen Me thoden in Cortison verwandelt werden.
<I>Beispiel, 2</I> a; 2,4b-Dimethj#1-7-äthylendioxy-2-methallyl 1-acetaldeliydo-1,2,3,4,4a,4b,5,6,7,8,10,10a- dodekahydrophenanthren-4-on. Zum Komplex, erhalten durch Lösen von 380 mg Chromtrioxyd in 3,5 em3 Pyridin, wurden 381. mg 2,4b-Dimethyl-7-ätliylendioxy -2-methallyl 1-(ss-oxy-ä.thyl)-1,2,3,4,4a.,4b,5,6,7,8,10,10a- dodekahy drophenanthren-4-ol in 3 cm3 Pyridin gegeben.
Nachdem die Reaktionsmischung über Nacht bei Raum temperatur stehengelassen worden war, wurde sie mit. Wasser verdünnt und mit Äther extrahiert. Die vereinigten Ätherextrakte wur den mit Wasser gewaschen, getrocknet und dann konzentriert. Das erhaltene kristalline Rohprodukt.
wurde dann in einer säure gewaschenen Aluminiumoxydsäule von 12 g ehromatographiert. Mit Petroleumäther-Äther wurde zuerst 2,4b-Dimethyl-7-ät.hylendioxy-2-methallyl l.-aeet.aldehydo-1,2,3,4,4a,4b,5,6,7,8,10,10a- dodekahydrophenanthren-4-on mit einem Schmelzpunkt von 148-150 C (nach Umkristallisieren aus Äther) eluiert. Es folgte dann 2,4b-Dimethyl-7-äthylendioxy-2-metIiall.yl 1-(ss-oxy-äthzTl)-1,2,3,4,4a,4b,5,6,7,8,10,10a- dodekahydrophenanthren-4-on mit einem Schmelzpunkt von 182-186 C (aus Äther).
Durch weiteres EI-Liieren mit Vietha- nol erhielt man 2,4b-Dimethyl-7-äthylendioxy-2-methally 1- 1-carboxy methyl-1,2,3,4,4a,4b,5,6,7,8,10,10a- dodekahydrophenanthren-4-oii.
mit einem Schmelzpunkt von 215-216 C (nach Reinigung durch Umkristallisieren aus Äthylacetat). Die oben genannten 1-(P-Oxy-ätht-1)- und 1-(Carboxy-methy l )-Verbinclungen können mit Lithium-aluminiumhydrid in Tetrahydrofuran reduziert werden, wobei das C-11-Epimere von 2,4b-Dimethyl- 7 -ätliy lenclioxy -2-metlia.ll.yl 1-(ss-oxy-äthyl)-1,2,3,4,4a,4b,5,6,7,8,10,10a- dodekahydrophenanthren-4-ol,
dem uraprüngliehen Ausgangsmaterial, ent steht.
b) 2,1b-Dimethyl-7-äthylendioxy-2-(ss,y-dioxy- isobutyl)-1-aceta.ldehydo-1,2,3,1,1a,4b,5,6,7. 8,10,10a-dodekahy drophenanthren-4-on.
Zu einer Lösung von 1.45 mg von 2,4b-Dimethyl-7-äthyl.endioxy-2-metliallyl 1-acetaldehydo-1,2,3,4,4a,4b,5,6,7,8,10,10a- dodekahy drophenanthren-4-on in 1,5 em3 Benzol und 0,5 em3 Tetrahydro- furan wurden 1.10 mg Osniiumtetroxyd gege ben.
Nach einer Stunde Stehenlassen bei Raumtemperaturwurde der ausgefällte Osmiat- ester in 6,5 em3 Äthanol aufgelöst und mit einer Lösung von 700 mg Natriumsulfit in 4 em3 Wasser behandelt. Die Mischung wurde dann während 20 Minuten tüchtig gerührt, um die Hv drolv se des Osiniatesters vorzuneh men. Nach kurzem Stehen dekantierte man die obere Schicht, wusch die untere Schicht mehr mals mit Äthanol und dekantierte wieder.
Die organischen Lösungen wurden zusammen geschüttet und auf ein kleines Volumen kon zentriert. Extraktion mit. Chloroform mit nachfolgendem Waschen mit Wasser, Trock nen und Konzentrieren ergab kristallines 2,1b-Dimethyl-7-äthylendioxy-2- (ss#,y-dioxv- i s obut- yl) -1-acetaldehydo-1,2,3,1,la,lb,5,6, 7,8,10,1.0a-dodekahy drophenanthren-4-on mit einem Schmelzpunkt von 174-177 (,
