Verfahren zur Herstellung von basischen Carbinolen Es wurde gefunden, dass man wertvolle basische Carbinole erhält, wenn man 2-Cyclo- hexyliden- oder 2-Cyclohexyl-cyclohexanon mit einem am Stickstoffatom substituierten Aminopropin umsetzt.
Im Falle der Verwendung von 2-Cyclo- hexyl-cyclöhexanon und 3-Piperidino-3-me- thyl-propin als Komponenten ergibt sich das Schema

Für die Umsetzungen eignen sich insbe sondere die nach dem Verfahren der deut schen Patentschriften Nrn. 724759 und 730850 erhältlichen Aminopropine.
Nach dem in der deutschen Patentschrift Nr. 724759 beschriebenen Verfahren erhält man Aminopropine, wenn man primäre oder sekundäre Amine mit Aldehyden umsetzt und die dadurch erhaltenen, an einem Kohlen stoffatom in a-Stellung zum Stickstoff hydro- xylierten Amine mit Acetylen umsetzt. Das in der deutschen Patentschrift Nr. 730850 beschriebene Verfahren ist dadurch gekenn- zeichnet, dass man in flüssiger Phase Acety len auf primäre oder sektuzdäre Amine in Ge genwart von Acetyleniden der Schwermetalle der ersten und zweiten Gruppe des periodi schen Systems bei Temperaturen bis etwa 120 C einwirken lässt.
Die nach der vorliegenden Erfindung zu verwendenden Aminopropine können z. B. aus Dimethyl-, Diäthyl-, Cyclohexyl- oder ssenzylamin, aus Pyrrolidin, Piperidin oder iVIorpholin hergestellt werden.
Die Umsetzungen des 2-Cyclohexyliden- bzw. 2-Cyclohexyl-cyclohexanons mit Amino- propinen werden vorteilhaft in Gegenwart der bekannten Äthinylierungskatalysatoren ausgeführt. Als solche seien u. a. genannt, die Alkalihydroxyde, -amide oder -alkoholate, starke organische Basen, wie Piperidin, so wie Schwermetalle der 1.
Gruppe des periodi-- sehen Systems, wie Kupfer oder Silber, und deren Acetylide und zur Acetylbildung be fähigte Verbindungen: Man kann auch Ge rnische mehrerer dieser Katalysatoren ver wenden.
Die Äthinylierungsreaktionen erfolgen im allgemeinen bereits bei gewöhnlicher Tempe ratur rasch; zuweilen ist Erwärmen förder lich, in andern Fällen muss man die Um setzung durch Kühlen mässigen. Die Mit verwendung indifferenter Lösungs- oder Ver dünnungsmittel, wie Äther, Tetrahydrofuran, Dioxan oder Acetale, ist vorteilhaft.
Die nach dem beschriebenen Verfahren leicht und in ausgezeichneter Ausbeute aus billigen technischen Ausgangsstoffen zugäng lichen Produkte besitzen als solche oder in Form ihrer Salze mit anorganischen oder organischen Säuren oder in Form ihrer quaternären Ammoniiim.derivate wertvolle spasmolytische, atropinartige Eigenschaften.
Durch .die Quaternisierung werden die thera peutischen Eigenschaften der Verfahrens- endprodukte nicht nennenswert beeinflusst, dagegen wird die Löslichkeit erhöht. Einige dieser Produkte sind besonders als Heilmittel gegen die Parkinsonsche Krankheit wirksam.
Es ist bereits bekannt, dass das 2-Cy clo- hexyliden- und das 2-Cyclohexyl-1-(3'-piperi- dino,propyl)-cyclohexanol atropinartige Ei genschaften zeigen und als Mittel gegen die Pa.rkinsonsehe Krankheit brauchbar sind.
Man hat diese beiden basischen Carbinole bisher nur durch Umsetzen von N-(1-Halogen- propyl-[3] )-piperidin mit einem Metall, wie Magnesiiun, Cadmium oder Zink, und dann mit 2-Cyclohexyliden- bzw. 2-Cyclohexyl- cyclohexanon herstellen können (vgl. die ame rikanische Patentschrift Nr. 2590638). Das vorliegende Verfahren führt auf wesentlich einfachere und billigere Weise zu wertvollen Spasmolytika. Die in den folgenden Beispielen genann ten Teile sind Gewichtsteile.
<I>Beispiel 1</I> In ein Gemisch aus 300 Teilen Tetrahydro- furan, 80 Teilen fein gepulvertem Kalium hydroxyd und 35 Teilen Natriummethylat lässt man allmählich unter Rühren bei ge wöhnlicher Temperatur eine Lösung von 55 Teilen 1-Piperidino-propin-(2) in 80 Teilen 2-Cyclohexyliden-cyclohexanon einlaufen. Das Reaktionsgemisch erwärmt sich auf ungefähr 35 . Man rührt noch etwa 16 Stunden weiter, fügt dann 100 Teile Wasser hinzu und trennt die sich absetzende wässrig-alkalische Schicht ab. Die organische Schicht wird mit festem Ätzkali getrocknet.
Dann verdampft man das Lösungsmittel und destilliert unverän dertes 1-Piperidino-propin-(2) bei vermin dertem Druck ab (ungefähr 9 Teile).
Der verbleibende Rückstand (etwa 121 Teile) wird mit etwa 10 /aiger .Salzsäure verrührt, bis die Lösung kongosauer ist. Die erhaltene wässrige, durch nichtumgesetztes 2-Cyclohexyliden-cyclohexanon getrübte Lö sung wird mit Äther ausgeschüttelt; nach Verdampfen des Äthers gewinnt man etwa 12 Teile 2-Cyclohexyliden-cyclohexanon zu rück. Die wässrige Schicht wird bei vermin dertem Druck eingedampft; den Rückstand kristallisiert man aus einem Gemisch von Al "kohol und Äther um.
Man erhält 120 Teile reines 2-Cyclohexyliden-l-(3'-piperidino-pro- pinyl)-cyclohexa.nol-hydrochlorid vom Smp. 170-l72 . Aus dem Hydrochlorid wird mit Alkali die Base freigesetzt.
<I>Beispiel 2</I> In ein Gemisch aus 300 Teilen Tetra- @hydrofuran, 80 Teilen fein gepulvertem Ka- liumhydroxyd und 35 Teilen Natriummethy- lat lässt man allmählich unter Rühren bei gewöhnlicher Temperatur 35 Teile 1-Diäthyl- amino-propin-(2) einlaufen, wobei sich die Mischung von 25 auf etwa.
35 erwärmt. Hier auf versetzt man die Mischung im Verlauf einer halben Stunde mit 134 Teilen 2-Cyclo- hexyliden-cyclohexanon und rührt noch 16 Stunden weiter. Man gibt 100 Teile Wasser hinzu und trennt die wässrig-alkalische Schicht ab.
Die organische Lösung wird mit Ätzkali getrocknet und das Lösungsmittel verdampft, wobei gleichzeitig 12 Teile nicht umgesetztes Diäthylamin mit überdestillie- ren. Der Rückstand (160 Teile) wird mit etwa 10 /oiger Salzsäure verrührt, bis die Lö sung kongosauer reagiert. Die nichtbasischen Bestandteile werden durch dreimaliges Aus- äthern entfernt.
Hierauf wird die wä.ssrig- salzsaure Lösung mit starker Natronlauge alkalisch gemacht und das 2-Cyclohexyliden- 1- (3'-,diäthylamino-pr opinyl) -cyclohexanol-K 1) ausgeäthert. Nach Verdampfen des Äthers verbleiben 72 Teile roher Base.
20 Teile dieser Base werden in 100 Teilen absolutem Äther gelöst und mit 10 Teilen Methyljodid versetzt. Nach einigen Stunden fällt ein gelber Niederschlag aus, der in wenig Alkohol gelöst und mit der etwa 20fachen Menge absolutem Äther wieder aus gefällt wird. Man erhält farblose Kristalle vom Schmp. 167-168 , die reines 2-Cyclo- h.exyliden-1= (3'-diäthylaminopropinyl)-cycloa hexanol-(l.)-jodmethylat darstellen.
<I>Beispiel 3</I> Zu 25 Teilen Natriumdraht, die mit 1000 Teilen trockenem Äther bedeckt sind, gibt man unter Rühren 150 Teile 3-Pyrrolidino- propin. Man erhitzt das Gemisch einige Stunden am Rückfluss zum Sieden, gibt dann 180 Teile 2 - Cyclohexyliden - cyclohexanon hinzu und erhitzt das Ganze nochmals einige Stunden zum Sieden. Nach dem Erkalten versetzt man mit 250 Teilen Wasser, trennt die ätherische Schicht von der wässrig alkalischen Schicht ab, wäscht sie mit Wasser und destilliert nach dem Trocknen den Äther ab.
Man erhält 150 Teile 2-Cyclohexyliden-l- (3'-pyrrolidino-propinyl) - cyclohexanol, die unter 10-3 Torr bei einer Badtemperatur von 210 bei 150-156 übergehen.
<I>Beispiel</I> Man setzt in der im Beispiel 3 beschrie benen Weise 180 Teile 3-IIexamethylenimino- propin mit 180 Teilen 2-Cyclohexyliden-cyclo- hexanon und 25 Teilen Natrium um. Bei der Aufarbeitung, die wie im Beispiel 3 erfolgt, erhält man 220 Teile 2-Cyclohexyliden-1- (3' hexamethylenimino - propinyl) - cyclohexanol, das unter 10-3 Torr bei einer Badtemperatur von 220-230<B>0</B> bei 160-163<B>0</B> siedet.

<I> Example 1 </I> In a mixture of 300 parts of tetrahydrofuran, 80 parts of finely powdered potassium hydroxide and 35 parts of sodium methylate, a solution of 55 parts of 1-piperidino-propyne- ( 2) run in 80 parts of 2-cyclohexylidene-cyclohexanone. The reaction mixture warms up to approximately 35. The mixture is stirred for a further 16 hours, then 100 parts of water are added and the aqueous-alkaline layer which separates out is separated off. The organic layer is dried with solid caustic potash.
The solvent is then evaporated off and unaltered 1-piperidino-propyne- (2) is distilled off under reduced pressure (approximately 9 parts).
The remaining residue (about 121 parts) is stirred with about 10% hydrochloric acid until the solution is acidic to the Congo. The resulting aqueous solution, clouded by unreacted 2-cyclohexylidene-cyclohexanone, is extracted with ether; after evaporation of the ether, about 12 parts of 2-cyclohexylidene-cyclohexanone are recovered. The aqueous layer is evaporated under reduced pressure; the residue is recrystallized from a mixture of alcohol and ether.
