Die Verseifung wird vorzugsweise alkalisch in Wasser, organischen Lösungsmitteln, insbesondere niedermolekularen Alkoholen, oder Gemischen beider bei Temperaturen von 80 bis 200¯C, vorzugsweise 140 bis 180 C, durchgeführt. Die Decarboxylierung kann in einem besonderen Arbeitsgang stattfinden, erfolgt aber meist schon bei der Verseifung oder Destillation im Vakuum.
Die als Ausgangsstoffe ben¯tigten, Nitrilgruppen enthaltenden Ester von Säuren der oben angegebenen allgemeinen Formel erhält man z. B. durch Umsetzung von Cyclooetanon mit CyanessigsÏureester und Kondensation der Reaktionsprodukte mittels Natriumalkoho- lat mit Alkyl-, Alkenyl-, Alkinyl-, Cyclo alkenyl-oder Aralkylhalogeniden, wie Methyl- jodid, Äthylbromid, Propylbromid, n-Butylbromid, Dodecylbromid, Allylchlorid, Propa. rgylbromid, Cyclohexylchlorid, Cyclohexenylchlorid, Cyclooctylchlorid, ?1-Cyclooctenyl ehlorid oder Benzylehlorid. Auf diese Weise lassen sich z.
B. die folgend. en Cyanessigsäure- äthylester-derivate herstellen :

Die nach dem besehriebenen Verfahren hergestellten lmgesättigten Säuren sind in bezug auf die Lage der Doppelbindungen zum Teil Isomerengemische, wobei sich vorzugs- weise ein Gleichgewicht zwischen zwei Isomeren einstellt, und in alkalischer Lösung die ,
6-y-Stellung bevorzugt wird. Die isomeren Säuren lassen sieh gewünschtenfalls durch sorgfÏltige, fraktionierte Destillation oder andere iibliche VTethoden trennen.
Die nach dem vorliegenden Verfahren erhältliehen Säuren bilden mit anorganischen oder organischen Basen, wie Natriumhydroxyd, Magnesium-oder Calciumcarbonat, Ammoniak, DiÏthylamin, ¯thanolamin, TriÏthanolamin, Morpholin, Pyrrolidin, Hexylamin, Piperidin usw., wasserlösliche Salze, welehe die zur Verwendung als gallentreibende Mittel bevorzugten Verabreichungsformen darstellen. Ihre gallentreibende Wirkung übertrifft die Ïhnlich wirkender Produkte, z. B. die der Desoxycholsäure, bei weitem.
Die ?1-Cyclooctenyl-methyl- und -ÏthylessigsÏure und ihre Salze zeichnen sich durch besonders hohe Wirksamkeit allas. Die in die pharmakologisehe Prüfung miteinbezogene, analog gebaute Cyclooctancarbonsäure zeigt ebenfalls choleretische Eigenschaften, während die entsprechende Cyclohexancarbonsäure bei der verwendeten Versuchsweise praktisch wirkungslos ist.
Die in den nachstehenden Beispielen angegebenen Teile sind Gewichtsteile.
Beispiel 1
600 Teile ?1-Cyclooctenyl-Ïthyl-cyanessigsÏureÏthylester erhitzt man mit einer L¯sung von 1050 Teilen Kaliumhydroxyd in 985 Tei len 90%igem Alkohol 45 Stunden auf 160 bis 170 C. Nach dem Erkalten versetzt man das Reaktionsgemisch mit 2000 Teilen Wasser, filtriert und extrahiert zur Entfernung neutraler Anteile einmal mit Äther. Die Äther- Idsung wird abgetrennt und die wässrige alka lische Losung mit Salzsäure kongosauer ge- stellt.
Die sich dabei abscheidenden sauren organischen Anteile trennt man ab und extrahiert die wässrige Lösung noch einmal mit Ather. Nach Vereinigung der ¯therl¯sung mit den sauren organisehen Anteilen wäscht man noch einmal mit Wasser, trocknet über Na triumsulfat und verdampft das L¯sungsmittel.
Aus dem Rückstand erhält man durch Vakuumdestillation 350 Teile ?1-Cyclooctenyl äthylessigsäure als farblose viskose Flüssig- keit, die bei 122 bis 123 C unter 0, 4 mm
Druck siedet.
Den ?1-Cyclooctenyl-Ïthyl-cyanessigsÏure äthylester kann man auf folgende Weise er halten :
Man trägt 995 Teile Cyelooctyliden-eyan- essigsäureäthylester in eine Losung von 210
Teilen Natrium in 3480 Teilen absoluten Alkohols ein und lässt bei Raumtemperatur unter kräftiger Rührung 1395 Teile Diäthylsulfat mit soleher Geschwindigkeit zufliessen, dass die Reaktionstemperatur 700 C nicht überschreitet.
Nach beendeter Zugabe erhitzt man noch
6 Stunden auf 70 bis 75 C und destilliert ansehliessend den Alkohol ab. Der Rüekstand wird mit Benzol verdünnt und mit Wasser neutral gewasehen. Nach Verdampfen des
Benzols destilliert man den R ckstand im hohen Vakuum und erhält 945 Teile ?1-Cyclo oetenyläthyl-cyanessigsäure-athylester, die bei 113 bis 116¯C unter 0, 3 mm Druck sieden.
Beispiel 2
An Stelle der in Beispiel 1 angegebenen 600 Teile ?1-Cyclooctenyl-Ïthyl-cyanessigsÏure äthylester benutzt man als Ausgangsstoff 880 Teile ?1-Cyclooctenyl-methyl-cyanessigsÏure äthylester. Man erhält unter den in Beispiel : 1 angegebenen Bedingungen 430 Teile ?1-Cyclooctenyl-methylessigsÏure (Kp0.4 = 118 bis 122 C).
Den ?1-Cyclooctenyl-methyl-cyanessigsÏure äthylester kann man z. B. folgendermassen erhalten :
Man vermischt 995 Teile Cyclooctyliden evanessigsäureäthylester mit 1140 Teilen Dimethylsulfat, kühlt auf-8 C und lässt unter starker R hrung die HÏlfte einer Lösung von 217 Teilen Natrium in 2410 Teilen absolutem Methanol zufliessen. Nach wenigen Minuten steigt die Temperatur schnell auf 60 bis 65 C an. Sobald die Reaktionstemperatur abklingt, lässt. man die restliehe Natriummethylatlosung mit solcher Geschwindigkeit, zufliessen, dass die Innentemperatur 45 C nicht überschreitet.
Nach Zugabe des gesamten Methylates rührt man 1 Stunde bei Baumtemperatur und beendet die Umsetzung durch achtstündiges Er hitzen unter Rückfluss.
Das Methanol wird nun abdestilliert, der Rückstand mit Benzol verdünnt und mit Eiswasser. gewaschen. Man verdampft das Losungsmittel und erhält durch Destillation des Rückstandes unter hohem Vacuum 930 Teile ?1 Cyclooctenyl - methyl - cyanessigsÏureÏthylester mit einem Siedepunkt von 113 bis 1] 50 C bei 0, 4 mm.
