Verfahren zur Herstellung von Podophyllotoxinglucosid aus Podophyllumarten Es wurde gefunden, dass man aus den Rhizomen von Podophyllumarten, insbesondere aus der indischen, Podophyllum emodi Wall., ein Glucosid, Podophyllotoxinglucosid, gewinnen kann, dem eine mitosehemmende und tumorzerstörende Wirkung zukommt.
Dieses Glucosid wird hergestellt, indem man die durch Extraktion mit einem chlorierten Kohlenwasserstoff von der Harzfraktion befreite Droge mit einem wasserlöslichen Alkohol auszieht, den Extrakt konzentriert und dann mit -Wasser verdünnt, diese Lösung zur Trennung des CTerbstoff-Cllucosid-Komplexes mit gerbstoffällenden Mitteln behandelt, die Fällung abtrennt, die zuriiekbleibende Lösung vom organischen Lösungsmittel weitgehend befreit und mit -Wasser verdünnt, die wässerige Lösung zur Entfernung von Begleitstoffen mit. einem mit.
Wasser nicht, misehbaren Lösungsmittel extrahiert., dann das Glueosid mit einem mit Wasser nicht misehbaren Lösungsmittel, das höchstens 10 /a eines wasserlöslichen Alkohols enthält, ausschüttelt, das in die orga- nisehe Phase übergegangene Podophyllotoxin- glricosid nach Abdestillieren des Lösungsmittels in fester Form gewinnt und weiter reinigt.
Es war bekannt., da.ss die Rhizome von Podophyllumarten Wirkstoffe enthalten, die Krebsgewebe in therapeutisch günstigem Sinne beeinflussen können. Die wichtigste von den bisher untersuchten wirksamen Verbindungen ist das in Wasser praktisch unlösliche Podophyllotoxin, das aus der Harzfraktion der Droge isoliert werden konnte. Es war aber bisher nicht gelungen, aus Podophyllum irgendein Glykosid von einer der wirksamen Komponenten zu erhalten.
Nach dem erfindungsgemässen Verfahren, das besonders auch der Empfindlichkeit und der leiehten enzymatischen Spaltbarkeit von glykosidisclren Wirkstoffen Rechnung trägt, kann aus Podophyl- luma.rten ein Glueosid erhalten werden, das in Wasser löslich ist und als Podophyllotoxin-fl- D-glucosid identifiziert werden konnte.
Dass therapeutisch wichtige pflanzliche Drogen, deren wirksaanes Prinzip zu den Glucosiden gehört, diese in Form von Komplexen an Gerbstoffe gebunden enthalten, war schon länger bekannt. Die sehweiz. Patentschrift Nr.102141 beschreibt ein Verfahren zur Herstellung der herzwirksamen Glucoside aus der Meerzwiebel. Hierbei wird der Droge der GIucosid-Gerbstoff-Komplex durch Extraktion entzogen und in dem Extrakt durch Behandlung mit gerbstoffällenden Mitteln aufgetrennt. In der Schweiz.
Patentschrift Nr.130021 wird die Isolierung und Reinigung dieses Glueosid-Gerb'stoff-Komplexes in fester Form beschrieben sowie die Gewinnung des reinen Glucosidgemisches daraus durch Fällung des Gerbstoffes. Die Schweiz. Patentschrift Nr.134217 mit Zusatzpatent Nr.142364 hat ein Verfahren zum Gegenstand für die Auftrennung des Glucosidgemisehes der Meer-

zwiebel, das auf der verschiedenen Löslichkeit der Glucosid-Gerbstoff-Komplexe, der sogenannten Reintannoide , in gewissen Lösungsmitteln oder Lösungsmittelgemischen beruht.
Auch hier werden die reinen Glucoside schliesslich durch Behandlung der Reintannoide mit gerbstoffällenden Mitteln gewonnen. Auch die Glucoside der Digitalisblätter liegen in der Pflanze an Gerbstoffe gebunden vor und können erst durch Fällung der Gerbstoffe rein dargestellt werden. Siehe schweiz. Patentschrift Nr.154657. Auch das bisher unbekannte mitosehem- mende und tumorzerstörende Podophyllotoxin- glucosid liegt in den R.hizomen der Podophyl- lumarten als Glucosid-Gerbstoff-Komplex vor. Dieser Komplex wird bei der Behandlung mit gerbstoffällenden Mitteln, z. B. Bleisalzen, nach dem vorliegenden Verfahren aufgetrennt, wonach das freie Glucosid aus dem Filtrat von der Fällung nach bekannten Methoden gewonnen und gereinigt werden kann.
Es hat sich im vorliegenden Fall als nicht notwendig, ja sogar als nicht zweckmässig erwiesen, den Glucosid-Gerbstoff-Komplex vor seiner Auftrennung in festem Zustand zu isolieren. Das Verfahren gemäss der vorliegenden Erfindung arbeitet viel rationeller, wenn der genannte Komplex in der durch Extraktion der Droge erhaltenen Lösung direkt durch Behandlung mit gerbstoffällenden Mitteln aufgetrennt wird.
Das Podophyllotoxin-gluco- sid hat die Zusammensetzung C28H3201s und schmilzt bei 149-152 . Es zeigt in MethanollOsung die optische Drehung [a1 ri = -75 , in Pyridinlösung [cc1D = -117 . Das Gluco- sid ist in Alkohol und in Wasser löslich. Bei der enzymatischen Spaltung und bei der Hydrolyse mit Säure wird das Glueosid in Podo- phyllotoxin und D-Glucose zerlegt.
Das verfahrensgemäss aus Podophyllum- arten dargestellte neue Glucosid, das sich gegenüber den bis jetzt bekannten zuckerfreien Wirkstoffen besonders durch seine Löslichkeit in Wasser auszeichnet, soll für therapeutische Zwecke verwendet werden. Beispiel Getrocknete Podophyllumrhizome werden fein gemahlen und dann mehrmals bei gewöhnlicher Temperatur mit Chloroform extraliiert.. Das so vorgereinigte Pflanzenmaterial wird nun mit Methanol ausgezogen, worauf man die vereinigten gelb gefärbten Auszüge auf etwa 1/3 konzentriert Lind dann mit dem gleichen Volumen Wasser versetzt.
Die wässe- rig-methanolische Lösung wird nun zur Trennung des Gerbstoff-G@lueosid-Komplexes mit. einer konzentrierten wässerigen Lösung von Bleiacetat, solange eine Fällung entsteht, versetzt. Nach dem Abfiltrieren der entstandenen Fällung wird die Lösung durch Konzentrieren im Vakuum möglichst vollständig von Methanol befreit und mit Wasser verdünnt und dann mehrmals mit etwas reinem Chloroform ausgeschüttelt.
Diese Vorfraktion enthält neben harzigen Begleitstoffen auch schon kleine Mengen des neuen Glucosids. Die wässerige Restlösung wird anschliessend mehrmals mit Chloroform, dem 10 /o Alkohol zugesetzt sind, ausgeschüttelt, worauf man die vereinigten, fast farblosen Chloroformauszüge eindampft, den Eindampfrückstand mit Äther wäscht und trocknet.
Dieses Clucosidpräparatwird in Essigester gelöst und zur Reinigung in einer Säule aus Silicagel chromatographiert. Beim Eluieren der Säule mit Essigester, dem 1-5 0/0 Methanol beigemischt sind, erhält man nach einer Vorfraktion, die hauptsächlich noch gefärbte, etwas harzige Anteile enthält, Fraktionen, die das Podopliyllotoxin-glucosid enthalten. Das Glucosid wird daraus nach Eindampfen und Umfällen aus Aceton-Äther in reiner Form erhalten. Dieses Präparat besitzt die oben erwähnten Eigenschaften.
Die Ausbeute an Podophyllotoxin-glucosid aus den getroekne- ten Rhizomen (Handelsdroge) beträgt etwa 0,5-10/0.

The method according to the present invention works much more efficiently if the said complex in the solution obtained by extraction of the drug is separated directly by treatment with tannic agents.

This clucoside preparation is dissolved in ethyl acetate and chromatographed in a column of silica gel for purification. The glucoside is obtained therefrom in pure form after evaporation and reprecipitation from acetone-ether. This preparation has the properties mentioned above.
