Verfahren zur Herstellung von Verbindungen der Tropasäurereihe
Es wurde gefunden, dass man zu neuen Verbindungen der Tropasäurereihe gelant, wenn man gesättigte oder ungesättigte Bicycloalkanone mit Organometallverbindungen eines plienylessigsauren Salzes umsetzt und die so erhaltenen Tropasäureabkommlinge durch Umsetzung mit N, N'-disubstituierten Aminoalkylhalogeniden, deren Alkylgruppe den Aithyl-oder Propylrest bedeuten kann, in die entsprechenden substituierten Aminoalkylester überführt und diese quaternisiert. Dabei ist der Stickstoff der Aminogruppe vorzugsweise durch niedrige Alkylgruppen substituiert.
Diese neuen quaternisierten bicyclischen Abkömmlinge der Tropasäure zeiehnen sich durch wertvolle therapeutische Eigenschaften aus ; sie weisen bemerkenswerte spasmolytisehe Eigenschaften hinsichtlich der Stärke und der Dauer ihrer Wirkungen bei verhältnismässig geringer Giftigkeit und geringen Nebenwirkungen auf.
Man gewinnt beispielsweise das quartäre Salz des DiÏthylmethylammoniumÏthylesters der [2-Oxy-bicyclo (2, 2, 1) heptyl-2]-phenyl- essigsäure, indem man Bicyclo (2, 2, 1) heptanon-2 mit dem Natriumsalz der Brommagnesium- phenylessigsäure umsetzt und die gewonnene freie Säure in den entsprechenden Diäthyl- aminoÏthylester und diesen in das quaternäre Salz des Diäthylmethylammoniumäthylesters überführt.
Die quaternisierten Verbindungen der Tropasäurereihe sind meist wasserlöslich und gut kristallisierbar.
Beispiel 1 [2-Oxy-bicyclo (2, 2, 1) heptyl-2]- phenylessigsäure
79, 6 g fein pulverisiertes, troekenes phenylessigsaures Natrium werden unter guter K hlung im Verlaufe von 2 Stunden in eine ätheri- sche Lösung von Isopropylmagnesiumbromid, dargestellt aus 21, 0 g Magnesiumspanen und 111 g 2-Brompropan in 700 cm3 trockenem Äther, eingetragen, wobei unter Propanentwicklung die Organomagnesiumverbindung der Phenylessigsäure gebildet wird. Darauf werden 55, 0 g Bicyclo (2, 2, 1) heptanon-2, (C7HtoO) in 450 cm3 trockenem ¯ther hinzugetropft und zum Schluss die Reaktion durch Erwärmen am Rückfluss zu Ende geführt.
Das Reaktionsgemisch wird in der Kälte mit verdünnter Mineralsäure behandelt, die ätherische Schicht abgehoben und derselben mit verdünnter Natronlauge die freie Säure entzogen. Auf Zusatz von verdünnter Mineralsäure zur wässrigen Lösung des Natriumsalzes scheidet sich die bicyclisch substituierte Phenylessigsäure zuerst als dickes 61 ab, das nach einiger Zeit völlig erstarrt. Die Säure C5Ht803 kristallisiert aus Aceton-Hexan in büschelförmig angeordneten Nadeln vom F. 170 C. Ausbeute 65-75% der Theorie.
Das gesuchte Hydrochlorid des Dimethyl- aminoäthylesters, C gH2703N. HC1, kristalli- siert aus Methanol-Äther in quadratischen Plättchen vom F. 143¯C. Ausbeute 65-75% der Theorie.
[2-Oxy-bieyclo (2, 2, 1)heptyl-2]-phenylessig säure-trimethylammoniumäthylester methylsulfat
Zur Gewinnung eines quartären Salzes werden 13, 9 g der freien tertiären Base in 150 em3 Äther gelost und mit 7, 5 g Dimethylsulfat versetzt. Das Methomethylsulfat C21H33O7NS kristallisiert aus Methylacetat- Äthylaeetat in langen Säulen vom F. 145¯ C.
Ausbeute 75-85% der Theorie.
Beispiel 3 [2-Oxy-bicyclo (2, 2, 1) heptyl-2]-phenylessig säure-diäthylamino-propylesterhydrochlorid 80, 7 g Natriumsalz der in Beispiel 1 be schriebenen SÏure C15H18O3 und 56 g 1, 3-Di äthylaminopropylehlorid-hydrochlorid werden nach Beispiel 1 in 480 cm3 Propanol-2 zur Umsetzung gebracht und aufgearbeitet. Das Hydroehlorid des 1, 3-Diäthylaminopropylesters, C, 2H3303N. HCI, kristallisiert aus Ace ton-Äther in langen Prismen vom F. 111 bis 113"C. Ausbeute 75% der Theorie.
[2-Oxy-bicyelo (2, 2, 1) heptyl-2]-phenylessig- säure-diäthylmethylammonium-propylester methylsulfat
Durch Behandeln der freien Base C22H3303N mit Dimethylsulfat in Äther ge- winnt man das Methomethylsulfat, C24H39O7NS, das aus Methylacetat-Äthylaeetat in quadrati- schen PlÏttchen vom F. 103-105 C kristallisiert. Ausbeute 80-90% der Theorie.
Beispiel 4 [2-Oxy-?5-bicyclo(2, 2, 1) heptenyl-2]- phenylessigsäure
79, 6 g phenylessigsaures Natrium werden nach Beispiel 1 in die entspreehende Organomagnesiumverbindung übergeführt und mit [2-Oxy-bicyelo (2, 2, 1) heptyl-2]-phenylessig säure-diäthylaminoäthylester-hydrochlorid
123 g der SÏure C15H18O3 werden in eine Lösung von 11, 5 g Natrium in 400 cm3 Propanol-2 eingetragen und hierzu 86 g Diäthyl- aminoäthylchlorid-hydrochlorid in 400 em3 Propanol-2 hinzugefügt.
Das Gemisch wird 36 Stunden am Rückfluss gekocht und nach Entfernung des gebildeten Natriumchlorids die Lösung auf dem Wasserbad unter vermindertem Druck eingeengt. Durch Behandeln des Rückstandes mit troekenem Äther gewinnt man das Hydrochlorid des Diäthylamino Ïthylesters, C21H31O3. HCl, welches aus ¯thylacetat in schwach hygroskopischen langen Nadeln vom F. 132, 5 C kristallisiert. Ausbeute 75% der Theorie.
[2-Oxy-bicyclo (2, 2, 1) heptyl-2]-phenylessig säure-diäthylmethylammoniumäthylester- methylsulfat
Zur ¯berf hrung der tertiären Base in das Methomethylsulfat werden 34, 5 g der freien Base in 250 cm3 trockenem ¯ther gelöst und die Lösung mit 15, 1 g Dimethylsulfat versetzt.
Nach einiger Zeit scheidet sich das quartÏre Salz in fester Form nahezu vollständig ab, welches in Wasser, Methanol und Aceton leicht löslich ist, wenig löslich in Äthylacetat und unlöslich in Äther. Es kristallisiert aus einem Gemisch von Methylacetat-Äthylacetat in derben Drusen vom F. 114, 5 C. In gerin ger Menge erhält man eine Modifikation dieses Salzes, welche aus Äthanol-Äthylacetat in sechseckigen Blättchen vom F. 133 C kristallisiert. Ausbeute 85-90% der Theorie.
Beispiel 2 [2-Oxy-bicyclo (2, 2, 1) heptyl-2]-phenylessig- sÏure-dimethylaminoÏthylester-hydrochlorid 74 g der in Beispiel 1 beschriebenen SÏure C15H18O3 gel¯st in 240 em3 Propanol-2, werden mit einer Lösung von 6, 9 g Natrium in 240 cm3 Propanol-2 versetzt. Zu dieser Lösung fügt man 43, 2 g Dimethylaminoäthylchlorid-hydro- chlorid in fester Form hinzu und kocht 36 Stunden am Rückfluss. Die Aufarbeitung erfolgt nach Beispiel 1.
5. g S5-Bicyclo (2, 2, 1) heptenon-2, CTH8O, in 450 cm Äther umgesetzt. Das Reaktionsgemisch wird nach Beispiel 1 aufgearbeitet, und man erhÏlt die ungesÏttigte SÏure C15H16O3 aus Aceton-Petroläther in langen Prismen vom lui. 148, 5 C. Ausbeute 20-30% der Theorie.
[2-Oxy-@5-bicyclo (2, 2, 1) heptenyl-2]-phenyl essigsäure-däthylamino-äthylester
122 g der freien Säure C15H16O werden nach Beispiel 1 in eine Lösung von 11, 5 g Natrium und 400 cm3 Propanol-2 eingetragen und diese Losung mit 86 g Diäthylaminoäthyl- chlorid-hydrochlorid in 400 cm3 Propanol-2 36 Stunden am Rüekfluss gekocht. Das Reaktionsgemisch wird entsprechend aufgearbeitet.
Das Hydroehlorid des Diäthylaminoäthyl esters, C2tH2903N. HCl, ist ein schwach hygroskopisches Salz, welches aus Athylacetat in Prismen vom F. 103 C kristallisiert. Ausbeute 60-70% der Theorie.
[2-Oxy-?5-bicyclo (2, 2, 1) heptenyl-2]-phenyl essigsäure-diäthylmethylammoniumäthylester- bromid bzw.-methylsulfat
Das Methobromid der quartären Base, C22H32O3NBr, vom F. 130, 5¯ C, wird durch Anlagerung von 9, 5 g Methylbromid an 38 g der tertiären Base in ätherischer Lösung erhalten. Es ist leicht löslich in Wasser und Methanol, wenig löslich in Aceton und unlöslich in Äther. Ausbeute 90% der Theorie.
Das Methomethylsulfat, C23H35O7NS, in entsprechender Weise aus der tertiären Base mit Dimethylsulfat dargestellt, kristallisiert aus Aceton-Äther in kleinen Plättchen vom F. 120-121 C. Ausbeute 85-95 % der Theorie.
Process for the preparation of compounds of the tropic acid series
It has been found that new compounds of the tropic acid series are obtained if saturated or unsaturated bicycloalkanones are reacted with organometallic compounds of a plienylacetic acid salt and the tropic acid derivatives obtained in this way are reacted with N, N'-disubstituted aminoalkyl halides, the alkyl group of which can be the ethyl or propyl radical , converted into the corresponding substituted aminoalkyl esters and quaternized. The nitrogen of the amino group is preferably substituted by lower alkyl groups.
These new quaternized bicyclic derivatives of tropic acid are distinguished by valuable therapeutic properties; they have remarkable spasmolytic properties with regard to the strength and the duration of their effects with relatively low toxicity and few side effects.
For example, the quaternary salt of the diethylmethylammonium ethyl ester of [2-oxy-bicyclo (2, 2, 1) heptyl-2] -phenylacetic acid is obtained by mixing bicyclo (2, 2, 1) heptanone-2 with the sodium salt of the magnesium bromide converts phenylacetic acid and the free acid obtained is converted into the corresponding diethyl amino ethyl ester and this into the quaternary salt of diethyl methyl ammonium ethyl ester.
The quaternized compounds of the tropic acid series are mostly water-soluble and easily crystallizable.
Example 1 [2-Oxy-bicyclo (2, 2, 1) heptyl-2] - phenylacetic acid
79.6 g of finely powdered, dry sodium phenylacetic acid are added to an ethereal solution of isopropylmagnesium bromide, prepared from 21.0 g of magnesium shavings and 111 g of 2-bromopropane in 700 cm3 of dry ether, under good cooling over the course of 2 hours, with the evolution of propane, the organomagnesium compound of phenylacetic acid is formed. 55.0 g of bicyclo (2, 2, 1) heptanone-2, (C7HtoO) in 450 cm3 of dry ether are then added dropwise and finally the reaction is brought to an end by refluxing.
The reaction mixture is treated in the cold with dilute mineral acid, the ethereal layer is lifted off and the free acid is removed from it with dilute sodium hydroxide solution. When dilute mineral acid is added to the aqueous solution of the sodium salt, the bicyclically substituted phenylacetic acid first separates out as thick 61, which solidifies completely after a while. Example 3 [2-Oxy-bicyclo (2, 2, 1) heptyl-2] -phenylessig acid diethylamino-propyl ester hydrochloride 80.7 g of the sodium salt of the acid C15H18O3 described in Example 1 and 56 g of 1,3-diethylaminopropyl chloride hydrochloride are reacted according to Example 1 in 480 cm3 of 2-propanol and worked up. The mixture is refluxed for 36 hours and, after the sodium chloride formed has been removed, the solution is concentrated on a water bath under reduced pressure. The hydrochloride of diethylamino ethyl ester, C21H31O3, is obtained by treating the residue with dry ether. HCl, which crystallizes from ethyl acetate in weakly hygroscopic long needles with a melting point of 132.5 ° C. Yield 75% of theory.
[2-Oxy-bicyclo (2, 2, 1) heptyl-2] -phenyl-acetic acid diethylmethylammonium ethyl ester methyl sulfate

[2-Oxy-? 5-bicyclo (2, 2, 1) heptenyl-2] -phenylacetic acid diethylmethylammonium ethyl ester bromide or methyl sulfate
The methobromide of the quaternary base, C22H32O3NBr, with a melting point of 130.5¯ C, is obtained by adding 9.5 g of methyl bromide to 38 g of the tertiary base in an ethereal solution.
