Die Reduktion der Tropinone zu den Tropinen kann durch katalytiselie Hydrierung niit oder ohne Druck, bei normaler oder bei erhöhter Temperatur, aber auch durch Na trium in einem Alkohol durchgeführt werden.
Katalytische Hydrierung führt zu Verbin dungen der Tropinreihe, Reduktion mit Na trium und Alkohol zu mP-Tropinderivaten. Die Aeylierung der 6-Alkoxy-tropine wird vorzugsweise und in guter Ausbeute durch Alkoholyse der Alkylester der Säuren der Formel HOOC - R;, durchgeführt.
Das Verfahren wird beispielsweise so aus geführt: Ein 6-Alkoxy-tropinon wird in abs. Methanol oder Äthanol oder ohne Lösungs mittel mit Raney-Nickel als Katalysator bei leicht. erhöhter Temperatur unter Druck hydriert. Das auf übliche Weise isolierte und durch Fraktionierung im Vakuum. gereinigte 6-Alk oxy-tropin wird durch L: mesterung mit.
einem Alkylester einer Säure der Formel HOOC - R;, in den. 6- Alkoxy-tropinester über- 0, efübrt.
Die nach dein vorliegenden Verfahren er lialtenen neuen Ester von 6-Alkoxy-tropinen sind wegen ihrer starken pharmakodyna- mischen Wirkung als - Medikamente mit Seopolaminwirkung zu verwenden, dienen aber auch als Zwischenprodukte zur Her stellung solcher.
<I>Beispiel Z</I> 6-Methoxy-tropin-benzilsäureester 5 g 6-IVTethoxi>-tropinon-hy droehlorid wer den in 50 cin'3 abs. Methanol im Autoklav en finit R.aney-Niekel während 5 Stunden bei 40-45 und einem Anfangsdruck von 60 at hydriert.
Man filtriert vom Katalysator ab und verdampft den grössten Teil des Lösungs mittels im Vakuum, wobei das 6-Methoxy- tropin-hydrochlorid, CaHi30.1\TCl, auskristal- lisiert. Naeh dem Umkristallisieren aus Metha- noli'Äther zeigt es einen Smp. von 205-206 .
Die Hydrierung kann ebensogut mit der freien Base ausgeführt werden.
Man erhitzt eine Mischung von 7,29 g 6-Methoxy-tropin und 21,9 g Benzilsäure- äthy lester mit 0,2 g Natriummetall während 30 Stunden im Vakuum. (etwa 2 mm) auf 125-130 . Die anfangs starke, durch abdest.il- lierendes Äthanol verursachte Blasen- und Schaumbildung hört. gegen Ende der Reak tionszeit vollständig auf.
Nach dem Abkühlen wird das Reaktionsgemisch in 80 cm3 2n-Salz- säure aufgenommen und mit total 200 cin3 Äther extrahiert. Man macht die wässerige salzsaure Schicht mit 25 /oiger Ammonium- hydroxy dlösung alkalisch, extrahiert den ausfallenden 6 - Methoxy - tropin - benzilsäure- ester mit insgesamt 300 cm3 Chloroform,
trocknet die vereinigten Chloroformextrakte über Natriumsulfat und destilliert das Chloro form ab. Nach kurzem Stehen kristallisiert der Rückstand durch. Smp. 98-100 . Nach dem Umkristallisieren aus Benzol/Petroläther und Benzol allein schmilzt der 6-Methoxy- tropin-benzilsäureester, C23Hz70,IN, bei 99 bis 101 .
6-llet.lioxy - tropin-benzilsäureester - hydro- ehlorid, Smp. 146-148 aus Methanol;'Äther.
6-Methoxy - tropin-benzilsäureester - 1y dro- bromid, Smp. 197-199 aus Methanol/Äther. Beispiel <I>2</I> 6-Methoxy-tropin-benzoesäiireester Eine Mischung von 16,6 g gemäss Beispiel 1 hergestelltem 6-Methoxy-tropin und 29,1 g Benzoesäureäthylester wird mit 0,3 g Natrium während 30 Stunden im Vakuum (etwa 10 mm) auf 125-130 erhitzt.
Die zu Beginn starke Schaumbildung durch abdestillierendes Äthanol hört gegen Ende der Reaktionszeit vollständig auf. Nach Abkühlen wird das Reaktionsgemisch in 150 cm3 2n-Salzsäure gelöst und mit total 90 em3 Äther extra hiert. Die wässerige salzsaure Lösung wird darauf mit 25 obiger Ammoniumhy droxyd- lösung alkalisch gestellt und mit insgesamt.
100 eni3 Chloroform ausgesehüttelt. Die ver einigten Chloroformextrakte werden über Natriumsulfat getroeknet und das Chloro form im sehwachen Vakuum abdestilliert.
6-Methoxy-tropin-benzoesä.ureester - hydro- ehlorid, Smp. 216-218 aus Aeeton. 6-Methoxy-tropin-benzoesäureester-hy dro- bromid, Smp. 227-230 aus Methanol/Äther. <I>Beispiel 3</I> 6-Methoxv-tropin-veratrumsäureester Man erhitzt eine Mischung von 5,5 g emäss Beispiel 1 hergestelltem 6-Methoxy- tropin und 13,
5 g Verati#umsätireäthy lester mit etwa 0,2 g Natriummetall während 30 Stunden im Vakuum (5-10 mm Hg) auf 125-130 . Die anfangs starke, durch ab destillierendes Äthanol verursachte Blasen- und Schaumbildung hört gegen Ende der Reaktionszeit vollständig auf.
Nach dem Ab kühlen wird das Reaktionsgemisch in 40 em3 2n-Salzsäure aufgenommen und mit total <B>110</B> eins Äther extrahiert. -Man macht die wässerige salzsaure Schicht unter Eiskühlung mit 25 /oiger Ainmoniumhy droxy dlösung alka lisch, extrahiert den 6 - Methoxy - t.ropin- veratrumsäureester mit insgesamt 150 em3 Chloroform,
trocknet die vereinigten Chloro- formextrakte über Natriumsulfat und dampft das Chloroform im Vakuum ab. Nach kurzem Stehen kristallisiert der Rückstand, 6-Met.hoxy- tropin-veratrunisäureester, CisH.2505N, durch. Sinp. 96-98 nach dem Umkristallisieren aus Benzol/Petroläther.
Die Reinigung des Esters geschieht am besten über das .6-Methox,##-tropin-v eratrumsä.ureester- liy drobromid, Snip. <B>209-2110</B> aus 1e thänot Äthe r.
6-Methoxy -tropiii-v eratrunisäui-eester- hydrochlorid, Smp. 219-221 aus MethanollÄ ther. <I>Beispiel</I> 6-Methoxy-tropin-mandelsäureester (6-Methoxy-homatropin) Man erhitzt eine Misehung von 3,4 g gemäss Beispiel 1 hergestelltem 6-llethoxy-tropin, 7,2 g llaridelsäureäthylester und etwa 0,
1 g N at.riummetall während 30 Munden im Va kuum (etwa. 5 nun) auf 125-130 . Nach be endeter Reaktion wird das Reaktionsgemisch in 40 em3 2n-Salzsäure aufgenommen und mit total 100 ein- Äther ausgeschüttelt. Anschlie ssend wird noch mit insgesamt 100 cm3 Chloro form extrahiert.
Die saure wässerige Lösung wird darauf mit 100 cm3 Chloroform unter schichtet, unter Eiskühlung mit. gesättigter Sodalösung schwach alkalisch gestellt (schwach phenolphthaleiilalkaliseh) und mit total .100 ein-' Chloroform extrahiert. Man trocknet die verei- lii"rtenChloroformextrakte über 7.\7atriunisulfat und destilliert das Chloroform im Vakuum ab. Aus dem Rückstand, dem 6-Methoxy- tropin-mandelsäureester, werden direkt die Salze hergestellt.
Aus der schwach alkalischen wässerigen Lösung wird durch Versetzen mit konz. Natronlauge, Sättigen mit Kochsalz und Extrahieren mit Chloroform eine geringe Menge 6-Methoxy-tropin zurückgewonnen (l20 nig). Der saure ätherische Auszug wird über Natriumsulfat.
getrocknet, und der Äther abdestilliert. Nach Verseifen des Rück standes mit Natronlauge und Ansäiuern wer den noch 1,8 g Mandelsäure zurüekgewonnen. Beispiel <I>5</I> 6-Äthoxy-tropin-benzilsäureestei# 7 g 6-Äthoxy-tropinon-hydrochlorid wer den in 50 em3 abs. Äthanol im Autoklav en mit Raney-Niekel während 5 Stunden bei 40-.15 und 60 at hydriert.
Man filtriert vom Katalysator und destilliert den Alkohol ili schwachem Vakuum ab. Der ölige R.ück- #tand wird in 10 em3 Nasser aufgenommen, mit gesättigter Sololösung alkalisch gestellt, mit Kochsalz gesättigt und das 6-Äthoxy - tropin mit total 60 em3 Chloroform extrahiert.
Nach Trocknen über Natriumsulfat und Ab destillieren des Chloroforms wird der Rück stand im Hoehvakuum destilliert. Sdp. 106 bei 0,7 mm Hg.
Eine Mischung von 4,3 g 6-Äthoxy-tropin und 11,95g Benzilsäureäthylester wird mit etwa 0,2 g Natriummetallwährend 30 Stun den im Vakuum (etwa 1 mm) auf 125-130 erhitzt. Gegen Ende der Reaktionszeit ist keine Blasenbildung mehr feststellbar. Nach Abkühlen wird das Reaktionsgemisch in 40 cin3 2n-Salzsäure aufgenommen und mit total 110 em3 Äther extrahiert.
Die wässerige salzsaure Lösung wird darauf mit 80 cm3 Chloroform unterschielitet, eisgekühlt und mit 25'o/oiger Ammoniulnhydroxydlösung alkalisch gestellt. Darauf wird mit. - total 400 em3 Chloroform ausgeschüttelt und die vereinigten Chloroformextrakte über Natriumsulfat ge trocknet. Nach Abdestillieren des Chloro forms hinterbleibt ein öliger brauner Rück stand. Dieser wird in wenig Äthanol gelöst und durch Tierkohle filtriert.
Beim Einengen kristallisiert der 6-Äthoxy-tropin-benzilsäure- ester, Smp. 132-134 , aus Äthanol. Das 6-Äthoxy-tropin-benzilsäureester- hydroehlor id schmilzt unter Zersetzung bei 100 nach Um- kristallisieren aus .lthaiiol!ilther, bei 186 bis <B>1870</B> aus Aceton.
<I>Beispiel 6</I> 6-Äthoxy-tropin-benzoesäureester Man erhitzt eine Mischung von 4,26 g gemäss Beispiel 5 hergestelltem 6-Äthoxy- t.ropin und 6,9 g Benzoesäureäthylester mit etwa 0,1 g Natriummetall während 30 Stun den im Vakuum (12 mm) auf 125-130 . Nach dem Abkühlen wird das Reaktions- glemiseh in 40 cm3 2n-Salzsäure aufgenommen und mit total 100 em3 Äther extrahiert.
Die wässerige saure Lösung wird darauf mit 50 cm3 Chloroform unterschichtet, eisgekühlt und mit gesättigter Sodalösung schwach alka- liseh (Phenolphthalein) gestellt. Man extra hiert mit insgesamt 270 ems Chloroform, trocknet die vereinigten Chloroformextrakte über Natriumsulfat und destilliert. das Extraktionsmittel in schwaehem Vakuum ab.
Der ölige Rückstand (7,3 g) wird in 30 ein3 Äthanol aufgenommen und über Tierkohle und Aluminiumoxyd gereinigt.
Das 6 - Äthoxy-tropin - benzoesäur eester- hydrobromid besitzt den Smp. 192-194 nach Umkristallisieren aus @lthanol,"Äther.

Die wässerige Lösung wird darauf mit 50 em3 Chloroform unterschichtet, mit Eis (1#-ekühlt, mit 25 o/o.iger Ammoniumhydroxyd- lösung alkalisch gestellt und mit total 280 cm3 Chloroform extrahiert. Die vereinigten Chloro- formextrakte werden über Natriumsulfat ge trocknet, das Extraktionsmittel im Vakuum abgedampft und der ölige, braune Rückstand über Tierkohle und Aluminiumoxyd gereinigt.
