Verfahren zur Herstellung von N-Allyl-normorphin. N-Allyl-normorphin, in Mischung mit Morphin angewendet, hemmt oder verhindert die Wirkung dieses Alkaloides. Wenn es aber allein verwendet wird, besitzt. es eine anal "etische Wirkung, die derjenigen des Mor phins gleichkommt.
Bisher wurde N-Allyl-normorphin her gestellt, indem in einem organisehen Lösungs- niittel zwei Moläquivalente Normorphin mit einem Moläquivalent eines Allylhalogenides zur Reaktion gebracht. wurden. Dieses Vor gehen. bringt. den Nachteil mit. sich, dass im geschlossenen Gefäss gearbeitet werden muss.
Ausserdem war es bei diesem bisherigen Ver fahren zum Erreichen einer ins Gewicht. fal lenden Ausbeute unerlä.sslich,zwei 1@loläqui- valente Normorphin auf jedes Mol Allylhalo- renid anzuwenden. Die Gewinnung des N Allyl-normorphins aus der Reaktionsmi- seliuna- wurde durch die Notwendigkeit erschwert,
das N-Allyl-normorp-hin von dem als Nebenprodukt entstehenden Normorphin- hydrobromid zu trennen, welches sich in etwa lleieher Menge im Reaktionsgemisch vorfand wie das N-Allyl-normorphin. Die Gesamt- ausbeuten an N-Allyl-normorphin waren bei diesem. früheren Verfahren deshalb im allge meinen sehr gering, etwa zwischen 20 und 25% der Theorie.
Es wurde nun gefunden, dass sieh ausser- ordentlieli hohe Ausbeuten an N-Allyl-nor- morphin, welche sich den theoretisch erreich baren Werten annähern und jedenfalls über 85% der Theorie betragen, erzielen lassen,
wenn man erfindungsgemäss Normorphin und ein Allylhalogenid in mindestens annähernd äquimolarem Verhältnis zur Reaktion bringt (statt einen bisher als unerlässlich betrach- teten überschuss von 100% Normorphin zu verwenden) und in einem praktisch wasser freien organischen Lösungsmittel und in Ge genwart eines molaren Überschusses eines säurebindenden Stoffes arbeitet.
Die Mi schung wird am besten bis zur Beendigung der Reaktion erhitzt. Als Allylhalogenid kann Allylchlorid, Allylbromid usw., als säurebin dender Stoff etwa Natriumbicarbonat. und als Lösungsmittel z. B. Äthanol verwendet wer den.
Vorzugsweise wird-wie folgt vorgegangen: Eine Lösung von äquimolaren Mengen von Normorphin und einem Allylha.logenid, vor zugsweise Allylbromid, in Alkohol wird unter Rühren am Rückfluss in Gegenwart eines molaren Überschusses von Natriumbicarbonat während einiger Stunden erhitzt. Im allge meinen werden.
etwa 1,5 Moläquivalente Bi- carbonat verwendet. Unter diesen Umständen ist die Reaktion etwa nach fünf- bis sechs stündigem Erhitzen beendet und das N-Allyl- normorphin entsteht in einer Ausbeute von etwa 85% der Theorie. Es wird aus der Reaktionsmischung auf übliche Weise gewon- nee,
indem die Lösung eingedampft wird, vorzugsweise unter verringertem Druck, worauf der Rückstand mit einem heissen chlorierten Kohlenwasserstoff als Lösungs- mittel, etwa mit Chloroform, extrahiert wird. Die erhaltene Lösung, die durch Behandlung mit Aktivkohle weiter gereinigt werden kann, scheidet beim Eindampfen rohes N-Allyl- morphin aus.
Dieses kann durch Ausziehen mit einem wasserfreien Dialkyläther vor- gereinigt und dann zur weiteren Reinigung aus einem alkoholischen Lösungsmittel, etwa aus Methanol, umkristallisiert. werden.
<I>Beispiel:</I> 120<B>g</B> wasserfreies, analysenreines Nor- inorphin (0,44 Mol) wurden in 1500 cm.3 ab solutem Alkohol .stispen:
diert und der Suspen sion 53,5 g ALlylbromid (0,44 Mol) und 53,5 g Natriumbicarbonat (0,63 Mol)
zu- gesetzt. Die erhaltene Mischung wurde unter Rühren am R.ücldluss etwa 5 1Stunden erhitzt. Dann wurde die heisse Reaktions- mischung im Vakuum zur Trockne einge dampft und der Rüel-stand mit.
l-200 ein-' heissem Chloroform ausgezogen. Der heissen Chloroformlöstzng wurden etwa.<B>10</B> g Aktiv kohle zugesetzt, die Mischung gerührt und dann durch eine Schiehe Kieselgur filtriert. Das zurückbleibende unlösliche Material (Kohle, Kieselglzr und Salze) wurde mit je 75 eins siedendem Chloroform viermal aus gewaschen.
Das klare Filtrat tind die Wasch- flüssigkeit wurden zusammen im Vakuum eingedampft. Der Rückstand wurde liieratif mit 450 ein-' wasserfreiem Diät-hvläther zer rieben, bis er vollständig kristallin geworden war. Der kristalline Sehlamm wurde auf 2" (-' abgekühlt und etwa 15 Stunden bei dieser Temperatur gehalten.
Darre wurde abfiltriert, der kristalline R.üekstand dreimal mit je 50 ein- kaltem Diäthvläther (Temperatur etwa 0 C) gewaschen und da,s gewaschene Produkt an der Luft. getrocknet., wobei 117 g rohes N-Allyl-noimorphin mit einem Schmelz- punkt von 198 bis 201 C erhalten wurden.
Dieses zeigte mit reinem N-Allyl-noranorphin vermischt keine Sclimelzpunktsdepression. Die Ausbeute betrug 85,301o der Theorie.
Das so erhaltene Material wurde wie folgt gereinigt: 117 g rohes Produkt wurden in 1000 eins heissem Methanol gelöst, 5 em3 Ak tivkohle zugesetzt und die Miseliung gerührt,
dann heiss filtriert und der Kohlefilterkuchen mit 170 eins heissere Methanol aussgewasehen. Das Filtrat und die Waschflüssigkeit. wurden zusammen im Vakuum auf etwa 390 bis 400 em3 eingedampft.
Diese angereicherte Lösung wurde auf etwa 2 C abgekühlt und bei dieser Temperatur etwa 15 Stunden ste- bengelassen. Das ausgeschiedene, kristalline Material wurde nun. abfiltriert, dreimal mit Je 35 ein-' eiskaltem Methanol gewaschen und im Vakuum über konzentrierter Schwefel säure bis zur Cewich.tskottstaitz getrocknet. So wurden 97,
7 g praktisch reines N-Allyl-nor- morphin mit einem Schmelzpunkt 205-'207'(' erhalten, was einer Ausbeute beim Reini- gungsverfahren von etwa 83,50;
o entspricht. Weitere 8,3 g des Produktes wurden noeli aus der Mutterlösung gewonnen, so dass' sieh eine totale Ausbeute von<B>106</B> - N-Allyl-nor- morphin ergab, also etwa 90,6% der den i Reinigungsverfahren unterworfenen Menge.
