The N-oxy-methylamides of the general formula given above which are required from starting materials are easily accessible by allowing aqueous formaldehyde solutions to act on the corresponding α-alkoxy-fatty acid amides.
Some of the α-alkoxy-fatty acid amides which can be considered are known; those that have not yet become known are: can be produced by the methods described for the preparation of the known alkoxy fatty acid amides.
The present patent now relates to a process for the preparation of a derivative of 3- (4'-oxy-phenyl) -3- (4 "-oxy-3" - aminomethyl-phenyl) -oxindole. The process is characterized in that 3,3-bis- (4'-oxy-phenyl) -oxindole are condensed with N-oxymethyl-n-butoxyacetamide. The. Condensation is preferably accomplished by heating the components with zinc chloride in glacial acetic acid.
The new compound obtained, 3- (4'-Oxv-phenyl); -3 - (3 "-n-butoz-y-acetylaminomethyl-4" -oxy-phenyl) -oxindole, is a fine, amorphous one Powder before. It should be used as a medicinal product or as an intermediate product for medicinal products.
Example 1 26 parts of n-butoxyacetamide are suspended in a solution of 2 parts of potash in 20 parts by volume of water. A solution of 18 parts of 30% strength aqueous formaldehyde solution is added and the mixture is heated on the steam bath for half an hour.
The clear solution is then concentrated to a small volume in vacuo and extracted exhaustively with ether. The combined ethereal solutions are dried over potassium carbonate and evaporated. The N-oxymethyl-n-butoxyacetamide remains as a yellow oil that can be further processed directly. In the pure state, the compound melts at 46.
16 parts of 3,3-bis- (4'-oxy-phenyl) -oxindole are dissolved in a solution of 20 parts of anhydrous zinc chloride in 70 parts by volume of hot glacial acetic acid. The mixture is then cooled to about 10 and 18 parts of N-oxymethyl-n-butoxyacetamide are added, the temperature rising to 25 to 28, and the mixture is left to stand for 48 hours at room temperature. Then the yellow solution is allowed to flow in a thin stream with stirring on ice, whereupon the crude condensation product precipitates out as greasy.
It is dissolved in 5% sodium hydroxide solution in the cold, filtered through a little activated charcoal and then carefully acidified. The 3- (4'-oxy-phenyl) -3- (3 "-n-butoxyacetyl-aminomethyl-4" -oxy-phenyl) -oxindole precipitates out as a fine amorphous powder. It is suctioned off and dried at a temperature not exceeding 50 Tem.
The product produced in this way has a nitrogen content of 5.89%, the theoretical content is 6.10 / 0.
