verfahren zur Herstellung von 3-Acyloxy-bisnor-5-cholensäure. Naeh dem Verfahren der Patentschrift. I)RI' Nr.750212 gelangt man zu 3-Acyloxy- bisnor-5-eholensäure,
indem man Phytoester nach Absättigung der Kerndoppelbindung mit Brom der Ozonisierung unterwirft und die erhaltenen Ozonide unter gleichzeitiger Wie derherstellung der Kerndoppelbindung reduk tiv aufspaltet. Die Ausbeute an 3-Acyloxy- hisiior-5-cholensäure beträgt rund 28 %.
Es wurde nun gefunden, dass man die Aus beuten wesentlich steigern kann, wenn man an Stigmasterinester bzw. solche enthaltende Mischungen mit Sitosterinestern (Acylphyto- sterin) in einem organischen Lösungsmittel in Gegenwart einer organischen Base Brom an die Kerndoppelbindung anlagert, dann nach Pufferung mit einer organischen Säure Ozon im Überschuss durch die Reaktionslösung lei tet., die Reaktionslösung mit Wasser wäscht und nach Verdampfen des Lösungsmittels mit Chromsäure oxydiert,
dann mit Zinkstaub in Gegenwart einer organischen Säure reduziert.
Als organisches Lösungsmittel kann man zum Beispiel Chloroform, Tetrachlorkohlen- stoff oder Methylenchlorid verwenden. Die Anlagerung von Brom an die Kerndoppelbin- dung erfolgt zweckmässig in Gegenwart von Pyridin, Piperidin oder Trimethylamin. Als organische Säure verwendet. man zum Beispiel Eisessig, Propionsäure oder Ameisensäure.
Das Ozon -wird zum Beispiel in bis zu zehn fachem Lberschuss über die theoretisch erfor- derliche Menge durchgeleitet. Die Oxydation mit Chromsäure wird zweckmässigerweise bei Zimmertemperatur vorgenommen. Die an schliessende Reduktion mit Zinkstaub wird mit Vorteil ebenfalls bei Raumtemperatur eingeleitet. Bei dieser Reaktion steigt jedoch die Temperatur ausserordentlich rasch an und erreicht etwa 90 C. Der bei der Reduktion freiwerdende Wasserstoff spaltet die restlichen Ozonide und entfernt zugleich das Kernhalo gen.
Die entstandenen 3-Acyloxy-bisnor-5- cholensäure kann nach bekannten Methoden über das Natriumsalz isoliert und aufgearbei tet werden.
Das Verfahren liefert Ausbeuten um 60 %, wobei das aus dem Neutralanteil zurückgewon nene Stigmasterinacetat nicht in Anrechnung gesetzt ist.
In Anbetracht der Bedeutung der 3-Acet- oxy-bisnor-5-cholensäure als Zwischenprodukt für die Herstellung von Hormonen ist dieses Verfahren von grösstem Wert. Beispiel: Zu einer Lösung von 2,72 kg Phytosterin- acetat mit einem Gehalt von etwa 14 % Stig- masterinacetat (= 380 g Stigmasterinacetat)
in 7 1 trockenem Chloroform und 200 cm3 trockenem Pyridin lässt man unter Rühren bei 10 bis 20 C eine Brom-Chloroformlösung, bereitet. aus 306 cm3 (= 960 g) Brom und 5 1 Chloroform, innerhalb von 30 Minuten zufliessen. Darauf gibt man zur Reaktions- lösung 300 eins Eisessig, kühlt dabei auf 0 C und leitet einen 3,3prozentigen Ozonstrom 16 Stunden durch die gekühlte Lösung.
Die Sauerstoffstromgeschwindigheit beträgt 901 pro Stunde und die Gesamtozonmenge, die durch die Reaktionslösung geleitet wird, 68 g (theoretisch erforderlich: .10 g). Die Reak tionslösung wird danach mit 4 1 Wasser zwei Stunden gerührt, die Chloroformschicht ab getrennt Lind schliesslich im Vakuum einge engt.
Darauf nimmt. man den Rüekstand in 10 1 Eisessig auf und lässt zu dem Gemisch unter Rühren eine Chromsäurelösung, berei tet aus 160 g Cr03 und 400 cms H.0, inner halb einer Stunde zutropfen, wobei die Tem peratur 20 C nicht übersteigen soll.
Anschliessend wird die Reaktionslösung portionsweise mit 700 g Zinkstaub versetzt, wobei die Temperatur bis auf etwa 80 C an steigt. Die Reduktion dauert 30 bis -10 Mi nuten. Man giesst auf Eis, nimmt mit Äther auf und entzieht der ätherisehen Lösung die 3-Acetoxy-bisnor-5-cholensätire durch Schüt teln mit Natronlauge. Durch Kristallisation aus Äther und Toluol bzw. Benzol wird die Säure schliesslich rein erhalten. Der Schmelz- Punkt beträ-t \?31 bis n35 C. Die Ausbeute von 190 g entspricht. 58,5 Klo der Theorie.

The addition of bromine to the core double bond is expediently carried out in the presence of pyridine, piperidine or trimethylamine. Used as an organic acid. for example glacial acetic acid, propionic acid or formic acid.
The ozone, for example, is passed through in an excess of ten times the amount theoretically required. The oxidation with chromic acid is expediently carried out at room temperature. The subsequent reduction with zinc dust is also advantageously initiated at room temperature. During this reaction, however, the temperature rises extremely quickly and reaches around 90 C. The hydrogen released during the reduction splits the remaining ozonides and at the same time removes the nuclear halogen.
The resulting 3-acyloxy-bisnor-5-cholenic acid can be isolated and worked up using the sodium salt by known methods.
