Zwecks Herstellung von asymmetrischen Verbindungen kann man die Umsetzungen auch stufenweise vornehmen. Die Anlagerung der Halogenverbindungen an die tertiären Amine erfolgt beim Methyljodid schon bei Zimmertemperatur und unter Wärmeentwick lung, während in den meisten andern Fällen zur Erzielung völliger Umsetzung zweckmässig erwärmt wird.
Die Umsetzungen können in An- oder Abwesenheit von Lösungsmitteln durchgeführt werden; als solche kommen zum Beispiel Aceton, Butanon, Essigester, Benzol, Tohiol, Xylol, Äther, Dioxan, Methanol, abs. Äthanol oder auch ein Überschuss an Alkyl- halogenid in Frage.
Allgemein reagieren die Jodverbindungen am leichtesten, die Chlor verbindungen am schwersten. Quaternäre Am moniumjodide oder -bromide kann man auch mittels Silberchlorid in die quaterriären Am- monilimchloride überführen. Als besonders geeignete Alkylhalogenide seien Methyljodid, Methylbromid, Äthyljodid und Äthylbromid genannt. Nötigenfalls werden die Umsetzun gen in Druckgefässen ausgeführt.
Die als Ausgangsprodukte in Betracht fal lenden di-tertiären Bis-aminoalkoxy-alkane können beispielsweise durch Umsetzung von a,oo-Alkandiolen der allgemeinen Formel HO-(CH2)ri OH vorteilhaft in Form ihrer Metallverbindungen (Alkoholate) mit 2 Mol von tertiären Amino- alkylhälogeniden der allgemeinen Formel

Ihre Alkoholate können zum Bei spiel mit ss-Chlor-äthyl-y-Brom-propyl- oder b-Brom-butyl-dimethylamin, -methyl-äthyl- amin, -methyl-propylamin, -methyl-butylamin, -methylisobutylamin, -diäthylamin, -äthylpro- pyl-amin, -äthyl-butyl-amin, -äthyl-isobutyl- amin, -dipropylamin, -propyl-butylamin,
-pro- pyl-isobutylamin, -dibiitylamin, -butyl-isobu- tyl-amin, -diisobutylamin, -methyl-isopropyl- amin, -methyl-sec.biityl-amin;
N- (ss-Chlor- äthyl)-, N-(y-Brom-propyl)- oder N-(d-Brom- butyl) -pyrrolidin, -piperidin, -2-methyl-piperi- din, -morpholin umgesetzt werden.
Man kann die di-tertiären Bis-amino- alkoxy-alkane aber auch durch Tetraalkylie- rung von di-primären Diaminen herstellen und diese anschliessend oder gegebenenfalls im gleichen Arbeitsgang in die quaternären Ammoniumverbindungen überführen. Dieses Verfahren kommt besonders dann in Frage, wenn. die di-primären Diamine leicht zugäng lich sind, wie z.
B. die durch Anlagerung von einem Mol eines a,( -Diols an zwei Mol Acrylo- nitril und anschliessende Reduktion gemäss DRP Nr. 734475 erhältlichen Bis-(y-amino- propoxy)=alkane.

dem 1,2-Bis-(b-brom-biitoxy)-äthan, kann man z. B. durch Umsetzling von entsprechenden Bis-(co-oxy-alkoxy)-alkanen mit Phosphor- halogeniden nach Ziegler und Hall, Ann. 528, 152 (1937), wo auch ein Verfahren für die Herstellung von Bis-(o)-oxy-alkoxy)-allianen angegeben ist, gelangen.
Die so erhältlichen neuen quaternären Am moniumverbindungen werden in reiner Form als farblose, kristallisierte Produkte erhalten, die in Wasser mit fast neutraler Reaktion löslich sind. Sie sollen als Pharmazeutika, z. B. zur Beeinflussung des Muskeltonils, Verwen dung finden.
Gegenstand vorliegenden Patentes ist nun ein Verfahren zur Herstellung einer quater- nären Ammoniumverbindung eines Bis-amino- alkoxy-alkanes. Das Verfahren ist dadurch ge kennzeichnet, dass man auf 1 Mol 1,10-Bis- (ss-dimethylamino-äthoxy)-dekan 2 Mal Bu- tylbromid einwirken lässt.
Die erhaltene neue Verbindung, das 1,10- Bis - [ss - (dimethyl-biityl-ammoniiim)- äthoxy] - dekan-dibromid, schmilzt bei 148 bis 149 . Sie soll therapeutische Verwendung finden. <I>Beispiel:</I> 7.7 Teile 1,10-Dekan-diol, 25 Teile Dimethyl- amino-äthylchlorid und 87 Teile abs. Toluol werden gemischt und auf 90 bis 100 erhitzt.
Bei dieser Temperatur gibt man portionen- weise 8;2 Teile Natriumamid, welche zusam men mit abs. Toluol fein gemahlen wurden, zu und rührt weitere 2 Stunden bei der glei chen Temperatur. Anschliessend erhitzt man etwa 2 Stunden zum Sieden -unter Rückfluss. Nach Erkalten wird die gebildete Base durch Verrühren mit überschüssiger 9,n-Salzsäure in salzsaure Lösung übergeführt, diese mit Äther gewaschen, mit Tierkohle abgesaugt und unter Eiskühlung mit überschüssiger 301/aiger Natronlauge versetzt.
Die Base wird ausgeäthert und, nach Trocknen der Äther lösung mit Kaliumhydroxyd, durch fraktio nierte Destillation gereinigt.
Man erhält so 1,10-Bis (l-dimethylamino- äthoxy)-dekan als farbloses Öl vom Siede punkt 134 bis 137 - unter 0,04 mm Druck.
13 Teile dieser Base werden mit 80 Teilen trockenem Aceton und 17 Teilen Butylbromid 14 Stunden in einem Wasserbad von 50 bis 60 erwärmt. Nach Erkalten wird abgesaugt und das erhaltene 1,10-Bis-[ss-(dimethyl-butyl- ammonium) - äthoxy] -.dekan - dibromid ge- wünschtenfalls durch Umkristallisieren aus Butanon -I- wenig abs. Methanol gereinigt,
indem das Salz in möglichst wenig heissem Methanol gelöst und darauf so viel heisses Butanon zugefügt wird, dass in der Hitze noch alles gelöst bleibt. Das umkristallisierte Pro dukt schmilzt bei 148 bis 149 .
