Die Verbindung weist kreislauffördernde Eigenschaften auf.
Es ist bereits bekannt, dass man aus einem primären Amin und einem Aldehyd oder Keton durch katalytische Hydrierung der bei den Komponenten in geeigneten Lösungsmit teln sekundäre Amine darstellen kann. Die Reaktion verläuft so, dass die Aminogruppe des primären Amins sich an die Carbonyl- gruppe des Aldehyds oder Ketons zum Alde- hyd-Ammoniak oder unter Wasserabspaltung zur Sehiffschen Base anlagert, Verbindungen,
zur Herstellung eines Raeemates von 1.-(p- Oxy - phenyl) - 2 - (a- methyl - y - phenyl - propyl- amino)-propanol-(1.) der Formel die dann durch Aufnahme von 1 111o1 Wasser stoff in die sekundären Amine übergehen.
Auf die gemeinsame Hydrierung eines a Aminoket.ons mit einem Keton schien diese Reaktion nicht übertragbar zu sein, da zu er warten war, dass sich das a-Aniinoketon unter den Bedingungen, unter denen es mit dem zu gesetzten Keton reagieren soll, mit. sich selbst unter doppelter innerer Aldehyd-Ammoniak- Bildung und Wasserabspaltung zu einem Di- liydropyrazin vereinigen würde, das seiner seits durch Verlust zweier Wasserstoffatome in ein stabiles Pyrazin übergehen würde.
Es ist daher durchaus überraschend, dass es in dem speziellen Fall des 1-(p-Oxy-phenyl)-2- amino-propanons-(1) möglich ist, durch ge meinsame katalytische Hydrierung mit. Ben- zylaceton in sehr guter Ausbeute zu Verbin dungen der Formel 1 zu gelangen. Die Reaktion verläuft nach der Gleichung

Das Aminoketon kann auf übliche Weise, z. B. durch Umsetzen des p-Acetoxy-a-brom- propiophenons mit wässrigem Ammoniak, dar gestellt wenden.
Das Benzylaceton wird zweckmässig in äquimolekularerMenge oder auch in geringem L?bersehuss gegenüber dem Aminoketon ver wendet und das Gemisch unter Wasserstoff in Gegenwart eines Hydrierungskatalysators bei Atmosphärendruck oder auch bei erhöhtem Druck geschüttelt.
Als Lösungsmittel wählt man zweckmässig solche, die ein gewisses Lösungsvermögen für Wasser besitzen und keine Carbonylgruppe enthalten. Es ist besonders vorteilhaft, niedere Alkohole zu verwenden. Vorzugsweise ver wendet man Methanol; man kann aber auch Ethyl- oder Isopropylalkohol verwenden. Diese Alkohole können in wasserfreiem Zustand verwendet werden; es können aber auch was serhaltige Alkohole zur Anwendung gelangen, z. B. 80 o/oiges Methanol.
Als Hydrierungskatalysatoren sind Raney- Nickel oder Edelmetallkatalysatoren, wie Palladiumhydroxyd auf Bariumsulfat oder Platinmohr, vorzugsweise aber Platinoxyd, ge eignet. Bei der Hydrierung werden 2 Mol Wasserstoff aufgenommen. Das Ausgangs Aminoketon geht in Lösung und das Endpro dukt entsteht in guter Ausbeute. Es soll als Mittel zur Kreislaufförderung Verwendung finden.
Die Aufarbeitung der Reaktionslösung kann in üblicher Weise erfolgen. Da das End produkt 3 asymmetrische, ungleichwertige Kohlenstoffatome enthält, wäre an sich die Bildung von 4 stereoisomeren Racematen mög lich.
Von diesen Racematen entstehen jedoch, wenn man in der angegebenen Weise arbeitet, stets nur zwei, von denen das eine bei<B>111.</B> bis 112 C schmilzt und ein Chlorhydrat mit dem Smp. 222 bis 22.1 C, ein Acetat mit dem Smp. 184 bis 189 C und ein Propionat mit dem Smp. 175 bis 177 C bildet und starke kreislauffördernde Eigenschaften hat, wäh rend das andere bei 139 bis 1.10 C schmilzt und ein Chlorhydrat mit dem Smp. 223 bis 224 C, ein Bromhydrat mit dem Smp. 215 bis 216 C,
ein Acetat mit dem Smp. 195 bis 199 C und ein Propionat mit dem Smp. 187 bis 188 C bildet und wesentlich schwäehere kreis lauffördernde Eigenschaften aufweist.
Zur Trennung kann (las Racematgemisch in einem niederen Alkohol, insbesondere in Methanol, gelöst. und die Kristallisation einge leitet werden; hierbei fällt zuerst die Base mit dem Smp. 111 bis 112 C aus, welche ab getrennt wird.
<I>Beispiel:</I> 3<B><I>30,0</I></B> g 1-(p-Oxy-pheny1)-2-amino-propanon- (1) vom Smp. 168 bis 170 C (mit. 2 Mol Kri stallwasser) werden in 130 cm3 reinem Metha nol suspendiert und das Ganze nach Zusatz von 27,0g Benzylaceton und 0,5g Platinoxyd unter Wasserstoff geschüttelt. Nach 6 bis 8 Stunden sind etwas mehr als 2 Mol Wasserstoff aufgenommen, die Hydrierung kommt. praktisch zum Stillstand. Man arbeitet in üblicher Weise auf und erhält z.
B. durch Eindampfen im Vakuum nach dem Ansäuern mit Salzsäure und Auskochen des Rückstandes mit 70 cm3 Aceton 801/o der Theorie eines Chlorhydrat- Gemisches, das etwa. gleiche Teile der bei 111 bis 112 und der bei 139 bis 140 C schmelzen den Base enthält.
An Stelle von Methanol kann man mit demselben Ergebnis Äthanol als Lösungsmittel verwenden, wobei die Hydrierung nur etwas länger dauert. An Stelle von 0,5 g Platinoxyd kann man aticli 1.0 g eines 1.0 o/oigen Palla- diumhydroxyd-Bariumsulfat=Katalysators oder 25 g Raney-Niekel verwenden und zur Be schleunigung der Wasserstoffaufnahme in diesen Fällen unter Überdruck hydrieren.
Zur Basentrennung werden 20,0 g des Ge misches der Basen in 20 ml Methanol heiss gelöst und die Lösung nach dem Abkühlen mit der Base vom Smp. 111 bis 112 C ge impft. Nach 3stündigem Stehen bei Zimmer- temperatur sind 9,2 g der Base (bei 70 C getrocknet, wobei 1 Mol Kristallmethanol ent weicht) vom Smp. 108 bis 111 C auskristalli siert. Das Produkt kann durch Umkristallisie- ren weiter gereinigt werden.
