Verfahren zur Herstellung eines Morphin-6.glacosides. Es wurde gefunden, dass Morphin durch Verknüpfung des in 6-Stellung befindliehen alkoliolisehen Sauerstoffatoms mit einem Zuk- kerrest in Verbindungen übergeführt werden kann, deren therapeutische Breite im Ver gleich zum 311orphin erweitert ist; insbeson dere ist der Einfluss dieser Verbindungen auf das Atemzentrum geringer als beim Morphin selbst.
Die Herstellung dieser VerbindLingen er folgt zweckmässigerweise derart, dass man in 3-Stellung aeviiertes Morphin mit einer Aeeto- hafogenose in Gegenwart halogenwasserstoff- bindender Mittel umsetzt und das Reaktions produkt, verseift. Man erhält iNIorphin-6-g#iieo- side der allgemeinen Formel

Man kann zum Beispiel in der Weise ver fahren, dass zunächst die in 3-Stellung befind- lielie phenolisehe Hydroxylgruppe des Mor phins durch Einwirkung eines Säurehalo- M genids, wie z.
B. Aeetylehlorid, Benzoylehlorid oder Cliloi#aiiieisen,#"äiireester aeviiert wird und dass dann die in 6-Stellung befindliche alko- holisehe Hydroxylgruppe durch Einwirkung einer Aeetohalogenose, z.
B. AcetobromgILieose, in Gegenwart eines halogenwasserstoffbinden- den Mittels, wie Silberoxyd oder Silberearbo- nat, veräthert wird. An Stelle von Aeetobrom- glucose können auch entsprechende Verbin dungen von andern Hexosen, wie Mannose oder Galactose, oder auch von geeigneten Pent- osen, verwendet werden.
Aus den so erhaltenen Reaktionsprodukten gewinnt man durch Ver- seifung die 3#lorphin-6-glueoside der obigen allgemeinen Formel. Durch Salzsäure können diese Morphin-6-glucoside in die Hydroehlo- ride übergeführt werden.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines Morphin- 6-glucosides, welches dadurch gekennzeichnet ist, dass man in 3-Stellung aeyliertes Morphin mit Acetohalogenglueose in Gegenwart eines halogenwasserstoffbindenden Mittels umsetzt und das entstandene Aeylmorphintetraacet.v1- glucosid durch Verseifung in das Morphin-6-fl- zn d-glucosid überführt.

20 Teile Benzoylmorphin-tetraaeetylgluco- sid werden mit 348 Teilen 0,4-ii-Barium- hydroxyd Lösung und<B>320</B> Teilen 'Methanol 2 Stunden zum Sieden erhitzt und dann mit <B>70</B> Teilen 2-n-Sehweielsäure versetzt. Man ven- trifugiert und dampft die klare Lösung im Vakuum bis züm Sirup ein.
Der Rückstand wird in 1.30 Teilen Aceton gelöst und mit kon zentrierter Salzsäure anoesäuert. Die ausge schiedenen Kristalle werden mit verdünnter Salzsäure und mit Aeeton gewaschen. Mai) erhält 12,2 Teile Morphin-6-fl-d--##l-uieosi(1-liv- droehlori(l. Farblose Nadeln vom Fp. '2361'.
