Verfahren zur Herstellung von Bakterienpräparaten, insbesondere von milchsäureerzeugenden Bakterien.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Bakterienprä- paraten, z. B. in grossem Massstab, insbesondere von Bakterien, die Milchsäure als haupt- sächliches Stoffwechselprodukt erzeugen, wie z. B. Bakterien der Gruppen Bacterium cueu mens fermentati, Bacterium brassicae, Stre ptobaeterium plantarum, Bacterium acetylcholini usw.
Diese Bakterien haben die Fähig- keit, Milchsäure ausserordentlich rasch und in grossen Mengen zu bilden, weswegen man sie für verschiedene Zweeke anwenden kann, z. B. zum Ensilieren von Fntter vnd Konser- z von insbesondere vegetabilischen Lebensmitteln. Die genannten Bakterien liefern als Stoffweehselprodukte auch mehr oder weniger grosse Mengen Acetyleholin, welches vom medizinischen Gesiehtsplmkt aus wertvoll ist, da es eine ausserordentlich kräftige Wirkung auf den Gefäss-und Nluskeltonus hat, so dass es in starker Verdünnung als sehr wirksames Mittel gegen Verstopfung verwendet werden kann.
Das Verfahren gemäss der Erfindung zur Herstellung von Bakterienpräparaten, insbesondere von milchsäureerzeugenden Bakterien, bei welchem in einem mit einem saureneutralisierenden Mittel versetzten Nähr- substrat säurebildende Bakterien unter Umrühren gezüehtet werden, ist dadurch gekennzeichnet, dass ein Nährsubstrat verwendet wird, welches organische Partikeln enthält, an denen sich die Bakterien festsetzen, und die Kultur zum Schlusse einige Zeit stehengelassen und dann der dabei gebildete Bodensatz, bestehend aus den mit Bakterien behafteten organischen Partikeln, abgezogen wird und eventuell zu einer Paste oder.einem trockenen Pulver verarbeitet wird.
Das Nährsubstrat kann z. B. aus einer Maische von feinvermahlenem Gerstenmalz bestehen. Es hat sich gezeigt, dass der aus einem solchen Nährsubstrat beim Absetzen gebildete Bodensatz eine beträchtlich grössere Menge Bakterien enthält als die gesamte Kulturflüs- sigkeit. Dies beruht offenbar darauf, dass die in grosser Anzahl vorhandenen, äusserst klei- nen Schalenteilehen (Kleieteilchen) in hohem Masse mit Bakterien behaftet sind und beim Sinken diese und wahrscheinlich auch in der ..SubstratflüssigkeitfreischwebendeBakterien mit sich führen.
Um zu verhindern, dass die durch die Wir kung der Bakterien gebildete Säure, z. B.
Milchsäure, eine zu stark saure Reaktion des Substrates hervorruft, was ein für das Bakterienwachstum ungeeignetes Milieu bedeuten würde, wird ein säureneutralisierendes Mittel, z. B. Kreide oder Kalziumkarbonat, vorzugsweise in einer derartigen Menge zugesetzt, dass die gebildete Säure grösstenteils neutralisiert wird. In dieser Weise wird ein schädlicher Einfluss der Säure auf die Bakterien verhindert.
Die Nährsubstratflüssigkeit wird vorteilhaft, nachdem der aus den mit Bakterien be- hafteten organischen Partikeln bestehende Bodensatz abgetrennt ist, zentrifugiert sowie im Vakuum eingedunstet und sodann, eventuell mit dem Bodensatz, z. B. zu Tabletten, Pillen, Granulat oder dergleichen verarbeitet.
Es können so z. B. als Abführmittel dienende Präparate erhalten werden, welche man dureh Zusätze anderer Stoffe oder Substanzen noch variieren kann.
Ausfiihrungsbeispiele : 1. 42 kg sehr fein vermahlenem Gerstenmalz werden 258 kg Wasser mit einer Temperatur von 65 C zugesetzt. Die Temperatur der Maische soll ungefähr 600 C betragen.
Man lässt etwa eine Stunde verzuckern. Darauf werden 7,5 kg Kalziumkarbonat zugesetzt. Dann wird die Maische sterilisiert, auf 3200 abgekühlt und mit/4 to Stamm- kultur säurebildender Bakterien, auf den ganzen Satz berechnet, geimpft, worauf das Ganze ohne mit der Aussenluft in Berührung zu kommen einem besonderen Apparat zuge- führt wird, in dem eine gründliehe und dauerhafte Durchmischung der Maische erfolgt. Man züchtet nun die Bakterien etwa 4 Tage, bis man einen pu-vert von etwa 4,5 erhält. Darauf lässt man den Apparat stillstehen und die Maische sich absetzen.
Nach ungefähr 20 Stunden zieht man den Bodensatz ab, der ungefähr 95 kg beträgt, zentri fugiert die verbleibende Kulturflüssigkeit und verarbeitet den Bodensatz, das Bakteriensediment von der Zentrifugierung und steriles Kartoffelmehl zu einer homogenen Paste oder einem Pulver. Auf 95 kg Bodensatz verwendet man 125 kg Kartoffelmehl und 4, 5 kg Bakteriensediment. Das fertige Präparat wird luftdicht verpackt und zweck- mässig bei 0 C, doch nicht unter-2 C verwahrt.
Die beim Zentrifugieren der Klllturflüs- sigkeit erhaltene Ijöslmg wird im Vacuum eingedilnstet. Die so erhaltene Trockensubstanz aus Stoffwechselprodukten der Bakterien ergibt ein ausgezeichnetes Mittel gegen Verstopfung. Da die Dosierung sonst ziem- lich hoch gehalten werden müsste, etwa 7 bis 8 Halbgrammtabletten je Tag, werden zweckmässig zur Erhöhung der Wirkung noeh andere Substanzen zugesetzt.
Für andere medizinische Zwecke, z. B. bei gewissen Darmerkrankungen, hat es sich als vorteilhaft erwiesen, dem Verdauungssystem (Darmsystem) ein Präparat zuzuführen, wel ches die oben genannten trockenen Stoff wechselprodukte und erfindungsgemäss erhaltene Bakteriensedimente enthält. Gegebenen- falls wird solchen Präparaten zweckmässig ein Schutzstoff zwecks Verhinderung seiner Zersetzung, bevor es seinen Bestimmungsort erreicht, z. B. infolge der Einwirkung des sauren Magensaftes, zugegeben.
Bei der Herstellung von Acidophilus-oder Joghurt-Bakterienpräparaten hat man bisher stets Gewicht darauf gelegt, die von den Stoffwechselprodukten befreiten Bakterien in trockener Form in trockenen Mitteln einzubetten, meist in Milchzucker oder in Emul- sionen, um sie darauf in Form von Tabletten oder Kapseln in das erkrankte Verdauungssystem einzuführen, wo sie infolge ihrer spe zifischen Züehtungsfähigkeit die Erzeugung der für die Heilung erwünschten Stoffweeh- selprodukte einleiten sollen.
Dabei hat man bisher gänzlich ausser acht gelassen, dass die zugeführten Bakterien in dem kranken Verdauungssystem oft Milieuverhältnisse antreffen, die weder für das Wachstum der Bakterien noch für die Bildung von gewünschten Stoffweehselprodukten geeignet sind ; die therapeutisehe Wirkung entsprach wahrsehein- lich deshalb vielfaeh nicht den Erwartungen.
Untersucht man die vorhandene Literatur über die therapeutische Wirkung der Acido philnsmilch (Reformjoghurt und Joghurt), so erhält man den Eindruck, dass die in gro ssen Mengen (bis zu einem Liter pro Tag) eingenommene Bakterienmilch ihren Einfluss auf das Verdauungssystem, insbesondere das Darmsystem, in erster Linie durch die Wirkung der Stoffwechselprodukte der Bakterien ausübt. Die Stoffwechselprodukte stellen also offenbar die primär erwünschten Sub- stanzen im Darmsystem dar, während das Vorhandensein der Bakterien erst sekundär von Bedeutung ist. Der Bakterienwaehstumsprozess und damit auch die Erzeugung der Stoffwechselprodukte setzt erst ein, wenn die kranke Bakterienflora, z.
B. die Entwicklung der Fäulnisbakterien, abzunehmen beginnt, wobei bessere Milieuverhältnisse und Lebensbedingungen für die zugesetzte Bakterienflora auftreten. Eine Zufuhr von Stoffwech- selprodukten von aussen her wirkt also verbessernd auf das Milieu, so dass sich die zugesetzten Bakterien besser entwiekeln und damit weitere Mengen Stoffwechselprodukte beitragen.
Nach der vorliegenden Erfindung kann ein für den oben angegebenen Zweck besonders geeignetes Präparat hergestellt werden, das sowohl sämtliche Stoffwechselprodukte, beispielsweise von Milchsäurebakterien, wie aueh die Bakterien selbst enthält. Gegebenen- falls können auch Nährsubstanzen für die Bakterien zugesetzt werden. Ferner kann man einen säureneutralisierenden Stoff beigeben, der im wesentlichen als Schutz beim Durch- gang des Präparates durch den Magensack dienen soll.
2. Die Herstellung eines solchen Präpa- rates erfolgt zweckmässig in folgender Weise :
Als Nährsubstrat verwendet man z. B. mole, die nach dem Klären mit Laktose, Dextrose, Hefebouillon und Kalziumkarbonat versetzt wird. Nach dreimaligem Sterilisieren bei 100 C in besonderen Gefässen wird die Molkennährlosung mit 0,25 /o einer zwei Tage alten Acidophilus-Stammkultur (bestehend aus versehiedenen Acidophilusstämmen) geimpft und ohne Luftzutritt in einen Züchtungsapparat gebracht.
Nach Entfernung des Caseins enthält die Molke u. a. noch Albumin und Globulin, welche beim Sterilisieren koagulieren und sogenanntes Eiweissgerinnsel bilden, welches als organische Partikeln, an welchen sich die Bakterien festsetzen, wirkt.
Die Bakterien werden nun in der Nährlösung 4 bis 5 Tage bei einer Temperatur von etwa 38 C unter ständigem Umrühren gezüchtet.
Danach soll die zugesetzte Kalziumkarbonatmenge verbraucht sein, und die Lösung einen pH-Wert von 4,5 aufweisen. Die Kul- tur wird dann einige Zeit stehengelassen, und das Bakteriensediment wird abgezogen und im Kühlsehrank verwahrt. Die abgetrennte Plüssigkeit wird im Vakuum auf etwa 1/7 eingedlmstet. Die dabei erhaltene, etwas sirupartige Flüssigkeit wird darauf mit 5 /o (bezogen auf die Ausgangsflüssig- keit) feinvermahlenem, sterilem und leichtgeröstetem Zwiebackmehl versetzt. Das Ganze lässt man in Schalen erstarren ; darauf wird es zerkleinert, in Trockenschränken fertig- getrocknet sowie granuliert.
Das Bakteriensediment wird mit 5 Teilen sterilem Stärke- mehl und einem Teil Wasser zu einer homogenen Masse geknetet, die dann mit dem mit Kalziumkarbonat gemisehten G-ranulat der Stoffwechselprodukte gemischt wird. Die Masse wird durch ein Sieb geleitet und unter Zusatz von Laktose und Dextrose in gut- geschlossene Verpackungen gefüllt.
Soll das Acidophiluspräparat z. B. mit artspezifisehen Colistämmen kombiniert werden, so erfolgt dies zweckmässig im Ver hältnis 5 : 1. Für das Colinährsubstrat kann ebenfalls Molke verwendet werden, wie für die Acidophiluszüchtung. Man gibt jedoch zweckmässig etwas Fleisch-Peptonbouillon zu, da die Colibakterien in Gegenwart einer animalischen Substanz besser wvachsen. Diese Nährlösung, die zweckmässig einen pR-Wert von 7,0 hat, wird eine halbe Stunde bei 110 C sterilisiert und nach Abkühlung mit einigen physiologisch gleichartigen, artspezifischen Colistämmen geimpft und dann auf 36 C gehalten.
Nach 24,48 und 72 Stunden wird mit einer 40 /o igen Natriumkarbonatlösung neutralisiert, wozu zweckmässig 75 em3 auf 20 Liter Colikultur verwendet werden.
Die Züchtung ist nach 8 Tagen abgeschlossen, wobei die Kultur einen pu-vert von etwa 5, 2 aufweist. Darauf wird die Kultur einige Zeit stehengelassen und das Bakteriensediment abgezogen. Die abgetrennte Nährsubstratflüssigkeit wird mit der von der Acidophiluszüchtung erhaltenen eingedun stet. Zur Gewinnung des Endproduktes miseht man Aeidophilus-lmd Colisediment, neutralisiert aber das Gemisch so, dass es einen pH-Wert von 6,5 erhält. Danach vereinigt man das erhaltene Acidophilus-Coli-Konzen- trat mit den getrockneten Stoffwechsel- produkten.

The method according to the invention for the production of bacterial preparations, in particular lactic acid-producing bacteria, in which acid-forming bacteria are grown with stirring in a nutrient substrate mixed with an acid-neutralizing agent, is characterized in that a nutrient substrate is used which contains organic particles where the bacteria settle, and finally the culture is left to stand for some time and then the sediment formed, consisting of the organic particles contaminated with bacteria, is removed and possibly processed into a paste or a dry powder.
The nutrient substrate can, for. B. consist of a mash of finely ground barley malt. It has been shown that the sediment formed from such a nutrient substrate on settling contains a considerably larger amount of bacteria than the entire culture liquid. This is evidently based on the fact that the extremely small shell parts present in large numbers (bran particles) are contaminated to a high degree with bacteria and, when they sink, carry them with them and probably also bacteria floating freely in the substrate liquid.
To prevent the acid formed by the action of bacteria, e.g. B.
Lactic acid, which causes an excessively acidic reaction of the substrate, which would mean an unsuitable environment for bacterial growth, is an acid-neutralizing agent, e.g. B. chalk or calcium carbonate, preferably added in such an amount that the acid formed is largely neutralized. In this way a harmful influence of the acid on the bacteria is prevented.
The nutrient substrate liquid is advantageous after the sediment consisting of the organic particles contaminated with bacteria has been separated, centrifuged and evaporated in a vacuum and then, possibly with the sediment, e.g. B. processed into tablets, pills, granules or the like.
It can be B. preparations serving as laxatives can be obtained, which can be varied by adding other substances or substances.

The Ijöslmg obtained by centrifuging the cooling liquid is diluted in a vacuum. The dry substance obtained in this way from metabolic products of the bacteria is an excellent remedy for constipation. Since the dosage would otherwise have to be kept fairly high, around 7 to 8 half-gram tablets per day, other substances are expediently added to increase the effect.
For other medical purposes, e.g. B. in certain intestinal diseases, it has proven advantageous to supply the digestive system (intestinal system) with a preparation wel Ches contains the above-mentioned dry metabolic products and bacterial sediments obtained according to the invention. If necessary, such preparations are expediently given a protective substance to prevent their decomposition before it reaches its destination, e.g. B. as a result of the action of the acidic gastric juice added.
In the manufacture of acidophilus or yoghurt bacterial preparations, emphasis has always been placed on embedding the bacteria freed from metabolic products in dry form in dry agents, usually in lactose or in emulsions, and then in the form of tablets or capsules to be introduced into the diseased digestive system, where they are supposed to initiate the production of the metabolic products required for healing as a result of their specific tractability.
It has hitherto been completely disregarded that the introduced bacteria often encounter environmental conditions in the diseased digestive system which are neither suitable for the growth of the bacteria nor for the formation of the desired metabolic products; Therefore the therapeutic effect probably did not correspond to the expectations.
If one examines the existing literature on the therapeutic effects of acidic milk (reform yoghurt and yoghurt), one gets the impression that the bacterial milk consumed in large quantities (up to one liter per day) has an effect on the digestive system, especially the intestinal system, primarily through the action of the metabolic products the bacteria exerts. The metabolic products obviously represent the primarily desired substances in the intestinal system, while the presence of bacteria is only of secondary importance. The bacterial growth process and thus also the production of metabolic products does not begin until the diseased bacterial flora, e.g.
B. the development of putrefactive bacteria begins to decrease, with better environmental and living conditions for the added bacterial flora. A supply of metabolic products from the outside has an improving effect on the milieu, so that the added bacteria develop better and thus contribute additional quantities of metabolic products.
According to the present invention, a preparation which is particularly suitable for the purpose indicated above can be produced which contains all metabolic products, for example of lactic acid bacteria, as well as the bacteria themselves. If necessary, nutrient substances for the bacteria can also be added. An acid-neutralizing substance can also be added, which is essentially intended to serve as protection when the preparation passes through the gastric sac.
2. The production of such a preparation takes place appropriately in the following way:
As a nutrient substrate you use z. B. mole, which is mixed with lactose, dextrose, yeast bouillon and calcium carbonate after clarification. After being sterilized three times at 100 C in special vessels, the whey nutrient solution is inoculated with 0.25 / o of a two-day-old acidophilus stock culture (consisting of different acidophilus strains) and placed in a breeding apparatus without access to air.
After removing the casein, the whey contains u. a. Albumin and globulin, which coagulate during sterilization and form so-called protein clots, which act as organic particles on which the bacteria attach themselves.
The bacteria are now grown in the nutrient solution for 4 to 5 days at a temperature of about 38 C with constant stirring.
After that, the amount of calcium carbonate added should be used up and the solution should have a pH of 4.5. The culture is then left to stand for some time, and the bacterial sediment is drawn off and kept in the cooler. The separated liquid is reduced to about 1/7 in a vacuum. The somewhat syrupy liquid obtained in this way is then mixed with 5 / o (based on the starting liquid) finely ground, sterile and lightly roasted rusk flour. The whole thing is left to solidify in shells; it is then chopped up, dried in drying cabinets and granulated.
The bacterial sediment is kneaded with 5 parts of sterile starch flour and one part of water to form a homogeneous mass, which is then mixed with the granulate of the metabolic products mixed with calcium carbonate. The mass is passed through a sieve and filled into tightly closed packaging with the addition of lactose and dextrose.
If the acidophilus preparation z. B. are combined with species-specific coli strains, this is conveniently done in a ratio of 5: 1. Whey can also be used for the coli nutrient substrate, as for acidophilus breeding. However, it is advisable to add some meat-peptone broth, since the coli bacteria grow better in the presence of an animal substance. This nutrient solution, which suitably has a pR value of 7.0, is sterilized for half an hour at 110 ° C and, after cooling, inoculated with some physiologically similar, species-specific coli strains and then kept at 36 ° C.
After 24, 48 and 72 hours, it is neutralized with a 40% strength sodium carbonate solution, for which purpose 75 cubic meters per 20 liters of coliculture are used.
Cultivation is completed after 8 days, the culture having a pu-vert of about 5.2. The culture is then left to stand for a while and the bacterial sediment is removed. The separated nutrient substrate liquid is concentrated with that obtained from acidophilus cultivation. To obtain the end product, Aeidophilus lmd Colisediment is measured, but the mixture is neutralized so that it has a pH of 6.5. The acidophilus coli concentrate obtained is then combined with the dried metabolic products.
