Verfahren zur Herstellung einer Verbindung mit den Wirkungen der Pantothensäur e. Gegenstand vorliegender Erfindung ist ein Verfahren zur Herstellung einer Verbin dung mit den Wirkungen der Pantothensäure.
Die Pantothensäure ist eine in Naturstof fen (Nahrungsmitteln) wehverbreitete Sub stanz, die sich am besten durch ihre WaclLs- tumswirkungen bei niederen Organismen nachweisen lässt, die aber auch für die Er nährung der höheren Tiere wertvoll ist. Man kann die natürliche Pantothensäure hydroly tisch in zwei Bestandteile aufspalten und diese Bestandteile, von denen eines ss-Alanin ist, auf chemischem Wege wieder zur voll wirksamen Pantothensäure vereinigen.
Man nimmt an, dass hierbei eine peptidartige Ver knüpfung des ss-Alanins mit einer Diogy- valeriansäure eintritt. Man müsste also Panto- thensäure synthetisieren können, wenn man von den vielen Möglichkeiten diejenige Di- oxyvaleriansäure kennt, die ein Bestandteil der natürlichen Pantothensäure ist.
Im Gegensatz zu dieser Anschauung, zu der die Forschungen auf dem vorliegenden Gebiet bisher gekommen sind, wurde gefun den, dass man eine Verbindung herstellen kann, die sich in physiologischer, chemischer und physikalischer Hinsicht wie Pantothen- säure verhält, wenn man synthetisch gewon nenes, linksdrehendes a-Oxy-ss,ss-dimethyl-y- butyrolakton (also das Lakton einer Dioxy- kapronsäure)
mit ss-Alanin unter solchen Be dingungen kondensiert, dass eine peptidartige Verknüpfung (Bildung einer Säureamid- gruppierung) eintritt. Die Arbeitsweisen, die für diese Reaktion benutzt werden können, sind aus der Chemie der Herstellung der Peptide bekannt.
Das u-Oxy-ss,ss-dimethyl-butyrolakton ist bekannt (E. Glaser, Monatshefte für Chemie, Band 39, 1918, Seite 293). Zur Kondensation kann man es in der Racemform benutzen, oder als 1-Form, die man durch die übliche Spal tung mit Hilfe optisch-aktiver Basen (z. B. Chinin) leicht erhält. Das ss-Alanin kann für die Kondensation als solches oder als geeigne tes Derivat, z. B. als Ester oder als Salz, an- gewendet werden.
Aus dem linksdrehenden Lakton und ss-Alanin bildet sich nach der Erfindung eine rechtsdrehende Verbindung folo:ender Konstitution:

<I>Beispiele:</I> 1. ss-Alanin wird durch Erhitzen mit der 5fachen Menge 10%iger methanolischer Salzsäure in den Methylester übergeführt. 400 mg a-Oxy-f,ss-dimethyl-butyrolakto-n werden mit ungefähr 900 mg ss-Alanin- methylester gemischt und während 2 Stun den auf 70 erhitzt.
Das Reaktionsgemisch wird mit 0,45 n-Bariumhydroxyd in der Kälte verseift und die Lösung kontinuier lich mit Äther extrahiert, um das unverän derte Laktnn zu entfernen. Das Barium wird quantitativ mit Schwefelsäure entfernt und die Lösung nach Zugabe von etwas Py ridin zur Trockne eingedampft.
Der Rückstand wird mit trockenem Aceton extrahiert, in welchem ss-Alanin unlöslich ist. Das aceton- lösliche Material wird mit Diazomethan ver- estert, der Ester mit Essigsäureanhy drid und Pyridin acetyliert und bei 95 bis 110 und 10-5 mm Druck destilliert.
Das Reaktions produkt wird mit 0,45 n-Bariiunhydroxyd verseift, das Barium mit Schwefelsäure quantitativ entfernt, die Lösung mit NaOH neutralisiert (pA = 7,2); zur Trockne einge dampft und das so erhaltene Natriumsalz aus Isopropylalkohol fraktioniert. Das Natrium salz wird acetyliert, mit Bariumhydroxyd hydrolisiert, das Barium entfernt, die Lö sung zier Trockne eingedampft und der Nie derschlag mit Äther aufgenommen.
Aus dem Äther erhält man die reine Substanz, eine viskose gelbe Flüssigkeit, die die physiolo gische Wirksamkeit der Pantothensäure zeigt, deren Analyse mit der der Pantothensäure übereinstimmt und deren chemische Formel oben angegeben ist.
Die rae.emische Verbindung hat die halbe Wirksamkeit der c1-Form, die man aus<I>1-a-</I> Oxy-ss,ss-dimethyl-y-butyrola.l@ton nach der oben angegebenen Vorschrift erhält.
2. ss-Alanin und einem Äquivalent Na- triumäthylat in alkoholischer Lösung und Eindampfen stellt man das Natriumsalz dieser Aminosäure her. 12,8 g a-Oxy-i3,ss-di- methyl-y-buty rolakton werden mit einem Überschuss des Na-Salzes von ss-Alanin ver mischt und 1 Stunde im Rohr auf 95 erhitzt. Dabei ist auf unbedingten Wasserausschluss zu achten, da sonst die Ausbeute leidet.
Beim Erhitzen schmilzt das Lakton, und die Reaktionsteilnehmer mischen sich gut durch. Nach einstündigem Erhitzen wird die Reaktion abgebrochen, das Natriumsalz der Pantothensäure kann nach Beispiel 1 isoliert werden.
Die so hergestellte Pantothensäure ist ein Wirksteff für Pflanzen und niedere Orga nismen und kann deshalb insbesondere in der Gärungsindustrie und in der Landwirtschaft Verwendung finden. Die Pantothensäure übt gewisse Wirkungen auch im Säugetierorga- nismus aus. Es sind deshalb auch medizi nische Anwendungsmöglichkeiten offen.
Process for the preparation of a compound having the effects of pantothenic acid e. The present invention is a process for the preparation of a connec tion with the effects of pantothenic acid.
Pantothenic acid is a substance that is widespread in natural substances (food) and is best demonstrated by its growth effects in lower organisms, but it is also valuable for the nutrition of higher animals. The natural pantothenic acid can be split hydrolytically into two components and these components, one of which is ss-alanine, are chemically combined again to form the fully effective pantothenic acid.
It is assumed that a peptide-like linkage occurs between the ss-alanine and a diogyvaleric acid. One should therefore be able to synthesize pantothenic acid if one knows of the many possibilities that dioxyvaleric acid, which is a component of natural pantothenic acid.
In contrast to this view, which research in the present field has so far reached, it has been found that a compound can be produced which behaves physiologically, chemically and physically like pantothenic acid if it is synthetically obtained, levorotatory a-oxy-ss, ss-dimethyl-y-butyrolactone (i.e. the lactone of a dioxycaproic acid)
condensed with β-alanine under such conditions that a peptide-like linkage (formation of an acid amide grouping) occurs. The procedures that can be used for this reaction are known from the chemistry of the preparation of the peptides.
The u-oxy-ss, ss-dimethyl-butyrolactone is known (E. Glaser, monthly for chemistry, volume 39, 1918, page 293). For condensation, it can be used in the racemic form or as the 1-form, which can easily be obtained through the usual cleavage with the aid of optically active bases (e.g. quinine). The ss-alanine can be used for the condensation as such or as a suitable derivative, z. B. can be used as an ester or a salt.
According to the invention, a dextrorotatory compound is formed from the levorotatory lactone and ss-alanine, folo: ender constitution:

The reaction mixture is saponified with 0.45 n-barium hydroxide in the cold and the solution is continuously extracted with ether in order to remove the unchanged lactine. The barium is removed quantitatively with sulfuric acid and the solution is evaporated to dryness after adding some pyridine.
The residue is extracted with dry acetone, in which ß-alanine is insoluble. The acetone-soluble material is esterified with diazomethane, the ester is acetylated with acetic anhydride and pyridine and distilled at 95 to 110 and 10-5 mm pressure.
The reaction product is saponified with 0.45 N barium hydroxide, the barium is removed quantitatively with sulfuric acid, the solution is neutralized with NaOH (pA = 7.2); evaporated to dryness and the sodium salt thus obtained fractionated from isopropyl alcohol. The sodium salt is acetylated, hydrolysed with barium hydroxide, the barium removed, the solution evaporated to dryness and the precipitate absorbed with ether.
The pure substance is obtained from the ether, a viscous yellow liquid which shows the physiological effectiveness of pantothenic acid, the analysis of which corresponds to that of pantothenic acid and whose chemical formula is given above.
The chemical compound has half the effectiveness of the c1 form, which is obtained from <I> 1-a- </I> Oxy-ss, ss-dimethyl-y-butyrola.l @ ton according to the procedure given above.
2. SS-alanine and one equivalent of sodium ethylate in alcoholic solution and evaporation are used to produce the sodium salt of this amino acid. 12.8 g of α-oxy-13, ß-dimethyl-γ-buty rolactone are mixed with an excess of the sodium salt of ß-alanine and heated to 95 in a tube for 1 hour. It is important to ensure the exclusion of water, otherwise the yield will suffer.
When heated, the lactone melts and the reactants mix well. After heating for one hour, the reaction is terminated; the sodium salt of pantothenic acid can be isolated according to Example 1.
The pantothenic acid produced in this way is an active ingredient for plants and lower organisms and can therefore be used in particular in the fermentation industry and in agriculture. Pantothenic acid also has certain effects in the mammalian organism. Medical applications are therefore also open.
