Method for representing a new tetraeyelic neton It has been found that a new tetracyclic concrete can be obtained if a dI-3-oxy-17a-acyloxy-D-homo-androstene is treated with oxidizing agents and the hydrolyzing agent thus obtained is allowed to act.
While the free 17a-oxy compound is used as the starting material in main patent no. 230498, 17a-acyloxy compounds are used here to prevent oxidation in the 17a position.
The starting materials are accessible, for example, from cyclopentanopolyhydrophenanthene compounds by ring expansion, by total synthesis and optionally subsequent methods known per se for converting, introducing or eliminating substituents and / or multiple bonds.
The chemical, biochemical or electrochemical methods that are known for converting secondary alcohols into concretes and especially those that were also used in the steroid series can be used for oxidation. Under certain circumstances, intermediate protection of the carbon double bond is advantageous, for example when using oxidizing agents such as chromic acid.
The oxidation according to the invention can also be carried out by means of dehydrating agents, in particular concretes, in the presence of metal alcoholates or metal phenates.
The d4-3-keto-17a-oxy-D-homo-androsten (D-homo-testosterone) of the formula obtained according to the new process
EMI0001.0037
forms colorless crystals. It should find therapeutic use or serve as an intermediate for the preparation of therapeutically ver usable compounds.
Example: 1 part d'-3-oxy-17a-acetoxy-D-homo-androstene is added (shown for example by reducing a d'-3J17-dioxy-ethio-cholenic acid ester with sodium and alcohol, dehydration, for example by means of amei sensic acid, addition of two hydroxyl groups to the doubly unsaturated compound resulting from ring expansion, elimination of water by means of potassium bisulfate,
Conversion of the keto group in the d'-3-oxy-17a-keto-D-homo-androstene in a manner known per se into a hydroxyl group, acetylation and finally partial saponification of the ester group in the 3-position) with 100 parts of absolute toluene and 10 parts of cyclohexanone, 10 parts of the solvent mixture evaporated again in vacuo for dehydration,
add 1 part of aluminum tertiary butoxide to the residue and reflux the whole for 11 / E hours. Then is steam distilled for 3 hours with water, the residue acidified and extracted with ether. On evaporation, the ether solution yields d4-3-keto-17a-acetoxy-D-homoandrosten, which can be crystallized or sublimed from dilute acetone.
It is then converted into d4-3-keto-17a-oxy-D-homo-androstene (D-homo-testosterone) of the formula by hydrolysing Dlitti, for example by boiling with 4% methanolic potassium hydroxide solution

convicted. This forms colorless crystals.
Instead of the 17a-acetate one can also start from other diol-17a-monoesters, for example propionate, butyrate or benzoate.
Instead of working in the presence of a metal alcoholate, it is also possible to work in the presence of a metal phenolate. Oxidation can also be done, for example, by heating with a metal such as copper in a vacuum, in a biochemical or electrochemical manner, or with chromic acid. When using chromic acid as the oxidizing agent, intermediate protection of the double bond, for example by means of halogen or hydrogen halide, is advantageous.
