Description

  

      'Verfahren    zur Herstellung von Progesteron.    Im Patent Nr. 215337 wurde beschrieben,  dass man aus     45"-Pregnenol-3-on-20    ein Pro  dukt mit dem Ausgangsmaterial gegenüber  erhöhter physiologischer Wirkung erhält,  wenn man     A",-Pregnenol-3-on-20    in einem  Lösungsmittel, welches weder     Hydroxyl-          gruppen    noch     Ketogruppen    enthält, mit       Leichtmetallalkoholaten    behandelt und aus  dem Reaktionsgemisch die Metallverbindun  gen und das Lösungsmittel entfernt.  



  Es wurde nun gefunden, dass man aus  dem so erhaltenen Produkt reines Progeste  ron gewinnen kann, wenn man aus dem  rohen Gemisch die     hydroxylhaltigen    Anteile  entfernt.  



  Diese Trennung kann in beliebiger, an  sich bekannter Weise vorgenommen werden,  z. B. durch Verestern mit einem     Acylie-          rungsmittel,    welches nach erfolgter     Vereste-          rung    noch eine zur Salzbildung befähigte  Gruppe enthält.  



  Die Ester können dann in Form ihrer  wasserlöslichen Salze vom     unveresterten        hy-          droxylfreien    Anteil getrennt werden. Der         hy        droxylfreie    Anteil besteht aus Progesteron,  welches durch     Umkristallisieren    gereinigt  werden kann.  



  Gegenstand des vorliegenden Patentes ist  somit ein Verfahren zur Herstellung der zu  Heilzwecken verwendbaren Progesteron, wel  ches dadurch gekennzeichnet ist, dass man       d5-Pregnen-3-ol-20-on    in einem Lösungsmit  tel, welches weder     Hydroxylgruppen    noch       Ketogruppen    enthält, mit     Leichtmetallalko-          holaten    behandelt, aus dem Reaktionsgemisch  die Metallverbindungen und das Lösungs  mittel entfernt und aus dem so erhaltenen  Rohprodukt die     hydroxylhaltigen    Anteile  entfernt.  



  <I>Beispiel:</I>  24 g     45,s        Pregnen-ol-3-on-20    werden in  600 cm'     Toluol    mit 60 g tertiärem     Alumi-          niumbutylat    24 Stunden unter     Rückfluss    er  wärmt. Das gelbe Reaktionsgemisch wird  darauf mit verdünnter Schwefelsäure gründ  lich ausgeschüttelt, mit Wasser gewaschen,  mit Natriumsulfat getrocknet und im Va  kuum trocken gedampft. Der Rückstand      wird in 180 cm'     Pyridin    mit 50 g     Phtal-          säureanhydrid    3 Stunden auf dem Wasser  bad erwärmt.

   Dann wird das Gemisch in  2 Liter     n/2-NaOH    gegossen und das Ganze       ausgeäthert.    Die ätherische Schicht wird mit  Wasser gründlich gewaschen, mit Natrium  sulfat getrocknet und trocken gedampft. Das  hinterbleibende rohe Progesteron wird aus  Methanol umkristallisiert. Ausbeute 4,3 g.       Smp.    129 bis 130  .

  The esters can then be separated from the unesterified, hydroxyl-free portion in the form of their water-soluble salts. The hydroxyl-free part consists of progesterone, which can be purified by recrystallization.

The yellow reaction mixture is then shaken out thoroughly with dilute sulfuric acid, washed with water, dried with sodium sulphate and evaporated dry in a vacuum. The residue is heated in 180 cm 'pyridine with 50 g of phthalic anhydride on a water bath for 3 hours.

   The mixture is then poured into 2 liters of n / 2 NaOH and the whole thing is extracted with ether. The ethereal layer is washed thoroughly with water, dried with sodium sulfate and evaporated dry. The remaining crude progesterone is recrystallized from methanol. Yield 4.3g. M.p. 129 to 130.

PATENTANSPRUCH: Verfahren zur Herstellung von Progeste ron, dadurch gekennzeichnet, dass man d; Pregnen-3-ol-20-on in einem Lösungsmittel, welches weder Hydrogylgruppen noch Keto- gruppen enthält, mit Leichtmetallalkoholaten behandelt, aus dem Reaktionsgemisch die Metallverbindungen und das Lösungsmittel entfernt und aus dem so erhaltenen Rohpro dukt die hydroxylhaltigen Anteile entfernt.
