Verfahren zur Darstellung von 2,4-Dioxo-3-methyl-3-allyl-1,2,3,4-tetrahydrochinolin. Die Darstellung von 2,4-Dioxy-3-alkyl- chinolin ist bekannt (Berichte der Deutschen Chemischen Gesellschaft 60 [1927], S. 832; deutsche Patentschrift Nr. 490274).
An glei cher Stelle wird die Gewinnung des 2,4-Di- oxo - 3,3 - diäthyl -1,2,3,4-tetrahydrochinolins beschrieben. Es ist aber ummöglich, die Di- äthylverbindung auf diesem Wege herzustel len.
Die Forscher gewannen durch Um setzung von Diäthylmalonester mit Anilin in schlechter Ausbeute (S. 834) ein Dianilid vom Schmelzpunkt 217-218'. Eigene Ver suche ergaben, dass das Monoäthylmalon- säure-dianilid, dass aus reinem MonoätIlyl- malonester hergestellt wird, einen Schmelz punkt von 217-218 hat,
und dass man durch Umsetzung von käuflichem Diäthyl- malones.ter mit Anilin in geringer Menge ein Dianilid vom Schmelzpunkt 217-218' iso lieren kann, das mit dem Monoäthylmalon- säuredianilid aus Monoäthylmalonester keine Depression des Schmelzpunktes zeigt. Offen bar handelt es sich bei der in den Berichten der Deutschen Chemischen Gesellschaft 60 1927], S.
840 beschriebenen Verbindung um Monoäthylmalonsäuredianilid. Diese Auf- fassung findet eine weitere Stütze darin, dass der Schmelzpunkt von<B>260'</B> des vermeint lichen 2,4-Dioxo--3,3-diäthyl-1,2;3,4-tetra- hydrochinolins dem auf S. 839 angegebenen Schmelzpunkt des 2;4-Dioxy-3-äthsl-cbino- lins (259 ) fast gleich ist. Ausserdem wurde Diäthylmalon äuredianild aus Diäthylmalo- nylchlorid und Anilin hergestellt.
Es gelingt aber unter den auf S. 840 angegebenen Be- dingungen keinesfalls; Anilin abzuspalten und zum 2,4-Diogo-3,3-diäthyl-1,2,3,4-tetra- hydrochinolin zu gelangen.
Die irrtümlichen Angaben finden ihre Erklärung darin, dass der käufliche Diäthyl- malonester immer geringe Mengen Mono- äthylmalonester enthält. Es muss deshalb als erwiesen gelten, dass bis jetzt keine 2,4-Di- oxo - 3,3 - dialkyl -1,2,3,4-tetrahydrochinoline bekannt geworden sind.
Es wurde nun gefunden, dass man zu dem 2,4 - Dioxo - 3 - methyl - 3 -allyl -1,2,3,4 -tetra- hydrochinolin gelangen kann, wenn man die Alkalisalze des 2,4-Dioxy-3-methyl-chinolins in wässeriger Lösung mit Allylhalogeniden bei Gegenwart von Spuren Kupfer umsetzt.
Das entstehende 2,4-Dioxo-3-methyl-3- allyl-1,2,3,4-tetrahydrochinolin ist eine farb lose kristallisierte Verbindung, die bei 99 bis<B>100'</B> schmilzt und sich in den gebräuch lichen organischen Lösungsmitteln leicht löst. In Wasser ist sie schwer löslich. Ihre Alkali salze zeigen in wässeriger Lösung gelbe Farbe. Beim Ansäuern fällt die unveränderte farblose Verbindung wieder aus. Bei der ka talytischen Hydrierung geht der Allyl- in den Propylrest über.
Die neue Verbindung zeigt gute krampflösende und krampfhem mende Eigenschaften bei epileptiformen Krämpfen, die mit Hilfe des Diäthylamino- äthyl-phenyläther-hydrochlorids (Sivadjian, Anesth. et Analg. 3, [1937], S. 315) erzeugt wurden.
Sie soll als Arzneimittel Verwendung finden.
<I>Beispiel:</I> '\ 175 Gewichtsteile 2,4-Dioxy - 3 -methyl- chinolin (Schmelzpunkt = 265') werden ,in 800 Gewichtsteilen Wasser, in welchen 40 Gewichtsteile Ätznatron gelöst sind, durch Rühren in Lösung gebracht. Nachdem man 0,5 Gewichtsteile Kupferpulver zu gefügt hat, giesst man auf einmal 121 Ge wicUtsteile Allylbromid unter kräftigem Rühren zu. Nach kurzer Zeit beginnt die Temperatur infolge der Umsetzungswärme langsam zu steigen.
Das 2,4-Dioxo-3-methyl- 3-allyl-1,2,3,4-tetrahydrochinoliii fällt zu erst ölig aus und wird gegen den Schluss der Umsetzung fest. Wenn neutrale Reaktion eingetreten ist, wird genutscht. Zur Reini gung löst man in der dreifachen Menge Me thylalkohol und fügt wenig Wasser bis zur beginnenden Trübung zu.
Process for the preparation of 2,4-dioxo-3-methyl-3-allyl-1,2,3,4-tetrahydroquinoline. The preparation of 2,4-dioxy-3-alkylquinoline is known (reports from the Deutsche Chemischen Gesellschaft 60 [1927], p. 832; German patent specification No. 490274).
At the same point, the production of 2,4-dioxo-3,3-diethyl -1,2,3,4-tetrahydroquinoline is described. But it is impossible to produce the diethyl compound in this way.
By reacting diethyl malonic ester with aniline, the researchers obtained a dianilide with a melting point of 217-218 'in poor yield (p. 834). Our own tests showed that the monoethyl malonic acid dianilide, which is produced from pure monoethyl malonic ester, has a melting point of 217-218,
and that by reacting commercially available diethyl malones.ter with aniline in small amounts, a dianilide with a melting point of 217-218 'can be isolated which does not show any depression of the melting point with the monoethylmalonic dianilide made from monoethylmalonic ester. Apparently it is the case in the reports of the German Chemical Society 60 1927], p.
840 described compound to Monoäthylmalonsäuredianilid. This view is further supported by the fact that the melting point of <B> 260 '</B> of the supposed 2,4-dioxo-3,3-diethyl-1,2; 3,4-tetrahydroquinoline is almost the same as the melting point of 2; 4-dioxy-3-ethyl-cbino-lin (259) given on p. 839. In addition, diethylmalon acid dianild was produced from diethylmalonyl chloride and aniline.
But under the conditions given on p. 840 it does not succeed under any circumstances; To split off aniline and to arrive at 2,4-diogo-3,3-diethyl-1,2,3,4-tetrahydroquinoline.
The erroneous information is explained by the fact that the commercially available diethyl malonic ester always contains small amounts of monoethyl malonic ester. It must therefore be taken as proven that no 2,4-dioxo-3,3-dialkyl -1,2,3,4-tetrahydroquinolines have become known to date.
It has now been found that the 2,4-dioxo-3-methyl-3-allyl -1,2,3,4-tetra-hydroquinoline can be obtained if the alkali metal salts of 2,4-dioxy-3- methyl quinoline in aqueous solution with allyl halides in the presence of traces of copper.
The resulting 2,4-dioxo-3-methyl-3-allyl-1,2,3,4-tetrahydroquinoline is a colorless crystallized compound that melts at 99 to 100 'and is in the easily dissolves common organic solvents. It is sparingly soluble in water. Their alkali salts show yellow color in aqueous solution. When acidified, the unchanged colorless compound precipitates out again. In the catalytic hydrogenation, the allyl changes to the propyl radical.
The new compound shows good antispasmodic and antispasmodic properties in epileptiform cramps, which were generated with the help of diethylaminoethyl phenyl ether hydrochloride (Sivadjian, Anesth. Et Analg. 3, [1937], p. 315).
It should be used as a medicine.
<I> Example: </I> '\ 175 parts by weight of 2,4-dioxy-3-methylquinoline (melting point = 265') are dissolved in 800 parts by weight of water, in which 40 parts by weight of caustic soda are dissolved, by stirring . After 0.5 part by weight of copper powder has been added, 121 parts by weight of allyl bromide are poured in all at once with vigorous stirring. After a short time, the temperature begins to rise slowly due to the heat of conversion.
The 2,4-dioxo-3-methyl-3-allyl-1,2,3,4-tetrahydrochinoliii precipitates out oily at first and solidifies towards the end of the reaction. If there is a neutral reaction, it is sucked. For cleaning purposes, three times the amount of methyl alcohol is dissolved and a little water is added until it starts to become cloudy.
