Description

  

  Verfahren zur Darstellung einer     in4-Stellung    eine basisch substituierte     Aminogruppe     enthaltenden     Chinolinverbindung.       Durch das D.     R.-Patent    486079 und durch  seine Zusatzpatente werden Verfahren zur  Herstellung von in ihrer     Aminogruppe    basisch  substituierten     Aminochinolinen    und deren     Sub-          stitutionsprodukte    geschützt. Die nach den  genannten Patenten erhältlichen     Chinolinver-          bindungen    sind durch ihre Wirkung gegen  Blutparasiten, insbesondere gegen die Er  reger der Malaria, ausgezeichnet.  



  Es hat sich nun weiter gezeigt, dass un  ter den nach dem Verfahren des D.     R.-Pa-          tentes    486079 und seiner Zusatzpatente er  hältlichen     Chinolinverbindungen    diejenigen be  sonders gut auf die ungeschlechtliche Form  der     Malariaparasiten    wirken, bei denen die  basisch substituierte     Aminogruppe    in der       4-Stellung    des     Chinolinkerns    steht und die  zugleich mindestens in der     7-Stellung    einen  weiteren     Substituenten    tragen.  



  Gegenstand des vorliegenden Patentes ist  ein Verfahren zur Herstellung einer in 4-Stel-         lung    eine basisch substituierte     Aminogruppe     enthaltenden     Chinolinverbindung,    dadurch ge  kennzeichnet, dass man     4,7-Dichlorchinolin     mit     1-Diäthylamino-4-aminopentan    zu     4-(5'-          Diäthylaminopentyl-2'-amino)-7-chlorchinolin     umsetzt. Die Umsetzung erfolgt zum Bei  spiel durch Erwärmen der genannten Aus  gangsstoffe auf höhere Temperatur, zum Bei  spiel auf etwa<B>1800.</B> Die Aufarbeitung der  Reaktionsmischung kann in üblicher Weise  erfolgen.

   Das so erhaltene neue Produkt  siedet unter 0,2 mm Druck bei     212-2140.     Es erstarrt zu Kristallen, die nach dem Um  lösen aus Benzol bei<B>880</B> schmelzen.  



  Das neue Produkt soll therapeutische An  wendung finden.         Beispiel:     105 g     4,7-Dichlorchinolin    vom F. 93-94   werden mit 200 g     1-Diäthylamino-4-amino-          pentan    7 Stunden unter Rühren in einem      Ölbad auf 180   erhitzt, bis eine in verdünn  ter Salpetersäure gelöste Probe mit     Natrium-          acetatlösung    keine Fällung mehr gibt. Das  Gemisch wird in verdünnter Essigsäure ge  löst und durch Zusatz von Natronlauge al  kalisch gemacht. Die Base wird mit Äther  extrahiert, über     Kaliumcarbonat    getrocknet  und nach Verdampfen des Äthers fraktio  niert.

   Man erhält so das     4-(5'-Diäthylamino-          pentyl-2'-amino)-7-chlorchiriolin    vom Kp.     o,2     212-214  . Beim Abkühlen erstarrt die Ver  bindung kristallin; sie schmilzt, aus Benzol  umgelöst, bei<B>880.</B> Mit     2,4-Dioxybeiizoesäui-e     liefert die Base ein farbloses, kristallines  Salz vom F.     228-229'.    Das     Pikrat    der  Base zeigt den F. 207  .



  Process for the preparation of a quinoline compound containing a basic substituted amino group in the 4-position. The D. R. patent 486079 and its additional patents protect processes for the production of aminoquinolines with basic substituted amino groups and their substitution products. The quinoline compounds obtainable according to the patents mentioned are distinguished by their action against blood parasites, in particular against the pathogens causing malaria.



  It has now also been shown that among the quinoline compounds obtainable by the process of DR patent 486079 and its additional patents, those in which the basic substituted amino group is in the 4-position have a particularly good effect on the asexual form of malaria parasites of the quinoline nucleus and which at the same time carry a further substituent at least in the 7-position.



  The present patent relates to a process for the preparation of a quinoline compound containing a basic substituted amino group in the 4-position, characterized in that 4,7-dichloroquinoline is converted with 1-diethylamino-4-aminopentane to give 4- (5'-diethylaminopentyl -2'-amino) -7-chloroquinoline converts. The reaction takes place, for example, by heating the starting materials mentioned to a higher temperature, for example to about 1800. The reaction mixture can be worked up in the customary manner.

   The new product thus obtained boils at 212-2140 under 0.2 mm pressure. It solidifies to form crystals which, after dissolving from benzene, melt at <B> 880 </B>.



  The new product is intended to find therapeutic applications. Example: 105 g of 4,7-dichloroquinoline of F. 93-94 are heated with 200 g of 1-diethylamino-4-aminopentane for 7 hours in an oil bath with stirring until a sample dissolved in dilute nitric acid is acetate solution no longer gives rise to precipitation. The mixture is dissolved in dilute acetic acid and made alkaline by adding sodium hydroxide solution. The base is extracted with ether, dried over potassium carbonate and fractionated after evaporation of the ether.

   This gives 4- (5'-diethylaminopentyl-2'-amino) -7-chlorochirioline of bp 0.212-214. On cooling, the connection solidifies in crystalline form; when redissolved from benzene, it melts at <B> 880. </B> With 2,4-dioxybeiizoesäui-e the base gives a colorless, crystalline salt with a melting point of 228-229 '. The picrate of the base shows the F. 207.
