Verfahren zur Darstellung von 3-Nitro-4.6-diaminocbinaldin. Es wurde gefunden, dass Derivate von 4.6-Diaminochitiolinen, die eine 1Vitro- oder Aniinogruppe in 3-Stellung enthalten und deren 6-Amitiogruppe einen Acylrest ent halten kann, als Therapeutika und als Aus gangsmaterialien für Therapeutika von be sonderem Wert sind.
Die neuen Produkte können durch Ni trierung von 4. 6-Diaminochinolinen, z. B. der in den Schweizer Patentschriften Nr. 166787, 170877 und 173620 beschriebenen, und durch Reduktion der so erhaltenen Nitroverbin- dungen hergestellt werden.
Man kann so die 3-Nitroverbindungen in einer Ausbeute von zum Beispiel etwa 86 % der Theorie erhal- ten, wobei aus den Mutterlaugen nur unde finierbare Nebenprodukte abgetrennt werden können. Besonders die 6-Acylamino-3.4-di- aminochinoline zeichnen sich durch die glei chen guten chemotherapeutischen Wirkungen aus wie die Ausgangsstoffe und haben zum Teil noch eine starke Wirksamkeit gegen Blutparasiten.
Es war nicht vorauszusehen, dass die Nitrogruppe in die 3-Stellung eintreten würde, vielmehr war mit einem zumindest über wiegenden Eintritt in die 5-Stellung zu rechnen.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Darstellung von 3-Nitro- 4.6-diaminochinaldin, das dadurch gekenn zeichnet ist, dass man 4. 6-Diaminoehinaldin nitriert. Man kann die Nitrierung bei Tem peraturen unter 0 o C durchführen und die Temperatur dann langsam .auf Zimmertem peratur steigern.
Das neue Reaktionsprodukt ist rot ge färbt und schmilzt bei 225-226 o C. Es soll als Zwischenprodukt für die Herstellung von Arzneimitteln verwendet werden. <I>Beispiel:</I> 8,6 gr 4.6-Diaminochinaldin werden unter Eiskühlung in 40 cm3 konzentrierter Schwe felsäure eingetragen. Darnach wird unter Rühren und Abkühlen auf - 10 bis - 5<B>1</B> C 5 gr Kalisalpeter allmählich hinzugefügt. Nach kurzer Zeit gibt eine Probe beim Neu tralisieren einen gelbroten Niederschlag, der in Natronlauge und in Ammoniak löslich ist und durch Essigsäure wieder gefällt wird.
Es hat sich also zunächst offenbar ein Ni- tramin gebildet. Wird nun die Kältemischung entfernt und die Temperatur langsam auf Zimmertemperatur gebracht, so gibt jetzt eine Probe auf Zusatz von überschüssiger Natronlauge einen dunkelroten Niederschlag. Nach zweistündigem Stehen wird auf Eis gegossen, mit Natronlauge alkalisch gemacht, der braunrote Niederschlag abgesaugt und mit Wasser gewaschen. Er wird in heisser, stark verdünnter Essigsäure gelöst, worauf aus der klar filtrierten Lösung durch Zugabe von Ammoniak die schön rotgefärbte Base folgender Konstitution

ausgefällt wird. Sie schmilzt nach dem Um- kristallisieren aus wässerigem Alkohol bei 225-22U1 C.
Sie bildet mit überschüssiger Salzsäure ein in Alkohol schwer lösliches, fast farbloses Salz, das in Wasser mit rot gelber Farbe leicht löslich ist.
