Verfahren zur Herstellung von 1-(3,4-Dioayphenyl)-2-aminopropan-l-ol-ehlorhydrat. Es wurde gefunden, dass man durch, ge gebenenfalls in Gegenwart säurebindender Mittel erfolgendes Halogenieren von Mon.o bezw. Dibenzyloxypropiophenonen, Umsetzen der so erhaltenen Produkte mit Dibenzylamin und Behandlung der gebildeten Dibenzyl- aminoketonäther mit Wasserstoff in Gegen wart von Edelmetallkatalysatoren zu den im Phenylkern hydroxylierten 1-Phenyl-2-amino- propan-l-olen gelangt.
Die erhaltenen, an sich bekannten Produkte sind teils selber wertvolle Arzneimittel, teils sollen sie als Zwischenprodukte zur Herstellung anderer therapeutisch wertvoller Substanzen dienen.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 1-(3,4-Di- oxyphenyl) -2- aminopropan-l-ol-chlorhydrat, welches dadurch gekennzeichnet ist, dass man 3,4-Dibenzyloxypropiophenon halogeniert, das erhaltene halogenierte Produkt mit Dibenzyl- amin umsetzt,
den entstandenen Dibenzyl- aminoketonäther mit Chlorwasserstoff in sein Chlorhydrat überführt und dieses mit Was- serstoff in Gegenwart von Edelmetallkataly- satoren behandelt.
Das Verfahrensprodukt soll als Arznei mittel verwandt werden.
<I>Beispiel:</I> 34,6 gr 3,4 - Dibenzyloxypropiophenon werden in 300 cm3 Methylenchlorid gelöst und nach Zugabe von 12 gr Calciumkarbonat unter Rühren 16 gr Brom eingetropft. Nach beendigter Umsetzung wird vom Calcium- niederschlag abfiltriert und die Methylen- chloridlösung vollständig eingedampft.
Beim Erkalten kristallisiert aus dem Rückstand das 3,4-Dibenzylogy-a-brompropiophenou aus, das nach dem Umkristallisieren aus Hexahydro- benzol oder aus Alkohol bei 93 bis 94' schmilzt.
40 gr 3,4-Dibenzylogy-a-brompropiophenon werden in 100 cm' Benzol gelöst und nach Zugabe von 36 gr Dibenzylamin etwa sechs Stunden unter Rückfluss auf dem Wasserbad erhitzt. Nach mehrstündigem Stehen wird das abgeschiedene -Dibenzylamiuhydrobromid abgesaugt und die Mutterlauge vollständig vom Benzol befreit. Man nimmt alsdann den Rückstand in Äther auf, wäscht die Äther lösung mit etwas Wasser durch und leitet in die durch Natriumsulfat getrocknete Lö sung Salzsäuregas bis zur sauren Reaktion ein.
Das auskristallisierende Hydrochlorid wird abgesaugt, in Wasser verteilt und durch Zusatz von Ammoniak die Base wieder frei gemacht, die in Äther aufgenommen wird. Nach dem Trocknen über Natriumsulfat wird der Äther abdestilliert und das alsbald aus kristallisierende 3,
4-Dibenzyloxy-a-diben-zyl- aminopropiophenon aus Methanol umkristal lisiert.- Schmelzpunkt 84 bis 86 .- Man erhält daraus das Hydrochlorid durch Neutralisieren der Base mit der berechneten Menge alkoho lischer Salzsäure.
10 gr 3,4-Dibenzyloxy-a-dibenzylamirio- propiophenonhydrochlorid werden in 250 emä Alkohol gelöst und mit Palladium und Wasserstoff hydriert. Nach Aufnahme der berechneten Menge Wasserstoff wird vom Katalysator abgesaugt und die alkoholische Lösung im Vakuum bis zur Trockne ein gedampft. Man erhält durch Umkristallisie- ren des Rückstandes aus Methanol und Ace ton das 1-(3,4-Dioxyphenyl)-2-aminopropau- 1-ol-chlorhydrat vom Schmelzpunkt<B>178'.</B>
