Verfahren zur Herstellung von Alkylarylcarbinolen, welche in der aliphatischen Seitenkette einen basischen Rest aufweisen. Die bisher bekannten Verfahren zur ka talytischen Reduktion von a-Ketonen der Arylaminoäthanreihe und deren Homologen (vergleiche deutsche Patentschriften Nr. 254438 und 256750, sowie Mannich, Archiv der Pharm., 253, Seite 283<B>)</B> benutzen aus schliesslich Metalle der Platingruppe in kol loidaler oder anderer feinverteilter Form als Katalysatoren.
Es zeigte sich nun über raschenderweise, dass auch die auf bekann ten Trägern niedergeschlagenen Nickel katalysatoren ebenfalls in sehr guter Aus beute und wirtschaftlicher Weise die Re duktion zu den entsprechenden sekundären Alkoholen gestatten. Dies bedeutet gegen über den älteren Verfahren. eine erhebliche Ersparnis an teuerem Material, da für tech nische Zwecke die Hydrierung mit Metallen der Platinreihe nur in sehr wenigen Fällen lohnend ist.
Beispiele 1. 46 gr co-N-Methylaminoacetophenon- bi omhydrat werden in 500 cm' Wasser ge- löst, mit 110 %igem, auf bekannten Trägern niedergeschlagenem Nickelkatalysator ver setzt und mit Wasserstoff geschüttelt. Hier bei wird genau 1 Mol. Wasserstoff aufge nommen. Nach beendeter Hydrierung wird mit Alkali übersättigt und ausgeäthert. Nach dem Verjagen des Äthers hinterbleibt reines Phenyläthanolmethylamin vom Schmelzpunkt 7 7 C.
2,. 34,2 gr co-Aminoacetophenonchlor- hydrat werden in 250 cm' Wasser gelöst und nach Zugabe von Nickelkatalysator bei etwa 35 mit Wasserstoff geschüttelt, wo bei die für die Reduktion der Ketogruppe berechnete Menge Wasserstoff aufgenommen wird. Das nach Beispiel 1 abgeschiedene Phenyläthanolamin schmilzt bei 40 .
3. 50 gr a-N-Methylaminopropiophenon- chlorhydrat Cjl, . CO . CH (NH . CH3) CH'3 . HCl werden in 200 cm' Wasser und 200 cm' Alkohol gelöst und wie im Bei spiel 1 weiter behandelt. Nach dem Ab dämpfen des Lösungsmittels im Vakuum kristallisieren unmittelbar 40 gr Chlorhydrat des a-N-Methylaminoäthylphenylcarbinols, da.s nach einmaligem Umkristallisieren aus Alkohol den Schmelzpunkt 189,5 gibt.
4. 10,6 gr o-Dioxy-co-N-methylamino- acetophenonchlorhydrat (co-Methylamino- acetobrenzcatechinchlorhydrat) werden in 150 cm' 50 %igem Alkohol gelöst, mit 10 gr 10 %igem Nickel-Bimsstein-Katalysator ver setzt und bei 30 bis 40' mit Wasserstoff geschüttelt. Hierbei wird die berechnete Menge Wasserstoff aufgenommen.
Nach Filtration und Einengen der Lösung im Va kuum fällt man die racemische Adrenalin- base mit Ammoniak aus. Ausbeute: 6,6 gr = 80 % der Theorie. Durch Erhöhung von Temperatur und Druck lässt sich in obigen Beispielen die Dauer der Hydrierung wesentlich abkürzen.
Process for the preparation of alkylaryl carbinols which have a basic radical in the aliphatic side chain. The previously known processes for the catalytic reduction of α-ketones of the arylaminoethane series and their homologues (compare German Patent Nos. 254438 and 256750, and Mannich, Archiv der Pharm., 253, page 283) are used exclusively Platinum group metals in colloidal or other finely divided form as catalysts.
Surprisingly, it was now found that the nickel catalysts precipitated on known supports also allow the reduction to the corresponding secondary alcohols in very good yield and economically. This means compared to the older method. a considerable saving in expensive material, since the hydrogenation with metals of the platinum series is only worthwhile in very few cases for technical purposes.
EXAMPLES 1. 46 g of co-N-methylaminoacetophenone biomhydrate are dissolved in 500 cm 2 of water, mixed with 110% nickel catalyst deposited on known carriers and shaken with hydrogen. Here at exactly 1 mole of hydrogen is taken up. When the hydrogenation has ended, the mixture is oversaturated with alkali and extracted with ether. After the ether has been chased away, pure phenylethanolmethylamine with a melting point of 7 7 C.
2 ,. 34.2 g of co-aminoacetophenone chlorohydrate are dissolved in 250 cm 'of water and, after the addition of nickel catalyst, shaken with hydrogen at about 35, where the amount of hydrogen calculated for the reduction of the keto group is absorbed. The phenylethanolamine deposited according to Example 1 melts at 40.
3. 50 gr of a-N-methylaminopropiophenone chlorohydrate Cjl,. CO. CH (NH. CH3) CH'3. HCl are dissolved in 200 cm 'of water and 200 cm' of alcohol and treated as in Example 1. After the solvent has been vaporized in vacuo, 40 grams of a-N-methylaminoethylphenylcarbinol hydrate crystallize immediately, since it has a melting point of 189.5 after recrystallizing once from alcohol.
4. 10.6 grams of o-dioxy-co-N-methylaminoacetophenone chlorohydrate (co-methylaminoacetobrenzcatechol chlorohydrate) are dissolved in 150 cm 'of 50% alcohol, mixed with 10 grams of 10% nickel-pumice catalyst and at Shaken for 30 to 40 'with hydrogen. The calculated amount of hydrogen is taken up.
After filtration and concentration of the solution in vacuo, the racemic adrenaline base is precipitated with ammonia. Yield: 6.6 g = 80% of theory. In the above examples, the duration of the hydrogenation can be significantly shortened by increasing the temperature and pressure.
