Abstract
L'invention concerne une colonne de séparation améliorée et un procédé correspondant pour séparer un mélange de fragments d'ADN à double brin par chromatographie MIPC. La colonne cylindrique, qui a un diamètre interne supérieur à environ 5 mm, contient des perles de polymère dont le diamètre moyen est compris entre 1 et 100 microns. Il s'agit de perles non substituées ou substituées par une fraction hydrocarbure qui présente entre 1 atome de carbone et un million d'atomes de carbone. De préférence, ces perles sont sensiblement dépourvues de cations multivalents qui peuvent se lier librement avec l'ADN. La colonne améliorée assure une meilleure séparation des fragments d'ADN ayant une taille comprise entre environ 100 et 20 000 paires de base. En outre, cette colonne assure une séparation améliorée pour les molécules d'ADN hétéroduplex et homoduplex dans une procédure de détection de mutation où la chromatographie est accomplie selon des conditions de dénaturation partielle de l'ADN en un site de paires de base présentant un défaut d'appariement.
