Abstract

L'invention concerne une méthode pour l'identification et la représentation de différences interindividuelles de séquences d'ADN. Elle est caractérisée en ce que l'on isole l'ADN génomique des individus à analyser, on définit des segments des gènes, des régions génomiques et des génomes à comparer, on isole de manière ciblée ces segments au moyen de l'amplification en chaîne par polymérase en utilisant des oligonucléotides très spécifiques, et on en fabrique un nombre suffisant de copies, on réalise un séquençage simultané de tous les segments géniques obtenus qui se présentent désormais sous la forme de produits d'amplification en chaîne par polymérase, on sépare ensuite par voie électrophorétique sur un gel de séquençage le mélange ainsi formé issu des réactions de terminaison et contenant les séquences individuelles de tous les segments géniques, et on le transfère sur une membrane par électrophorèse directe de transfert, puis on met en évidence, par hybridation de la membrane avec un oligonucléotide très spécifique marqué, les séquences individuelles d'un segment génique déterminé provenant du mélange initial, on répète cette opération jusqu'à ce que tous les segments géniques provenant du groupe initial soient détectés, et on détecte ensuite par analyse d'image les différences interindividuelles de séquences d'ADN, et on établit et on évalue par traitement mathématique un modèle de profil génique.
