Abstract

L'invention porte sur un procédé de détection simultanée de polygènes comportant les étapes (a) à (f) suivantes: (a) conversion d'un ARNm en ADNc à l'aide d'amorces de transcription inverse selon le processus suivant: (1) présenter dans l'extrémité 3' terminale de l'oligo-désoxythymidine une partie d'ancrage de formule 5'-M(N)¿q?-3' (dans laquelle M représente une base sélectionnée parmi A, G et C; N représente une base sélectionnée parmi A, G, C et T, et q est 0 ou 1, (2) présenter dans l'extrémité 5' terminale de l'oligo-désoxythymidine un site de formation d'une phase solide, (3) présenter un site de coupure enzymatique entre le terminus 5'- de l'oligo-désoxythymidine et le site de formation d'une phase solide; (b) coupure de l'ADNc par une enzyme de restriction; (c) marquage du site de coupure de l'ADNc par l'enzyme de restriction; (d) formation d'une phase solide avec le fragment d'ADNc suivie de sa récupération; (e) coupure du site de coupure de l'enzyme au terminus (5) de l'amorce de transcription inverse par une enzyme pour obtenir un fragment d'ADNc solubilisé et marqué; et (f) soumission du fragment d'ADNc marqué à une électrophorèse bidimensionnelle sur gel.
