Abstract

Procédés améliorés destinés à empêcher le rejet de cellules transplantées chez un receveur allogénique ou xénogénique, qui consistent à modifier au moins un antigène sur la surface d'une cellule donneuse avant la transplantation pour réduire l'immunogénicité de la cellule chez le sujet receveur. De préférence, on modifie un antigène du complexe majeur d'histocompatibilité de classe I en mettant en contact la cellule avec une molécule qui se lie à l'antigène, telle qu'un anticorps ou fragment ou dérivé dudit anticorps. Selon les procédés de la présente invention, un sujet receveur est également traité avec un agent qui inhibe l'activité des lymphocytes T chez ledit sujet. Un agent préféré qui inhibe l'activité des lymphocytes T chez le receveur est un médicament immunodépresseur, tel que la cyclosporine A. Alternativement, des anticorps qui provoquent la déplétion des lymphocytes T ou empêchent la prolifération de lymphocytes T chez le sujet peuvent être utilisés. Typiquement, après administration d'une cellule donneuse modifiée à un sujet receveur allogénique ou xénogénique, un médicament immunodépresseur est administré au sujet receveur pendant une durée suffisante pour induire la tolérance aux cellules donneuses.
