Abstract

Le gène pour l'ADNase B de pyogène de streptocoque a été cloné et des vecteurs comprenant l'ADN cloné ont été utilisés pour transformer Escherichia coli, ceci permettant de produire efficacement et rapidement l'ADNase dans E. coli sans avoir besoin de développer de grandes quantités de S. pyogène. L'enzyme peut être produite avec un peptide de tête à sa terminaison amino. Un procédé amélioré de purification de l'enzyme ADNase B de S. pyogène d'origine naturelle est également décrit. L'enzyme d'ADNase B produit, soit par purification de l'enzyme d'origine naturelle soit par des techniques d'ADN de recombinaison, peut être utilisé afin de générer des anticorps et peut également être utilisé dans des dosages immunochimiques afin de détecter la présence d'anticorps anti-ADNase B dans du sérum comme un marqueur d'infection par S. pyogènes.
