The constrained sites SgfI and SnaBI were used to introduce the HN gene from plasmid pUCNDVlc, in which the HN gene was also flanked by the SgfI and SnaBI restriction sites (Fig. 1 B). Finally, the resulting plasmid pUCNDVH5_1c was used to replace the pflNDV NotI / BsWI fragment (Fig. 1). The constructed pflNDV / AIVH5 -B differs from plasmidopflNDV / AIVH5-A in that the non-coding regions of NDV HN were additionally inserted between the transcriptional control elements (GS, GE) and the H5 ORF.
