Abstract

"PROCESSO PARA PROPAGAR UM VìRUS DO HERPES MUTANTE, USO DE UMA LINHAGEM DE CéLULA, LINHAGEM DE CéLULA, VìRUS, E, COMPOSIçãO FARMACêUTICA" Processo para a propagação de um vírus do herpes mutante, tendo uma mutação em seu gene VP16 HSV endógeno ou um seu homólogo, processo este compreendendo infectar uma linhagem de célula com o vírus do herpes mutante e cultivar a linhagem de célula, em que a linhagem de célula compreende uma seq³ência de ácido nucleico codificando um polipeptídeo VP16 do vírus do herpes simples funcional, ou um seu homólogo, operavelmente ligado a uma seq³ência de controle permitindo a expressão do polipeptídeo em dita linhagem de célula; a seq³ência de ácido nucleico sendo (i) capaz de complementar o gene endógeno e (ii) incapaz de sofrer recombinação homóloga com o gene endógeno. Além disso, a presente invenção fornece linhagens de células que podem ser usadas para o crescimento dos vírus do herpes mutante que têm defeitos em certos genes prematuros imediatos junto com mutação em VP16 ou seu homólogo.
