Abstract

COMPOSIÇÕES E MÉTODOS PARA AUMENTAR CAPACIDADE DE DISTRIBUIÇÃO SISTÊMICA, TOLERÂNCIA E EFICÁCIA DE PEPTÍDEOS MACROCÍCLICOS CATIÔNICOS. A presente invenção refere-se a composições que são fornecidas para formulações de ¿-defensina e/ou um análogo de ¿-defensina, as quais são altamente adequadas para administração parenteral. Essas formulações proporcionam a ¿-defensina e/ou um análogo da ¿-defensina em um tampão levemente ácido que inclui propilenoglicol. Surpreendentemente, inventores verificaram que essas formulações aumentam a biodisponibilidade de uma ¿-defensina e/ou um análogo de ¿-defensina, proporcionado assim por pelo menos um fator de 10 em relação a soluções salinas isotônicas convencionais, e que essas formulações aumentaram dramaticamente a biodisponibilidade em indivíduos humanos em relação a modelos animais. Os inventores também verificaram que tais formulações apresentam vantajosamente baixa viscosidade em altas concentrações de peptídeo, reduzindo o volume de injeção que permite esterilização por filtração simples.
