Abstract
"DETECçãO DE FLAVIVìRUS E ANáLISES DE QUANTIFICAçãO". São descritas ondas de DNA fluorescentes específicas e flanqueando pares de iniciadores, com base na seq³ência de informação encontrada na região não-codificadora de 3' terminal conservada de flavivírus, por exemplo, nucleotídeos 10653-10678 de vírus da dengue. Reação em cadeia de polimerase fluorogênica, empregando esse iniciadores e sondas, produzem resultados que permitem a identificação específica de flavivírus. Essa análises podem ser ambas quantitativas e qualitativas. São descritas condições ótima de análise com antecedente zero, a qual permite a detecção de baixos níveis de flavivírus a partir de espécimes clínicas. Fluorogenic polymerase chain reaction, employing these primers and probes, yield results that enable specific identification of flaviviruses. These analyzes can be both quantitative and qualitative. Optimal conditions of zero antecedent analysis are described, which allows the detection of low flavivirus levels from clinical specimens. Specifically, dengue virus, isolated from different geographic regions, can be universally detected and identified by the described fluorogency RT-PCR analysis. The described RT-PCR fluorogence analysis promptly detected viremia in sera collected from dengue sick individuals.
