Abstract

Prior art

The beneficial properties of cherries, sour cherries and Japanese cherries have been known for centuries. These delicious fruits have long been attributed with a number of medicinal properties - from a cure for rheumatism and arthritis to a food for restful sleep. They contain vitamins A, B and C, bioflavonoids, ellagic acid, anthocyanin, melatonin. These ingredients help fight cancer, prevent heart disease, relieve arthritis pain, headaches, and fibromyalgia. Cherries help the body to produce melatonin, improve physiological and mental functions. Cherries contain a large amount of vitamin C and zinc, which are very important for the immune system, they also contain folic acid for the health of the heart and circulatory system, calcium for the bones and iron for the blood. However, the most valuable in cherries are plant pigments called anthocyanins. They strengthen connective tissue and prevent the appearance of wrinkles as they destroy the damaging enzymes that age and wrinkle the skin. Cherries can prevent the formation of cellulite, and anthocyanins have an anti-inflammatory effect and prevent calf cramps. Thanks to acids and fruit sugar, cherries improve the work of the stomach, intestines and pancreas, thus stimulating digestion. In addition, these delicious fruits are a wonderful diuretic that relieves the heart and activates the liver and kidneys. Cherry pits contain small amounts of amygdalin, also called vitamin B17 or laetrile, which is believed to have a positive effect on shrinking tumors and preventing the further development of cancer, also helping to relieve pain caused by carcinogenic processes. Tea from the stems of cherries is recommended for better urination in case of kidney stones. Cherries are considered an extremely rich source of the element boron, which, in combination with calcium and magnesium, has an extremely beneficial effect on bone health. Cherries contain few calories, no fat, but have a high concentration of water, thanks to which energy increases and the body's metabolism accelerates. Compared to most fruits, cherries have a lot of potassium and little sodium, which prevents fluid retention in the body. Cherries are one of the fruits with the lowest glycemic index. Cherries are also known to purify the blood and increase energy, and thanks to the presence of active phytochemicals in them, the risk of developing Alzheimer's disease is reduced.

Each of the ingredients in the natural antioxidant composition - quercetin, extracts of green coffee, green tea, onion, apple, acerola, blackcurrant, blueberry, olive and concentrates of broccoli sprouts, camu camu, tomato, broccoli, acai berry, turmeric, garlic , basil, oregano, cinnamon, elderberry, carrot, mangosteen, cherry, blackberry, chokeberry, raspberry, spinach, kale and Brussels sprouts, has also long proven its healing and immunostimulating effect.

An anti-inflammatory agent is known which includes as an active ingredient an extract of Japanese sour cherry (Prunus serrulata), described in JP 2012 229 170 A.

However, no prior technical solution is known for a nutritional supplement based on a combination of cherry (Prunus avium), sour cherry (Prunus cerasus) and Japanese sour cherry (Prunus serrulata) extracts with a natural antioxidant composition - a combination of quercetin, green coffee extracts, green tea , onion, apple, acerola, currant, blueberry, olive and broccoli sprout concentrates, camu camu, tomato, broccoli, acai berry, turmeric, garlic, basil, oregano, cinnamon, elder, carrot, mangosteen, cherry, blackberry, chokeberry , raspberry, spinach, kale and Brussels sprouts.

Technical essence of the invention

The task of the invention is to create a nutritional supplement based on a combination of cherry (Prunus avium), cherry (Prunus cerasus) and Japanese cherry (Prunus serrulata) cherry extracts with a natural antioxidant composition - a combination of quercetin, extracts of green coffee, green tea, onion, apple, acerola, blackcurrant, blueberry, olive and broccoli sprout concentrates, camu camu, tomato, broccoli, acai berry, turmeric, garlic, basil, oregano, cinnamon, elder, carrot, mangosteen, cherry, blackberry, chokeberry, raspberry, spinach, kale and Brussels sprouts.

The task was solved by the proposed nutritional supplement based on a combination of extracts of cherry (Prunus avium), sour cherry (Prunus cerasus) and Japanese sour cherry, representing a combination of extracts of flowers, fruits, leaves and stems of cherry, sour cherry and Japanese sour cherry with a natural antioxidant composition , which is a combination of equal amounts of quercetin, green coffee extract, green tea, onion, apple, acerola, blackcurrant, blueberry, olive and broccoli sprout concentrates, camu camu, tomato, broccoli, acai berry, turmeric, garlic, basil, oregano, cinnamon, elderberry, carrot, mangosteen, cherry, blackberry, chokeberry, raspberry, spinach, kale and Brussels sprouts, cysteine and D-biotin.

The dietary supplement is in the form of cellulose capsules.

The contents of the cellulose capsules are a powdered mixture of the following components (in % by weight):

- 62% to 69%: a combination of extracts of flowers, fruits, leaves and stems of cherry (Prunus avium), where 20-40% is the flower extract, 10-20% - the fruit extract, 1-15% - the extract of leaves and 1-15% is the stem extract; of sour cherry (Prunus cerasus) - 1-15% extracts of flowers, fruits, leaves and stems in equal parts; and Japanese cherry (Prunus serrulata) - 25-45% extracts of flowers, fruits, leaves and stems in equal parts;

Examples of implementation of the invention

According to an exemplary embodiment of the invention, which only clarifies it, but does not limit it, the contents of the cellulose capsules are a powdered mixture of the following components (in % by weight):

• A combination of extracts of flowers, fruits, leaves and stems of cherry, sour cherry and Japanese cherry - 65%;

• Natural antioxidant composition - 26% • Cysteine - 8.99% • D-biotin - 0.01%

To prepare the composition of the nutritional supplement, the specified components are used, and when they are mixed, it is necessary to obtain a powdery mixture with evenly distributed components, which is encapsulated in the cellulose capsules.

With the indicated exemplary embodiment of the invention, in which the content of the cellulose capsules is a powdered mixture (in wt. %) of a combination of cherry (Prunus avium), cherry (Prunus cerasus) and Japanese cherry (Prunus serrulata) extracts - 65%, natural antioxidant composition - 26%, cysteine - 8.99% and D-biotin - 0.01%, a clinical trial was conducted that proved a new, unexpected synergistic effect of the mentioned combination.

More specifically, the contents of the cellulose capsules are a powdered mixture of the following components (in % by weight):

• 65% - a combination of extracts of flowers, fruits, leaves and stems of cherry (Prunus avium), sour cherry (Prunus cerasus) and Japanese sour cherry (Prunus serrulata), where cherry (Prunus avium) extracts are respectively 25% flower extract, 15% fruit extract, 10% leaf extract and 10% stem extract; from sour cherry (Prunus cerasus) - 10% extracts of flowers, fruits, leaves and stems in equal parts; and from Japanese cherry (Prunus serrulata) - 30% extracts of flowers, fruits, leaves and stems in equal parts - total 100%;

• 26% - natural antioxidant complex - a combination of equal amounts of quercetin, extracts of green coffee (Coffea arabica), green tea (Camellia sinensis), onion (Allium sepa), apple (Malus domestica), acerola (Malpighia punicifolia), blackcurrant ( Ribes nigrum), blueberry (Vaccinium myrtillus), olive (Olea europaea) and concentrates from sprouts of broccoli (Brassica oleracea var. italica), camu camu (Myrciaria dubia), tomato (Solanum lycopersicum), broccoli (Brassica oleracea var. Italica) , acai berry (Euterpe oleracea), turmeric (Curcuma longa), garlic (Allium sativum), basil (Ocimum basilicum), oregano (Origanum vulgare), cinnamon (Cinnamomum verum), elder (Sambucus nigra), carrot (Daucus carota), mangosteen (Garcinia mangostana), cherry (Prunus avium), blackberry (Rubus fruticosus), chokeberry (Aronia melanocarpa), raspberry (Rubus idaeus), spinach (Spinacia oleracea), kale (Brassica oleraceae) and Brussels sprouts (Brassica oleracea var. gemmifera) );

Before being used for the production of plant extracts, the plant raw material is subject to inspection according to the following parameters:

• authenticity of the plant • humidity • content of impurities • content of foreign organic and mineral bodies

The initial plant raw material, which meets the quality requirements, is treated with ascorbic and malic acid. This is followed by water-ethanol extraction, which allows the valuable bioactive substances in the plants to be preserved to the maximum extent. After the extraction process, a filtration process follows. The filtered plant extract is concentrated to a certain degree or not concentrated, according to the requirements of the recipe. Maltodextrin is added to the resulting extract and sterilization, drying and quality control processes follow. The plant extracts are mixed in a mixing room according to the recipe and the powdered mixture is encapsulated in cellulose capsules. Capsules are subject to visual inspection to eliminate the possible presence of damaged or incomplete capsules.

Clinical trial I

Chloasma (melasma), acquired hypermelanosis, is often refractory to various treatments and an efficient as well as safe way to reduce pigmentation is needed.

In an open-label, uncontrolled study conducted over a period of 16 weeks, the reducing effect of the above-mentioned natural complex on chloasma was investigated without the use of topical lightening therapy.

The study, which was conducted over 4 months between November 2018 and February 2019, involved 20 women (aged 19-44) with chloasma from Bulgaria who took 1 capsule per day of the dietary supplement orally. containing a combination of extracts of cherry, sour cherry and Japanese sour cherry, antioxidant systaw, cysteine and D-biotin, in the proportions indicated in the above-described exemplary embodiment. Patients were evaluated every 4 weeks using the MASI (Melasma Area Severity Index) and the Mexameter.

The results showed a significant reduction (p<0.05) in the degree of pigmentation in both parameters, and an improved quality of life was also noted in terms of MELASQOL (Melasma Quality of Life Scale) scores. The pigment density of the patients was assessed biometrologically - using the Mexameter, and subjectively - using a visual examination before and after 16 weeks of treatment.

In 15 patients with bilateral epidermal melasma, the pigment spots decreased by 62% at the end of the follow-up period. In 5 patients with dermal melasma, pigment spots were reduced by 51%.

Melanin index significantly decreased after 4 months of administration (0.025 +/- 0.005 at baseline vs. 0.019 +/- 0.004 after 4 months) (p<0.01).

The clinical trial was conducted as an open-label, single-center study in accordance with Good Clinical Practices. Participants had dull skin complexion assessed by clinical assessment, phototypes I to II on the Fitzpatrick scale, with no history of allergy or hypersensitivity to the products or one of their components. After signing written, informed consent, subjects were first screened for inclusion and exclusion criteria: dietary habits (stable before and during the study); lack of concomitant treatment; no history of skin disease or treatment; no care for the skin of the face (peeling, mask, etc.) during the study and 15 days before the start of the study; smoking (no smoking or <5 cigarettes per day); no UV exposure during the study; not pregnant, breastfeeding or likely to become pregnant during the study; no use of oral nutritional supplements and/or vitamin supplements (not <1 month before enrollment and not during the study).

Environmental factors affect the aging of the skin, which leads to a decrease in the radiance of the skin. The way you eat, and especially the consumption of antioxidants, can help preserve the youthful appearance of the skin.

The aim of this study was to evaluate the effects of taking an oral supplement rich in specific antioxidants on improving skin radiance in women.

In the open clinical study, 15 women aged 45 - 65 years, with a dull complexion and skin without shine on the face, were included. Subjects took an oral, daily dose of 1 capsule of a dietary supplement containing a combination of cherry, sour cherry and Japanese cherry extracts, an antioxidant complex, cysteine and D-biotin (in ratios according to the above exemplary embodiment) for 16 weeks.

Each participant served as his own control. The C.L.B.T.™ test was used to assess facial skin color (C), luminosity (L), brightness (B) and transparency (T) involved in skin radiance.

Facial skin imperfections were assessed by clinical assessment. Firmness was assessed by clinical assessment and cutimeter measurement. Participants also completed an auto-questionnaire to assess their satisfaction with various parameters involved in skin radiance and overall efficacy of the supplement.

"Red-pink" and "olive" skin color were significantly improved after supplementation (P<0.0001). Radiance was increased by 45% (P<0.0001), while brightness and transparency were not affected.

Facial skin imperfections were significantly reduced after taking the antioxidant-rich formula (total reduction: - 25.0%; P<0.0001).

In fact, dark circles, redness and blemishes are significantly reduced after oral treatment. Stiffness and elasticity have been shown to be improved. Participants were completely satisfied with the product (80%) and reported improvements in facial skin. Additionally, 60% of participants reported at the end of the study that they looked better.

Conclusion

From the results of the above clinical studies, it is evident that the proposed food supplement, according to the present invention, shows a new, unexpected, synergistic effect on skin pigmentation with clarification and evenness of complexion, based on balancing melanin production, improves the health and beauty of the skin.
