"Procédé de préparation d'esters d'acides 1,4-dihydro-pyridine-dicarboxyliques dissymétriques".
La présente invention concerne un procédé chimiquement particulier pour la préparation d'esters d'acides 1,4-dihydro-pyridine-dicarboxyliques dissymétriques connus.
Plusieurs procédés pour leur préparation sont déjà connus.
C'est ainsi que, par exemple, dans la demande de brevet de la République Fédérale d'Allemagne DE-AS 2.117.573, on décrit la préparation, à une étape, de dihydro-pyridines dissymétriques par réaction d'aldéhydes avec des acides céto-carboxyliques
et des esters d'acides énamino-carboxyliques. Dans
un procédé à deux étapes, on rencontre des inconvénients, car il est très difficile d'isoler les esters d'acides ylidène-B-cétocarboxyliques pouvant être préparés à partir d'aldéhydes et d'esters d'acides cétocarboxyliques et, très souvent, cette isolation n'est obtenue qu'avec de faibles rendements sous
forme pure.
Toutefois, on a constaté que la préparation de dihydro-pyridines en une étape conduisait à des impuretés qui ne peuvent être éliminées que difficilement par des procédés de purification.
En ce qui concerne l'utilisation des 1,4dihydro-pyridines dissymétriques comme médicaments, après comme avant, il est nécessaire que ces composés soient disponibles avec un haut degré de pureté.
C'est ainsi que, par exemple, lors de la préparation de la 4-(2'-nitrophényl)-2,6-diméthyl-3,5-dicarbéthoxy1,4-dihydro-pyridine conformément au brevet des EtatsUnis d'Amérique 3.485.847, par chromatographie sur couche mince, on a pu constater sept sous-produits.
En conséquence, l'invention concerne un procédé de préparation de 1,4-dihydro-pyridines dissymétriques de formule I :

en présence de quantités catalytiques de sels acétates d'amines à des températures de -10[deg.]C à 100[deg.]C.
Comme sels acétates d'amines, on mentionnera, de préférence : l'acétate de pipéridine ou d'une alkyl-pipéridine, l'acétate de morpholine ou d'une alkyl-morpholine, l'acétate de pipérazine ou d'une alkyl-pipérazine, de même que l'acétate de pyrrolidine ou d'une alkyl-pyrrolidine. On mentionnera, en particulier, l'acétate de pipéridine. Les groupes alkyle des catalyseurs comportent, de préférence, 1 à 4 atomes de carbone.
Comme solvants, on utilise, de préférence, des alcools aliphatiques tels que le méthanol, l'éthanol et/ou l'isopropanol. Les températures réactionnelles se situent, de préférence, entre 20 et 60[deg.]C.
Le catalyseur est ajouté, de préférence,
en quantités de 0,01 à 0,7 mole et, de manière particulièrement préférée, en quantités de 0,02 à 0,2 mole, en particulier, de 0,04 à 0,2 mole par mole des composés ylidène.
Par mole d'acide céto-carboxylique de formule IV, on peut utiliser, de préférence, 1 ou 2 moles, en particulier, 1 mole d'aldéhyde.
De préférence, dans la formule I .

La réaction du composé ylidène de formule
II ou III avec le composé d'énamine de formule IV
ou V a lieu à des températures de -10 à 130[deg.]C, de préférence, de 50 à 100[deg.]C.
Par mole du composé ylidène, on peut utiliser, de préférence, 1 à 1,5 mole et, de manière particulièrement préférée, 1 à 1,3 mole, en particulier,
1 à 1,2 mole du composé d'énamine.
Suivant une forme de réalisation particulière, le composé ylidène cristallin subsiste dans
le récipient réactionnel et il est mis à réagir directement avec le composé d'énamine.
Il est remarquablement surprenant que, suivant la réaction de l'invention, les esters d'acides ylidène-3-céto-carboxyliques soient formés,en présence des catalyseurs mentionnés, avec une grande pureté et un rendement excellent et qu'ils puissent être très bien isolés.
En outre, il est surprenant que, de la manière décrite, les composés de 1,4-dihydro-pyridine puissent être obtenus et isolés avec une aussi grande pureté. Ils ne contiennent aucun sous-produit (sans un procédé de purification complémentaire).
Le procédé suivant l'invention offre une série d'avantages.
C'est ainsi que le rendement est supérieur à celui des procédés connus et que le produit isolé ne doit être soumis à aucune étape supplémentaire
de purification.
L'invention sera expliquée par les exemples suivants .
Exemple 1
a) Ester d'acide ylidène-carboxylique.
Tout en agitant à la température ambiante,
à 325 ml d'isopropanol, on ajoute, dans l'ordre suivant, 80 g (0,5 mole) d'ester 2-méthoxyéthylique d'acide acéto-acétique, 75,5 g (0,5 mole) de 3-nitrobenzaldéhyde, 1,8 g (0,03 mole) d'acide acétique glacial et 2,5 g (0,03 mole) de pipéridine.
On chauffe à 40[deg.]C et on abandonne pendant
30 minutes à cette température.
Ensuite, on refroidit à 20[deg.]C et on poursuit l'agitation pendant 16 heures. Ensuite, on refroidit à 0[deg.]C et on poursuit l'agitation à cette température pendant une heure. On essore alors la solution surnageante et on lave le produit de cristallisation avec 166 ml d'isopropanol glacé.
On fait réagir immédiatement l'ester 2méthoxyéthylique d'acide 2-(3-nitrobenzylidène)-acétoacétique formé dans le même récipient, comme décrit sub b).
Si on isole l'ester 2-méthoxyéthylique d'acide 2-(3-nitrobenzylidène)-acéto-acétique et si
on le sèche, on obtient 132 g d'un produit de cristallisation brun clair (90% du rendement théorique) d'un point de fusion de 68-72[deg.]C.
Si, au lieu de 0,03 mole d'acétate de pipéridine, on utilise d'autres quantités de catalyseurs et si l'on fait varier les temps de réaction, on obtient les rendements suivants :

b) A l'ester 2-méthoxy-éthylique d'acide 2-(3-nitrobenzylidène)-acéto-acétique imprégné d'isopropanol et préparé suivant a), on ajoute 270 ml d'isopropanol et 64,4 g (0,45 mole) d'ester isopropylique d'acide 3-aminocrotonique. On chauffe à reflux
(83[deg.]C) et on abandonne à cette température pendant
24 heures.
Après refroidissement à 0[deg.]C, on isole le produit de cristallisation formé, on le lave avec
86 ml d'isopropanol et l'essore jusqu'à siccité.
On obtient 173,2 g d'ester isopropyl-(2méthoxyéthylique) d'acide 4-(3-nitrophényl)-2,6-diméthyl-1,4-dihydropyridine-3,5-dicarboxylique d'un point de fusion de 122-127[deg.]C (92%-de la théorie).
La chromatographie sur couche mince sur
des plaques préparées de gel de silice de "Merck" (éluant : chloroforme : acétone : éther de pétrole = 3:2:5) ne révèle aucun sous-produit visible.
Exemple 2
De la même manière qu'à l'exemple 1, à partir de l'ester éthylique d'acide 3-nitrobenzylidène-acéto-acétique préparé conformément à l'exemple la) et à partir de l'ester méthylique d'acide 3-aminocrotonique, après chauffage pendant 10 heures en solution éthanolique et après avoir ensuite laissé reposer pendant 16 heures à 20[deg.]C, on obtient l'ester 3-méthyl5-éthylique de l'acide 2,6-diméthyl-4-(3'-nitrophényl)-1,4-dihydro-pyridine-3,5-dicarboxylique que l'on recristallise dans du méthanol (point de fusion :
159[deg.]C ; rendement : 83% de la théorie).
Exemple 3
a) Ester d'acide ylidène-carboxylique.
Tout en agitant à la température ambiante,
à 325 ml d'isopropanol, on ajoute, dans l'ordre suivant : 58 g (0,5 mole) d'ester méthylique d'acide acéto-acétique, 82,5 g (1,5 mole) de 2,3-dichlorobenzaldéhyde, 1,8 g (0,03 mole) d'acide acétique glacial et 2,5 g (0,03 mole) de pipéridine.
On chauffe à 40[deg.]C et on abandonne à cette température pendant 30 minutes.
Ensuite, on refroidit à 20[deg.]C et on poursuit l'agitation pendant 16 heures.
Ensuite, on refroidit à 0[deg.]C et on poursuit l'agitation à cette température pendant une heure.
On fait réagir immédiatement l'ester méthylique d'acide 2-(2,3-dichlorobénzylidène)-acéto-acétique formé dans le même récipient, comme décrit sub b).
b) A l'ester méthylique d'acide 2-(2,3dichlorobenzyliàène)-acéto-acétique préparé suivant a), on ajoute 270 ml d'isopropanol et 58,9 g (0,45 mole) d'ester éthylique d'acide 3-aminocrotonique.
On chauffe à reflux (83[deg.]C) et on abandonne à cette température pendant 24 heures.
Après refroidissement à 0[deg.]C, on isole le produit de cristallisation formé, on le lave avec
86 ml d'isopropanol et on l'essore jusqu'à siccité.
On obtient 138 g d'ester méthyl-éthylique d'acide 4-(2,3-dichlorophényl)-2,6-diméthyl-1,4-dihydro-pyridine-3,5-dicarboxylique d'un point de fusion de 146[deg.]C (80% de la théorie).
"Process for the preparation of esters of asymmetric 1,4-dihydro-pyridine-dicarboxylic acids".
The present invention relates to a chemically specific process for the preparation of esters of known asymmetric 1,4-dihydro-pyridine-dicarboxylic acids.
Several methods for their preparation are already known.
Thus, for example, in patent application of the Federal Republic of Germany DE-AS 2,117,573, describes the preparation, in one step, of asymmetric dihydro-pyridines by reaction of aldehydes with acids keto-carboxylic
and esters of enamino-carboxylic acids. In
a two-step process, there are drawbacks, since it is very difficult to isolate the esters of ylidene-B-ketocarboxylic acids which can be prepared from aldehydes and esters of ketocarboxylic acids and, very often, this insulation is only obtained with low yields under
pure form.
However, it has been found that the preparation of dihydro-pyridines in one step leads to impurities which can only be removed with difficulty by purification methods.
With regard to the use of asymmetric 1,4dihydro-pyridines as medicaments, after as before, it is necessary that these compounds are available with a high degree of purity.
Thus, for example, during the preparation of 4- (2'-nitrophenyl) -2,6-dimethyl-3,5-dicarbethoxy1,4-dihydro-pyridine in accordance with United States patent 3,485 .847, by thin layer chromatography, seven by-products were found.
Consequently, the invention relates to a process for the preparation of asymmetric 1,4-dihydro-pyridines of formula I:

in the presence of catalytic amounts of acetate salts of amines at temperatures of -10 [deg.] C to 100 [deg.] C.
As acetate salts of amines, mention will preferably be made of: piperidine acetate or an alkyl-piperidine, morpholine acetate or an alkyl-morpholine, piperazine acetate or an alkyl- piperazine, as well as pyrrolidine acetate or an alkyl-pyrrolidine. Mention will be made, in particular, of piperidine acetate. The alkyl groups of the catalysts preferably have 1 to 4 carbon atoms.
As solvents, aliphatic alcohols such as methanol, ethanol and / or isopropanol are preferably used. The reaction temperatures are preferably between 20 and 60 [deg.] C.
The catalyst is preferably added
in amounts of 0.01 to 0.7 moles and, in a particularly preferred manner, in amounts of 0.02 to 0.2 moles, in particular, from 0.04 to 0.2 moles per mole of the ylidene compounds.
Per mole of keto-carboxylic acid of formula IV, it is possible to use, preferably, 1 or 2 moles, in particular, 1 mole of aldehyde.

The reaction of the ylidene compound of formula
II or III with the enamine compound of formula IV
or V takes place at temperatures from -10 to 130 [deg.] C, preferably from 50 to 100 [deg.] C.
Per mole of the ylidene compound, it is possible to use, preferably, 1 to 1.5 moles and, particularly preferably, 1 to 1.3 moles, in particular,
1 to 1.2 moles of the enamine compound.
According to a particular embodiment, the crystalline ylidene compound remains in
the reaction vessel and it is reacted directly with the enamine compound.
It is remarkably surprising that, according to the reaction of the invention, the esters of ylidene-3-keto-carboxylic acids are formed, in the presence of the catalysts mentioned, with high purity and excellent yield and that they can be very well insulated.
Furthermore, it is surprising that, as described, the 1,4-dihydro-pyridine compounds can be obtained and isolated with such high purity. They do not contain any by-products (without an additional purification process).
The process according to the invention offers a series of advantages.
This is how the yield is higher than that of known methods and that the isolated product must not be subjected to any additional step.

Example 1
a) Ester of ylidene-carboxylic acid.
While stirring at room temperature,
to 325 ml of isopropanol, 80 g (0.5 mole) of 2-methoxyethyl ester of acetoacetic acid, 75.5 g (0.5 mole) of 3-, are added in the following order nitrobenzaldehyde, 1.8 g (0.03 mole) of glacial acetic acid and 2.5 g (0.03 mole) of piperidine.
We heat to 40 [deg.] C and leave for
30 minutes at this temperature.
Then, it is cooled to 20 ° C. and the stirring is continued for 16 hours. Then, it is cooled to 0 [deg.] C and the stirring is continued at this temperature for one hour. The supernatant solution is then drained and the crystallization product is washed with 166 ml of ice-cold isopropanol.
The 2-methoxyethyl ester of 2- (3-nitrobenzylidene) -acetoacetic acid formed in the same container is immediately reacted, as described under b).
If the 2- (3-nitrobenzylidene) -aceto-acetic acid 2-methoxyethyl ester is isolated and if
dried, 132 g of a light brown crystallization product are obtained (90% of the theoretical yield) with a melting point of 68-72 [deg.] C.
If, instead of 0.03 mole of piperidine acetate, other quantities of catalysts are used and if the reaction times are varied, the following yields are obtained:

b) To the 2-methoxyethyl ester of 2- (3-nitrobenzylidene) -acetoacetic acid impregnated with isopropanol and prepared according to a), 270 ml of isopropanol and 64.4 g (0, 45 mol) of isopropyl ester of 3-aminocrotonic acid. We heat at reflux
(83 [deg.] C) and we leave at this temperature for
24 hours.
After cooling to 0 [deg.] C, the crystallization product formed is isolated, washed with
86 ml of isopropanol and wring it until dry.
173.2 g of isopropyl- (2-methoxyethyl) ester of 4- (3-nitrophenyl) -2,6-dimethyl-1,4-dihydropyridine-3,5-dicarboxylic acid with a melting point of 122 are obtained. -127 [deg.] C (92% -of theory).
Thin layer chromatography on
plates prepared from "Merck" silica gel (eluent: chloroform: acetone: petroleum ether = 3: 2: 5) does not reveal any visible by-product.
Example 2
In the same way as in Example 1, from the ethyl ester of 3-nitrobenzylidene-aceto-acetic acid prepared in accordance with Example 1a) and from the methyl ester of 3- aminocrotonic, after heating for 10 hours in ethanolic solution and after having then allowed to stand for 16 hours at 20 [deg.] C, there is obtained the 3-methyl5-ethyl ester of 2,6-dimethyl-4- acid ( 3'-nitrophenyl) -1,4-dihydro-pyridine-3,5-dicarboxylic which is recrystallized from methanol (melting point:
159 [deg.] C; yield: 83% of theory).
Example 3
a) Ester of ylidene-carboxylic acid.
While stirring at room temperature,
to 325 ml of isopropanol, 58 g (0.5 mole) of acetoacetic acid methyl ester, 82.5 g (1.5 mole) of 2,3- are added in the following order: dichlorobenzaldehyde, 1.8 g (0.03 mole) of glacial acetic acid and 2.5 g (0.03 mole) of piperidine.
It is heated to 40 [deg.] C and left at this temperature for 30 minutes.
Then, it is cooled to 20 ° C. and the stirring is continued for 16 hours.
Then, it is cooled to 0 [deg.] C and the stirring is continued at this temperature for one hour.
The 2- (2,3-dichlorobenzylidene) -aceto-acetic acid methyl ester formed in the same container is immediately reacted, as described under b).
b) To the methyl ester of 2- (2,3dichlorobenzylienene) -acetoacetic acid prepared according to a), 270 ml of isopropanol and 58.9 g (0.45 mole) of ethyl ester are added. 3-aminocrotonic acid.
It is heated to reflux (83 [deg.] C) and left at this temperature for 24 hours.
After cooling to 0 [deg.] C, the crystallization product formed is isolated, washed with
86 ml of isopropanol and it is wrung until dry.
