D'après les demandes de brevets n[deg.] 2.231.311 et n[deg.] 2.522.135 publiées en République fédérale d'Allemagne, on sait qu'en introduisant un groupe hydroperoxy -OOH dans la position quatre de la molécule des cytostatiques connus :

zaphosphorine (trofosfamide), 3-(2-chloréthyl-amino)-2-(2'chloréthylamino)-2-oxo-tétrahydro-2H-1,3,2-oxazaphosphorine
(ifosfamide), 3-(2-chloréthylamino)-2-(2'-méthanésulfonyléthylamino)-2-oxo-trétrahydro-2H-1,3,2-oxazaphosphorine
(sufosfamide) et d'autres cyclophosphamides d'une formule développée similaire, il est possible de créer des composés présentant des propriétés cytostatiques précieuses; mais, ces composés possèdent une stabilité tellement faible que leur transformation en préparations pharmaceutiques, tel que ceci est nécessaire dans la médecine des hommes, n'est pas possible. Le but de la présente invention consiste donc à créer des cyclophosphamides substitués dans la position 4 par un groupe hydroxy transformé ultérieurement et qui se distinguent par une haute activité cytostatique et une stabilité améliorée.

Le processus réactionnel peut être suivi, conformément au procédé de l'invention, à l'aide d'un chromatographie en couche mince. L'isolement des substances homogènes par la chromatographie en couches minces est établi par un procédé d'examen usuel de ces produits, en particulier par cristallisation ou purification chromatographique. La garantie de la structure se fait par le point de fusion, la chromatrographie en couche mince, l'analyse.élémentaire et/ou l'analyse spectrale aux infrarouges et par le spectre de résonance

Les composés utilisés comme matières premières dans le procédé conforme à l'invention sont largement connus, peuvent être employés à l'état cristallin ou comme produits bruts et peuvent être synthétisés comme suit d'une manière connue en soi.
Les 4-hydroxy-oxazaphosphorines sont obtenues par réduction des dérivés du 4-hydroperoxy (par exemple, A. Takamizawa et autres, J. Med. Chem., 18, 376, 1975). Les 4-méthoxy- ou 4éthoxy-oxazaphosphorines se forment par catalyse acide à partir des dérivés du 4-hydroxy dans du méthanol ou de l'éthanol ou dans des solvants inertes qui contiennent du méthanol et de l'éthanol. Les dérivés de l'hydroxy-urée sont fabriqués par la réaction des chlorures de l'acide carbamique ou des isocyanates substitués d'une manière correspondante avec une hydroxylamine ou des hydroxylamines N-monosubstituées. Pareillement, les esters de l'acide hydroxy-carbamique (hydroxycarbamate) . peuvent être produits par la réaction des esters de l'acide chlorocarbonique avec une hydroxylamine ou des hydroxulamines N-mono substituées.
Pour la synthèse des esters de l'acide hydroxy-urée-carbamique ou N-hydroxy-carbamique avec un groupe hydroxy, carboxy ou acide sulfonique supplémentaire, une voie de synthèse avec un groupe de protection est recommandable.
Les oxymes de la formule IV sont préparables selon diverses voies de synthèse. Ainsi, elles peuvent être obtenues par la réaction des 4-hydroxy-oxazaphosphorinès correspondantes avec une hydroxylamine ou des sels d'hydroxylamine. La valeur pH doit être maintenue autant que possible entre 2 et
7. Les isomères racémiques cis et trans(cis = 2R, 4R / 2S, 4S; trans = 2R, 4S / 2S; 4R) peuvent être fabriqués en partant des 4-carbamoyloxy-oxazaphosphorines conformes à l'invention. Le rattachement est en concordance avec les règles de la nomenclature IUPAC et avec la littérature sur les dérivés correspondants- des oxazaphosphorines. La forme cis ou trans peut être établie également avec précision selon le choix des conditions de la réaction. Pharmacologiquement,
les isomères ne montrent aucune différence importante.
Les composés conformes à l'invention se caractérisent par des propriétés remarquables, chimiothérapeutiquement précieuses. En comparaison de la substance de référence connue antérieurement, soit le cyclophosphamide, elles possèdent une forte activité chimiothérapeutique cancérotoxique approximativement égale sur des tumeurs de transplantation expérimentales des rats. En soltuion aqueuse, elles agissent directement comme alcoylant et ont in vitro une haute zytotoxicité, tandis que le cyclophosphamide exige une activation enzymatique et en pratique n'agit pas zytotoxiquement in vitro. La toxicité aiguë des composés conformes à l'invention est environ 4 fois plus petite que celle de la substance
de référence, le cyclophosphamide; l'ampleur thérapeutique est par conséquent nettement augmentée. Pareillement, en ce qui concerne les actions secondaires organotoxiques telles que la dépression des leucocytes et la suppression de l'immu-nisation, les composés conformes à l'invention présentent des avantages distincts par rapport à la substance de référence, le cyclophosphamide.
Les composés conformes à l'invention conviennent au traitement de tumeurs malignes et de maladies malignes semblables comme la leucémie des humains. Dès lors, on utilise des quantités de l'ordre de 0,01 à 100 mg/kg. les préparations pharmaceutiques ainsi mises en oeuvre sont fabriquées selon des méthodes usuelles en utilisant les supports et les additifs habituels.
Les exemples suivants servent à expliquer en détail la présente invention, sans cependant la limiter à ceux-ci.
Exemple 1

15 g (54 millimoles) de 4-hydroxycyclophosphamide
(c'est-à-dire le 2-(bis-chloréthyl)-amino)-4-hydroxy-tétrahydro-2H-,1,3,2-oxazaphosphorine-2-oxyde) et 4,4 g (58 millimoles) d'hydroxy-urée sont dissous dans 70 ml de diméthylformamide, acidifiés avec de " acide trichloracétique (pH 3-4)

phosphorine-2-oxyde). Ensuite, 1,2 g de 4-méthoxycyclophosphamide et 304 mg d'hydroxyurée sont dissous dans 3 ml de diméthylformamide et conservés au réfrigérateur pendant 20 h à -25[deg.]C. Le magma cristallin est dilué avec 3 ml d'ester acétique, aspiré, lavé, séché et recristallisé dans du méthanol.
Rendement : 670 mg (50% de la théorie) du même produit que
celui de l'exemple 1.
Exemple 3

Exemple 4

20 g (50 millimoles) de 4-hydroxytrofosfamide (c'est-àdire 3-(2-chloréthyl)-2-(bis-(2-chloréthyl)-amino)-4-hydroxy-tétrahydro-2H-1,3,2-oxazaphosphorine-2-oxyde- et 5,3 g
(70 millimoles) d'hydroxy-urée sont dissous dans 100 ml de diméthylformamide et refroidis à. -15[deg.]C. Ensuite, on acidifie avec de l'acide trichloracétique (pH 3-4) et on agite pendant 5 h à -15[deg.]C. Après abandon pendant une nuit à. 0[deg.]C, on dilue la solution réactionnelle avec deux fois sa quantité d'eau.
Puis, on extrait 4 fois en secouant avec chaque fois 300 ml d'ester acétique-méthanol (10/1), on lave 2 fois à l'eau les phases purifiées d'ester acétique, on sèche sur du sulfate de sodium et on concentre sous vide 22 g d'huile. Après absorption dans de l'ester acétique-méthanol, 4,2 g se séparent à l'état cristallin (point de fusion : 106-110[deg.]C). La liqueur mère est séparée par chromatographie sur colonne sur du gel de silice avec du chloroforme-méthanol (10/1) et recristallisée conjointement avec le premier produit de cristallisation dans de l'ester acétique-méthanol.
Rendement : 7,0 g (35% de la théorie). Point de fusion :
115-116[deg.]C (décomposition).
Exemple 5

A 900 mg (3,25 millimoles) de 4-hydroxycyclophosphamide dans 1 ml de chlorure de méthylène, on ajoute 540 mg (3,25 millimoles) de 3-benzyl-1-hydroxy-urée dans 40 ml d'acétone et une quantité catalytique d'acide trichloracétique. Le mélange est conservé pendant une nuit à -25[deg.]C, les cristaux sont ensuite aspirés, lavés à l'acétone et à l'éther et recristallisés dans de l'ester acétique.

uréido-acétate. Point de fusion : 113-120[deg.]C.
22,4 g (0,1 mole) de benzyl-3-hydroxy-uréido-acétate dans
300 ml de méthanol sont additionnés de 5 g de charbon palladié et hydrogénés dans un récipient pour hydrogénation catalytique. L'absorption correspondante d'hydrogène est achevée après 20 minutes environ. Le catalyseur est aspiré, le filtrat est concentré sous vide et le résidu solide est recristallisé dans du dioxane.
Rendement : 9,8 g (73% de la théorie) d'acide 3-hydroxy-uréi-
do-acétique. Point de fusion : 135[deg.]C.
b) 2,4 g (18 millimoles) d'acide 3-hydroxy-uréido-acétïque dans 10 ml d'eau et 25 ml d'acétone sont additionnés de 6,1 g (2 millimoles) de 4-hydroxycyclophosphamide, puis <EMI ID=52.1>
ajoute 25 ml d'acétone et 1,8 g (18 millimoles) de cyclohexylamine dans 10 ml d'acétone, on aspire après abandon de 2 h et on recristallise dans de l'acétone avec un peu de méthanol.
Rendement : 3,1 g (44% de la théorie). Point de fusion

La valeur pH est contrôlée sous agitation à 5-10[deg.]C avec un pH-mètre, est maintenue entre 4,5 et 5,5 avec de l'acide sulfurique 2 normal
et une solution concentrée de thiosulfate de sodium est ajoutée goutte à goutte jusqu'à ce que la valeur pH n'augmente plus. On agite de nouveau pendant une demi-heure à 10[deg.]C environ, on ajoute goutte à goutte une solution aqueuse de
950 mg de chlorhydrate d'hydroxylamine, on maintient la valeur pH à 5 avec une lessive de soude 2 normale, on laisse au repos pendant une nuit à 5[deg.]C le mélange réactionnel, on extrait quatre fois avec chaque fois 50 ml d'ester acétique et on concentre sous vide à 30[deg.]C les extraits organiques après séchage sur du sulfate de sodium. Le résidu est absorbé dans du chlorure de méthylène et les cristaux sont aspirés après 1 jour.
Rendement : 3,4 g (85% de la théorie). Point de fusion :

Exemples d'utilisations pharmaceutiques.
Exemple 72 (comprimés revêtus d'une pellicule et résistant
au suc gastrique)-
100 g du composé correspondant à l'exemple 5 sont additionnés de 7,0 g d'Aérosil par l'intermédiaire d'un tamis, puis sont mélangés. A ce mélange, on ajoute 76,0 g d'Avicel pH 105, 10 g d'amidon de maïs et 7,0 g d'acide stéarique et on mélange jusqu'à la répartition homogène de tous les constituants. Cette masse est pressée, comme on le sait, en noyaux de 200 mg comprenant 100 mg de constituant actif.
Ces noyaux sont revêtus d'une pellicule résistant au suc gastrique, par exemple, à base de dérivés de la cellulose ou d'un générateur de pellicules entièrement synthétique sous la forme d'une solution organique ou d'une dispersion aqueuse qui peut contenir des additifs tels que des plastifiants, des colorants, des aromatisants ou des antimousses.
Exemple 73 (gélules de gélatine résistant au suc gastrique)
250 g du composé correspondant à l'exemple 5 sont additionnés de 7,5 g d'Aérosil par l'intermédiaire d'un tamis, puis sont mélangés. A ce mélange, on ajoute 40 g de lactose et 2,5 g de stéarate de magnésium et on mélange jusqu'à la répartition homogène de tous les constituants.
Cette masse est prélevée en doses individuelles de 300 mg dans des gélules de gélatine. Une gélule contient 250 mg de constituant actif. Conformément à l'exemple 1, les gélules de gélatine peuvent être munies d'un revêtement pelliculaire résistant au suc gastrique.

The aim of the present invention therefore consists in creating cyclophosphamides substituted in position 4 by a hydroxy group which is subsequently transformed and which are distinguished by high cytostatic activity and improved stability.

The 4-hydroxy-oxazaphosphorines are obtained by reduction of the 4-hydroperoxy derivatives (for example, A. Takamizawa and others, J. Med. Chem., 18, 376, 1975). 4-methoxy- or 4ethoxy-oxazaphosphorines are formed by acid catalysis from 4-hydroxy derivatives in methanol or ethanol or in inert solvents which contain methanol and ethanol. Hydroxyurea derivatives are produced by the reaction of carbamic acid chlorides or correspondingly substituted isocyanates with hydroxylamine or N-monosubstituted hydroxylamines. Similarly, the esters of hydroxycarbamic acid (hydroxycarbamate). can be produced by the reaction of esters of chlorocarbonic acid with a hydroxylamine or N-mono substituted hydroxulamines.
For the synthesis of hydroxy-urea-carbamic or N-hydroxy-carbamic acid esters with an additional hydroxy, carboxy or sulfonic acid group, a synthetic route with a protecting group is recommended.

Compared to the previously known reference substance, cyclophosphamide, they have a roughly equal carcinotoxic chemotherapeutic activity on experimental transplant tumors in rats. In aqueous solution, they act directly as an alkylating agent and have a high zytotoxicity in vitro, while cyclophosphamide requires an enzymatic activation and in practice does not act zytotoxically in vitro. The acute toxicity of the compounds according to the invention is approximately 4 times lower than that of the substance
cyclophosphamide; the therapeutic scope is therefore significantly increased. Similarly, as regards secondary organotoxic actions such as leukocyte depression and the suppression of immunization, the compounds according to the invention have distinct advantages compared to the reference substance, cyclophosphamide.
The compounds according to the invention are suitable for the treatment of malignant tumors and similar malignant diseases such as human leukemia. Therefore, quantities of the order of 0.01 to 100 mg / kg are used. the pharmaceutical preparations thus used are manufactured according to usual methods using the usual supports and additives.
