L'invention concerne un procédé de préparation du dérivé 5'-0- triphosphoryl-5- (3-allylaminobiotine) uridine (en abréviation Bio-UTP) et du dérivé 5'-0-triphosphoryl-2'-désoxy-5-93-allyl- aminobiotine) uridine, (en abréviation Bio-dl1I'P) obtenus par un procédé de synthèse simplifié.
De tels dérivés de la biotine et de l'uridine triphosphate (UTP) ou de la désoxyuridine triphosphate (dUTP) sont représentés par la formule générale suivante.

Avec R et R'= OH pour le dérivé de l'uridine et R = OH et R' pour le dérivé de la désoxyuridine.
De tels analogues de nucléotides, (Bio-UTP et Bio-dUTP) qui peuvent être utilisés notamment ccmne marqueurs par incotporation dans des polynucléotides, trouvent de nombreuses applications dans les recherches biomédicales et de recombinaison génétique.
Le procédé de synthèse du Bio-UrP et du Bio-dUTP décrit par P. R.
Langer, A. A. Waldrop. et D. C. Ward (Proc. Natl. Acad. Sci. U. S. A.
78, (1981) 6633) fait appel à trois réactions successives.
Dans une première phase, l'uridine triphosphate (UTP) ou la désoxyuridine triphosphate (dUTP) est mis en réaction avec de l'acétate mercurique pour obtenir les dérivés respectifs 5-mercurique de l'UTP ou de la dUTP.

Dans une deuxième phase, les dérivés mercuriques de l'UTP ou de la DUTP sont mis en réaction avec l'allylamine en présence d'un catalyseur pour obtenir les dérivés 5-allylarnino-UTP ou dUTP.

Ces dérivés doivent être purifiés en deux étapes, d'abord par chromatographie sur colonne DEAE Sephadex, ensuite par chramatographie liquide à haute performance.
Dans une troisième étape, les produits de réaction de la deuxième phase sont mis en réaction avec l'ester N-hydroxysuccimidique de la biotine pour obtenir respectivement le Bio-UTP et le Bio-dUTP.
Le produit final est enfin purifié par chromatographie sur colonne de DEAE Sephadex.
Le schéma de synthèse décrit ci-dessus nécessite 3 reactions et 3 étapes de purification mettant en jeu ure fonction triphosphate particulièrement labile. Il a des désavantages d'être long, laborieux et sujet à dégradation de par l'instabilité relative des intermédiaires mis en jeu. De plus, les caractéristiques structurales du produit final et du produit de la seconde phase dans le procédé décrit, ne permettent pas une interprétation immédiate en spectrophotanétrie UV du résultat de la réaction car ces produits ont des spectres UV semblables.
La présente invention a pour objet de remédier à ces divers inconvénients. Suivant l'invention, on prépare le dérivé 5'-0-tripos- phoryl-5 (3-allylaminobiotine) uridine (Bio-UTP) et le dérivé 5' -0- triphosphoryl -2'-désoxy-5- (3-allylaminobiotine) uridine (Bio-dUTP) par la réaction, en présence d'un catalyseur, d'un dérivé 5-mercurique de l'uridine triphosphate ou de la 21 -désoxyuridine triphosphate de formule générale (I).

L'exemple donné se réfère à l'obtention du Bio-DUTP, Préparation du dérivé 5-mercurique de la dut On agite, par exemple, 4 h à 500C une solution aqueuse d'acétate de

sodium 0. 1 M, pH 6, de désoxyuridine trip 10 mM, d'acétate mercuri- que 50 mM. On ajoute du chlorurede lithium jusqu'à une concentration de 0.17 M. La solution est extraite avec de l'acétate d'éthyle. La phase acqueuse est précipitée avec de l'éthanol. Le précipité est lavé avec de l'éthanol et avec de l'éther diéthylique.
Préparation de l'allylaminobiotine.
A une solution de N-hydroxysuccinimidebiotine (45mg, 0,13 mmol) dans la dimethylformamide anhydre (0,5 ml), on ajoute l'allylamine (15 mg, 0,26 mol). La solution est agitée pendant 2h à 20oC, puis concentrée sous vide. Le résidu est recristallisé dans l'isopropa- nol-éther anhydres.
On a obtenu 35 mg de cristaux incolores.

Préparation du 5'-0-triphosphoryl (-5-allylaminobiotine) désoxyuridine A une solution d'allylaminobiotine (2 mg, 0, 01 mmol) dans l'acétate de sodium acqueux (0,1 M, pH 5, 0,8 ml) on ajoute le dérivé 5-mercurique de la 2'-désoxyuridine triphosphate (1) (5 mg, 0,01 mmol) et ensuite le tetrachloro palladate de potassium (2,3 mg, 0,01 mmol) en suspension dans 50 ul d'eau. Le mélange est agité 11 h à 20 C, filtré sur milipore ou centrifugé. Le surnageant est déposé sur une colonne de Sephadex DEAE A25 de 2,5 ml et élué dans un gradient linéaire 0,1 M à 0,9 M de bicarbonate de triéthylammonium (TEAB, pH 7,5).
Le produit éluant en fin de gradient présente le spectre UV attendu (gk max = 240 nm. min = 265 nm, max = 275 nm dans l'eau) et une pureté de # 90% d'après analyse par HPLC.
Le dérivé ainsi obtenu a été introduit dans du DNA correspondant au gène de globine par nicktranslation. Le DNA ainsi substitué réagit avec l'avidine fixée fixée sur colonne d'ultragel avec la même efficacité que le produit obtenu par la méthode antérieurement connue, décrite dans l'article de P. R. Langer et al. (Proc. Nat. Acad. Sci.
USA 78 (1981) 6633).
La même procédure de synthèse peut être utilisée pour l'obtention du Bio-UTP.
Comme avantages du procédé nouveau de synthèse faisant l'objet de la présente invention citons : - la rapidité et la simplicité ; - la diminution des risques de dégradation des dérivés triphospohorylés qui ne sont engagés qu'une seule fois dans une réaction et une fois dans une étape de purification ; - l'obtention, après mercuration en une étape, d'un produit final dont la structure permet une purification aisée en une seule étape par chromatographie sur colonne de Sephadex - l'identification facile du produit final car sa structure lectrsnique caractéristique se différencie aisément par spectrophotométrie UV de celle du produit de départ.

Les dérivés 5'-0-triphosphoryl-5- (3-allylaminobiotine) uridine et 5' -0- triphosphoryl -2'-désoxy-5-93-allylaminobiotine) uridine obtenus sont utilisés, notamment carme marqueurs internes des acides nucléiques.
Des possibilités d'applications sont, par exemple, l'isolement ou la mise en évidence d'un PNA. d'une espèce bien définie dans une population hétérogène de RNN, ou la mise en évidence de séquences de RNA ou de DNA par hybridation in situ ou après transfert de ces molécules sur filtre de nitrocellulose (Southern blot ou Northern blot), ou encore l'isolement de RNN spécifiques de protéines induites.

The invention relates to a process for the preparation of the 5'-0-triphosphoryl-5- (3-allylaminobiotin) uridine derivative (in abbreviation Bio-UTP) and of the 5'-0-triphosphoryl-2'-deoxy-5-93 derivative -allyl- aminobiotin) uridine, (abbreviated to Bio-dl1I'P) obtained by a simplified synthesis process.
Such derivatives of biotin and uridine triphosphate (UTP) or deoxyuridine triphosphate (dUTP) are represented by the following general formula.

In a second phase, the mercuric derivatives of UTP or DUTP are reacted with allylamine in the presence of a catalyst to obtain the 5-allylarnino-UTP or dUTP derivatives.

These derivatives must be purified in two stages, first by chromatography on a DEAE Sephadex column, then by high performance liquid chramatography.
In a third step, the reaction products of the second phase are reacted with the N-hydroxysuccimidic ester of biotin to obtain Bio-UTP and Bio-dUTP respectively.
The final product is finally purified by chromatography on a DEAE Sephadex column.
The synthesis scheme described above requires 3 reactions and 3 purification steps involving a particularly labile triphosphate function. It has the disadvantages of being long, laborious and subject to degradation due to the relative instability of the intermediaries involved. In addition, the structural characteristics of the final product and of the product of the second phase in the process described, do not allow an immediate interpretation in UV spectrophotometry of the reaction result because these products have similar UV spectra.
The object of the present invention is to remedy these various drawbacks. According to the invention, the 5'-0-triposphoryl-5 (3-allylaminobiotin) uridine derivative (Bio-UTP) and the 5 '-0- triphosphoryl -2'-deoxy-5- (3- allylaminobiotin) uridine (Bio-dUTP) by the reaction, in the presence of a catalyst, of a 5-mercuric derivative of uridine triphosphate or of 21-deoxyuridine triphosphate of general formula (I).

Preparation of 5'-0-triphosphoryl (-5-allylaminobiotin) deoxyuridine To a solution of allylaminobiotin (2 mg, 0.01 mmol) in aqueous sodium acetate (0.1 M, pH 5, 0.8 ml ) add the 5-mercuric derivative of 2'-deoxyuridine triphosphate (1) (5 mg, 0.01 mmol) and then potassium tetrachloro palladate (2.3 mg, 0.01 mmol) suspended in 50 μl of water. The mixture is stirred for 11 h at 20 ° C., filtered through a milipore or centrifuged. The supernatant is deposited on a 2.5 ml Sephadex DEAE A25 column and eluted in a linear gradient 0.1 M to 0.9 M triethylammonium bicarbonate (TEAB, pH 7.5).
The product eluting at the end of the gradient has the expected UV spectrum (gk max = 240 nm. Min = 265 nm, max = 275 nm in water) and a purity of # 90% according to analysis by HPLC.
The derivative thus obtained was introduced into DNA corresponding to the globin gene by nicktranslation. As advantages of the new synthesis process which is the subject of the present invention, let us cite: - speed and simplicity; the reduction in the risks of degradation of the triphospohorylated derivatives which are used only once in a reaction and once in a purification step; - obtaining, after mercury in one step, a final product whose structure allows easy purification in a single step by chromatography on a Sephadex column - easy identification of the final product because its characteristic lectrsnique structure is easily differentiated by UV spectrophotometry of that of the starting product.
