Priorité des deux demandes de brevet déposées au Japon, respectivement, le 14 novembre 1978 sous le n[deg.] 139325/78, et le
21 novembre 1978 sous le n[deg.] 142813/78, toutes deux au nom de la Société susdite.
La présente invention concerne de nouveaux dérivés d'imidazole ayant une excellente efficacité du point de vue médical, la préparation de ces dérivés, ainsi que des médicaments contenant ces dérivés. Plus particulièrement, l'invention concerne des dérivés d'imidazole répondant à la formule générale

Les dérivés d'imidazole suivant la présente invention sont de nouveaux composés inconnus jusqu'à présent qui n'ont qu'une très faible toxicité, qui manifestent une excellente efficacité antidépressive et qui sont utiles comme remède ou à titre prophylactique pour la dépression ou les états dépressifs. En tant qu'agents antidépressifs, on a utilisé cliniquement des composés d'une structure tricyclique tels que l'Imipramine, l'Amitriptyline et analogues. Les composés de la présente invention ont une structure totalement différente de celle de ces composés.
En conséquence, un objet de la présente invention est de fournir de nouveaux dérivés d'imidazole qui sont très utiles comme agents antidépressifs et qui offrent de hautes garanties de sécurité.
Un autre objet de la présente invention est de fournir un procédé de préparation de nouveaux dérivés d'imidazole du type mentionné ci-dessus.
Un autre objet encore de l'invention est de fournir un agent antidépressif comprenant de nouveaux dérivés d'imidazole.

On utilise chaque composé d'essai sous forme de son chlorhydrate.
Procédés expérimentaux
(1) Activité anti-réserpine :
On examine l'antagonisme vis-à-vis de l'hypothermie provoquée par la réserpine conformément au procédé de B.M. Askew ("Life Science", 4, 725-730, 1963).
Comme animaux d'essai, on utilise des souris mâles ddy pesant 19 à 25 g. On injecte de la réserpine par voie intrapéritonéale en une dose de 2,5 mg/kg et, 18 heures après l'injection, on administre le composé d'essai par voie orale et l'on examine le changement de la température du corps
à mesure que le temps s'écoule en utilisant un thermomètre du type à Thermistor (modèle "MGA III" fabriqué par "Shibaura Denshi").
On examine l'effet exercé par chaque composé à 2-4 niveaux de dosage.
On utilise chaque composé d'essai sous forme d'une solution aqueuse. On traite l'animal témoin avec une solution de sérum physiologique. Pour chaque dose, on utilise quatre animaux dont la température du corps se situe entre 24 et 25[deg.]C et ce, 18 heures après l'injection de la réserpine. On effectue l'expérience dans un local dont la température se situe entre
23 et 25[deg.]C.
(2) Influence sur la motilité spontanée :
On examine la motilité spontanée conformément au

Dans une boîte d'observation noire en résine acrylique, on place un groupe de trois souris mâles ddy pesant 20 à 25 g puis,

Suède), on mesure et enregistre la quantité de motilité spontanée au cours d'une période de 60 minutes.
60 minutes avant le début de la mesure de la motilité spontanée, on administre le composé d'essai par voie orale en une dose de 40 mg/kg. On traite l'animal témoin avec une solution de sérum physiologique. Pour chaque niveau de dosage, on utilise cinq groupes (15 souris).
(3) Toxicité aiguë :
On administre le composé d'essai en une dose de
100 ou de 400 mg/kg à des souris afin d'examiner l'action létale. Pour chaque niveau de dosage, on utilise cinq souris. On mesure la dose létale 24 heures après administration orale.
Résultats expérimentaux
Le tableau A ci-après donne les résultats de l'expérience effectuée en vue de déterminer l'activité antiréserpine, l'influence sur la motilité spontanée et la toxicité aiguë.

Comme on le constate clairement d'après le tableau A, les composés de l'invention exercent une puissante activité anti-réserpine et ont une très faible toxicité.
De plus, on a étudié l'effet des composés de la présente invention (composés H et I) sur la teneur en dopamine et ses métabolites dans le cerveau de la souris. Suite à cette étude, on a trouvé que ces composés réduisaient le renouvellement en dopamine dans le cerveau, ce qui signifie que les composés de l'invention sont des antagonistes de la dépression provoquée par la réserpine d'une manière totalement différente de celle des antidépressifs tricycliques connus.
Dès lors, les composés de la présente invention exercent une puissante activité anti-réserpine exceptionnelle et ils offrent un haut degré de sécurité en raison de leur faible toxicité, si bien qu'ils constituent des produits excellents en tant que remèdes ou pour le traitement prophylactique de la dépression ou des états dépressifs. En outre, on considère que les composés de l'invention sont chimiquement et pharmacologiquement différents des antidépressifs existants
(Imipramine, Amitriptyline et analogues). En d'autres mots, les composés de la présente invention sont des médicaments
d'un type exceptionnel et totalement différent des antidépressifs connus.
Les composés suivant la présente invention sont administrés en doses se situant entre 10 et 1.000 mg/jour, de préférence, entre 30 et 300 mg/jour, pour le traitement clinique de patients souffrant de dépression ou de patients souffrant de maladies internes et se plaignant de l'état dépressif.
Les composés suivant la présente invention peuvent se présenter sous diverses préparations administrées par des procédés facultatifs. En conséquence, la présente invention englobe une composition pharmaceutique comprenant au moins un composé de l'invention approprié comme médicament pour le corps humain. On peut préparer cette composition pharmaceutique en utilisant un support ou un excipient pharmaceutique facultatif
et on peut l'administrer suivant la méthode habituelle.
Il est préférable d'administrer la composition pharmaceutique sous une forme appropriée pour l'absorption
par les canaux digestifs. On peut mentionner un comprimé ou
une capsule comme unité pour l'administration par voie orale,
ce comprimé ou cette capsule pouvant contenir des agents liants tels qu'un sirop, la gomme arabique, la gélatine, le sorbitol,
la gomme adragante et l'alcool polyvinylique, des excipients
tels que le lactose, l'amidon de maïs, le phosphate de calcium, le sorbitol et la glycine, des lubrifiants tels que le stéarate de magnésium, le talc, le polyéthylène-glycol et la silice, des agents désintégrants tels que la fécule de pomme de terre,
des agents mouillants tels que le lauryl-sulfate de sodium,
ainsi que d'autres adjuvants classiques. Les comprimés peuvent être enrobés suivant les méthodes connues et habituellement adoptées dans la technique. Parmi les préparations liquides
pour administration par voie orale, il y a les suspensions et
les solutions aqueuses et huileuses, les sirops, les élixirs
et analogues. De plus, on peut utiliser un produit sec que
l'on redissout dans l'eau ou dans un autre véhicule approprié avant l'administration. Ces préparations liquides peuvent contenir des additifs habituellement utilisés dans ce domaine, par exemple, des agents de mise en suspension tels qu'un sirop
de sorbitol, la méthyl-cellulose, le sirop de glucose/sucre,
la gélatine, l'hydroxyéthyl-cellulose, la carboxyméthyl-cellulose, le gel de stéarate d'aluminium et une huile comestible hydrogénée, des agents émulsionnants tels que la lécithine, le monooléate de sorbitol et la gomme arabique, des véhicules non aqueux tels que l'huile d'amande, l'huile de coco fractionnée, un ester huileux, le propylène-glycol et l'alcool éthylique,
de même que des agents antiseptiques tels que le p-hydroxybenzoate de méthyle, le p-hydroxybenzoate de propyle et l'acide sorbique.
Une composition pour injection se présente sous forme d'une ampoule ou d'une fiole contenant un dosage unitaire en vue d'une administration avec un agent antiseptique. Cette composition pour injection peut se présenter sous forme d'une suspension, d'une solution ou d'une émulsion dans un véhicule huileux ou aqueux pouvant contenir un agent de mise en suspension, un stabilisant et/ou un agent dispersant. L'ingrédient actif peut être une poudre que l'on redissout dans un véhicule approprié tel que l'eau stérile exempte de substance pyrogène et ce, avant l'administration.
La présente invention sera illustrée plus particulièrement par les exemples suivants, mais il est évidemment entendu qu'elle n'y est nullement limitée.
Exemple 1
Synthèse de l-(3,4-diméthylphényl)-2-méthylimidazole :
Tout en agitant, pendant 40 minutes, on chauffe, dans un bain d'huile porté à une température externe de 180[deg.]C,
un mélange de 2 g d'acide 2-méthyloxazole-4-carboxylique et de

Ensuite, au mélange réactionnel, on ajoute 0,6 g de chlorure de méthane-sulfonyle et l'on chauffe le mélange à reflux tout en agitant pendant 3 heures. Au terme de la réaction, on refroidit naturellement le mélange réactionnel et on élimine le solvant par distillation sous pression réduite.
On ajoute de l'eau au résidu et on alcalinise le liquide obtenu par addition d'une solution aqueuse de carbonate de sodium.

The imidazole derivatives according to the present invention are new compounds hitherto unknown which have only very low toxicity, which exhibit excellent antidepressant efficacy and which are useful as a remedy or as a prophylactic for depression or depressive states. As antidepressant agents, compounds of a tricyclic structure such as Imipramine, Amitriptyline and the like have been used clinically. The compounds of the present invention have a completely different structure from that of these compounds.
It is therefore an object of the present invention to provide new imidazole derivatives which are very useful as antidepressant agents and which offer high guarantees of safety.
Another object of the present invention is to provide a process for the preparation of new imidazole derivatives of the type mentioned above.
Yet another object of the invention is to provide an antidepressant agent comprising new imidazole derivatives.
