Le brevet principal était plus particulièrement illustré par l'obtention d'une souche de virus résistante
aux inhibiteurs sériques préalablement obtenue par recombinaison de la souche de virus grippal A/England/42/72 avec la souche de virus grippal A/PR8/34 et l'utilisation ce la souche rendue résistante dans la préparation de vaccins vivants.
On sait que, presque chaque année, le sérotype des souches de virus grippal de type A circulant dans le monde diffère légèrement du sérotype des souches observées précédemment. Cette modification antigénique crée des problèmes particuliers pour l'immunisation contre le virus grippal
de type A. En effet, pour obtenir l'efficacité maximale de l'antigène vaccinal, celui-ci doit être le plus proche possible de celui du virus dominant et, de ce fait, les vaccins contre la grippe doivent être périodiquement adaptés de façon à conférer une protection maximale contre ces nouvelles souches.
La présente invention concerne un tel perfectionnement. En soumettant, suivant le procédé décrit dans le brevet <EMI ID=2.1>

Le présent perfectionnement concerne également un procédé pour préparer le dit vaccin caractérisé en ce qu'on incube dans la cavité allantoïdienne d'oeufs embryonnés de poule la souche de virus grippal P/76/ 7 résistante aux inhibiteurs sériques, l'incubation étant effectuée pendant un temps suffisamment long pour permettre la production d'une grande quantité du dit virus et qu'on récolte le matériel viral résultant qui est ensuite, de préférence, lyophilisé.
Pour administrer le vaccin.de l'invention, celui-ci, qui est de préférence conservé sous forme lyophilisée,
est alors reconstitué extemporanément par addition d'eau ou de tout autre diluant ou composition connu dans la technique pour l'obtention de préparations nasales telles que gouttes ou aérosols et le vaccin est inoculé dans les narines. Eventuellement,l'administration est répétée environ 1 à 2 semaines après la première inoculation.

On utilise comme matériel viral de départ une souche sensible aux inhibiteurs sériques obtenue par recombinaison de la souche A/PR8/34.avec la souche A/Victoria/3/75, soumise à un passage en dilution limite et qui a reçu dans notre collection la dénomination RIT 4057. On mélange différentes dilutions

Afin de tester la résistance aux inhibiteurs sériques de cobaye et de cheval, on mélange des séries de doubles dilutions des séra préalablement chauffés à 75[deg.]C pendant 10 minutes
à 4 unités hémagglutinantes de la souche parentale ou de la souche modifiée. Après une heure d'incubation à température ambiante, on ajoute des érythrocytes de poulets et on enregistre les dilutions de sérum provoquant une inhibition de l'hémagglutination.
Les résultats sont indiqués au Tableau I.

EXEMPLE 2
Préparation du vaccin
On utilise le virus du dernier passage du procédé décrit dans l'exemple I comme inoculum pour la production
du lot de base pour la fabrication de vaccin.
On récolte et on regroupe les liquides allantoïdiens. on en vérifie la stérilité et l'inocuité et on les mélange
à de la peptone de façon à atteindre une concentration finale de peptone de 5%.
Dans des flacons de 3 ml on répartit la suspension virale en fractions de 0,5 ml de façon à obtenir des doses

lyophilise ensuite le produit et on obture hermétiquement les flacons.
Le. vaccin est reconstitué au moment de l'inoculation par addition de 0,5ml d'un diluant qui peut être par exemple de l'eau distillée, du sérum physiologique ou une solution

EXEMPLE 3
Essais cliniaues

obtenu par la méthode décrite dans l'exemple 2 ont été reconstituées par addition de 0,5ml d'une solution aqueuse stérile de saccharose à 5% et inoculées à 15 volontaires sains (âge moyen: 22 ans) à raison de 5 gouttes par narine.
Une deuxième dose a été inoculée à 8 des volontaires 7 jours après la première inoculation.
Tous les volontaires ont été examinés quotidiennement pour détecter d'éventuels symptômes grippaux.
On a prélevé des échantillons de sang avant et 2 à 3 semaines après inoculation afin de déterminer le nombre de séroconversions et la moyenne géométrique du titre d'inhibition de l'hémagglutination (MGT).
Les résultats des essais cliniques sont repris dans
les tableaux II et III ci-après:

In order to test the resistance to guinea pig and horse serum inhibitors, series of double dilutions of the sera previously heated to 75 [deg.] C for 10 minutes are mixed.

Preparing the vaccine
The virus from the last pass of the process described in Example I is used as inoculum for the production
of the basic batch for the manufacture of vaccine.
The allantoic fluids are collected and pooled. we check their sterility and harmlessness and mix them
to peptone so as to achieve a final peptone concentration of 5%.
In 3 ml vials, the viral suspension is divided into 0.5 ml fractions so as to obtain doses

Clinical trials

All volunteers were examined daily for possible flu symptoms.
Blood samples were taken before and 2 to 3 weeks after inoculation to determine the number of seroconversions and the geometric mean hemagglutination inhibition titer (GMT).
