La présente invention est relative à de nouveaux composés anti-inflammatoires, En particulier, elle est relative à une nouvelle classe de composés appartenant aux séries de l'indol.
D'une façon encore plus spécifique, la présente invention est relative à de nouveaux acides a-(j-indolyl)-groupe aliphatique inférieur, à leurs sels, esters, anhydrides et dérivés amidés.
Les nouveaux composés substitués d'acide indolyl-aliphatique de la présente invention ont la formule de structure suivante :

Les dérivés indolyles répondant à la description donnée plus haut possèdent un degré élevé d'activité anti-inflammatoire.
Ils.présentent aussi de la valeur dans le traitement des désordres: arthritiques et dermatologiques ou dans le traitement de désordres justifiant l'utilisant de médicaments anti-inflammatoires, Sont comprises dans cette catégorie, les maladies telles que l'arthri- j te rhumatismale, l'ostéo-arthrite, la goutte, les arthrites infec- tieuses et la fièvre rhumatismale. Les dérivés indolyles possèdent aussi un degré d'activité utile comme analgésiques et anti-pyré- . tiques.
A ces fins, on peut administrer les composés de la présente invention par voie bucale, par voie topique, par voie parentérale, par inhalation de produit pulvérisé, ou par vcie ¯ rectale dans des formulations contenant des excipients, adjuvants ; et véhicules acceptables habituels, pharmaceuticuement non toxi- ques. En plus du traitement des animaux à sang chaud tels que les souris, les rats, etc.., les composés de la présente invention'

sont efficaces dans le traitement des êtres humains.
Les excipients pharmaceutiques non toxiques indioués plus haut comprennent soit des solides,soit des liquides. Des exemples d'excipients solides sont le lactose, l'amidon de mais, la gélatine, le talc, le stérotix, l'acide stéarique, le stéarate de magnésium, le bolus alba, le sucrose, la gélose, la pectine, le carbosil et la gomme acacia. Des exemples d'excipients liqui- des sont l'huile d'arachide, l'huile d'olive, l'huile de sésame et l'eau. De même, l'excipient ou diluant peut comprendre un produit retardateur tel que le monostéarate de glycéryle ou le distéarate de glycéryle, seuls ou avec de la cire.
On peut utiliser plusieurs formes pharmaceutiques des compositions thérapeutiquement utiles. Par exemple, si on utilise un véhicule solide, les compositions peuvent prendre la forme de comprimés, de capsules, de poudres, de pastilles ou de tablettes, préparées par des technicues pharmaceutiques standard.
Si on utilise un véhicule liquide, la préparation peut avoir la forme d'une capsule molle en gélatine, d'un sirop ou d'une suspension liquide. On peut préparer des suppositoires de la manière habituelle.
Comme on l'indique plus haut, les dérivés indolyles de la présente invention peuvent être administrés d'une façon efficace par voie parentérale (c'est-à-dire sous ou à travers une ou plusieurs des couches'de la peau ou des muqueuses) comprenant des techniques d'injection intramusculaires,sous-cutanées ou intraveineuse.
On peut réaliser l'administration parentérale,des composés de la présente invention en utilisant des solutions ou suspensions aqueuses ou oléagineuses stériles. Parmi les véhicules acceptables et les solvants qui peuvent être employés on note

l'eau, la solution de Ringer, et une solution isotonique de chlorure de sodium. De plus, on emploie de façon habituelle les huiles solidifées, stériles, comme solvant ou milieu de suspension. On peut employer dans ce but n'importe quelle huile douce solidifiée comprenant des mono- ou diglycérides synthétiques. De plus, des acides gras tels que l'acide oléique trouvent une utilisation dans la préparation des produits injectables.
Comme utilisation sous forme de topique, on emploie des crèmes, des pommades, des gelées ou des solutions ou suspensions contenant les agents anti-inflammatoires.
Des dosages de l'ordre de 10 mg à 7 g par jour sont utiles dans le traitement des désordres indiqués plus haut.
Par exemple, les iflammations sont traitées efficacement, et une activité anti-pyrétique et analgésique se manifeste, par l'administration d'environ 0,3 à 100 mg du composé par kg de poids du corps. Des dosages quotidens d'environ 2 mg à environ

On fait réagir un mélange d'o-nitrodiphénylméthane (0,8 mole), de méthanol (500 ml) et de charbon palladié à 5% de palladium,sous une pression de 2,812 kg/cm2 d'hydrogène, à température ambiante, la réaction s'arrête quand la quantité théorique d'hydrogène a été consommée, on filtre le mélange, on lave bien le gâteau avec du méthanol, les filtrats combinés concentrés sous vide donnent l'o-aminodiphénylméthane.

On élimine l'iodate par filtration, élimine les solvants sous vide, traite le résidu par un mélange chloroforma/eau, décante la couche formée par le chloroforme, la sèche, la filtre et la concentre sous vide.
