La présente invention concerne des médicaments anti- bietiques à tokleité atténuée convenant en particulier pour la médecine humane, ainsi que des procèdes pour les préparer ,les effets toxiques des antibiotiques actifs de ces médicaments étant sensiblement diminues sans que leurs effets antibactériens soient atténues.
On sait que les antibiotiques à propriétés basiques de la classe des streptomycines ont des effets secondaires à caractère texique qui rident leur adninistration difficile (Streptomycine, @ihydrostreptsmycine, Kanamycine etc. ) ou limitent leur champ d'ap-

et,comme terpènes,,des produits d'oxydation de l'essence de térében- thine tels qu'ils peuvent être obtenus par exemple suivant le procédé du brevet français n 1.060.855, de la terpine (ou 1,8-p-mentane- diol) ou son hydrate, de même que les mélanges de tels composés.
Les produits d'oxydation de l'essence de térébenthine (appelés ci-après simplement produits d'oxydation) contiennent, entre autresdes composés à fonction oxo (.comme des cétones ou aldéhydes), c'est-à-dire à radicaux susceptibles de réagir avec les radicaux basiques des antibiotiques, comme les radicaux guanidino de la Streptomycine ou de la Dihydrostreptomycine.
Par mélange de ces terpènes avec un antibiotique basique ou un de ses sels, on peut donc former au moins partiellement des pro- duits de réaction correspondants,de même que des composés d'addi- tion ou complexes.
L'atténuation de la toxicité au moyen d'un mélange d'hydrate de terpine et des produits d'oxydation est mise en évi- dence de la manière suivante:
On administre pendant 10 jours quotidiennement une dose de 250 mg de Streptomycine (sous forme de sulfate) par kg de poids du corps en injection intramusculaire à de jeunes cobayes bien por- . tants d'un poids de 200 g.
A cette fin, on administre le sulfate de Streptomycine aux animaux d'un groupe sous forme de solution dans de l'eau pure et aux animaux de l'autre groupe sous la forme d'une solution comprenant.,Pour 5 ml d'eau,du sulfate de
Streptomycine en une quantité correspondant à 1 g de Streptomycine ainsi que 7,5mg d'hydrate de terpine et 2,5 mg des produits d'oxyda- tion.Au lime jour de l'essai on commence entre autres, des examens électro- physiologiques pour apprécier par mesure du biopotentiel après exci- tation adéquate du système acoustique l'état de fonctionnement ou le degré d'une altération éventuelle de l'organe auditif.

Au cours des essais décrits ci-dessus, on utilise pour 1 mole de Streptomycine 0,023 mole d'hydrate de terpine ainsi que 2,5o/oodes produits d'oxydation,par rapport à la quantité de .
Streptomycine base libre,soit au total 1% de composés terpéniques par rapport à la quantité de Streptomycine base libre. Ces quan- fîtes permettent de réaliser une atténuation significative de la , toxicité de la Streptomycine. Il est évident qu'on peut prendre d'autres valeurs pour les rapports entre l'hydrate de terpine et les produits d'oxydation ou entre ces dernières substances et l'antibiotique ou son sel. Ainsi on peut envisager de prendre des quantités d'environ 0,01mole ou davantage d'hydrate de terpine et de et de 1%. ou davantage des produits d'oxydation (rapportées aux anti- biotiques),Là quantité totale de terpènes ne peut de manière gêné- @ raie excéder 2,5% de la quantité d'antibiotiques (base libre).
Pour la préparation de solutions des antibiotiques à toxicité atténuée, on peut ajouter à l'antibiotique ou son sel une solution d'un mélange d'hydrate de terpine et des produits d'oxy- dation. ou, une solution des divers constituants de ce mélange. Il est évident qu'on peut aussi incorporer une solution de l'anti- biotique ou de son sel avec une solution des composés terpéniques ou de leurs mélanges.
De plus, on peut aussi, par exemple, fai- re agir sur 1 mole d'antibiotique basique 1 mole d'hydrate de ter- pine et/ou une quantité s'élevant,par exemple,à 5 - 10% des pro-

duits d'oxydation,puis ajouter à cette solution une nouvelle quantité d'un antibiotique de nature identique. ou différente ou un sel correspondant,éventuellement à l'état dissous,sans que l'atténuation de toxicité se perde. Dans ce cas, il peut être nécessaire d'ajouter des auxiliaires de dissolution en raison de .la médiocrité de la soluibilité dans l'eau des composés terpé- niques et en particulier des produits d'oxydation.
Les exemples suiv ants illustrent davantage l'inven- tion sans la limiter.
EXEMPLE 1 -
On dissout 7,5 mg d'hydrate de terpine dans 5 ml d'eau, puis on y ajoute du sulfate de Streptomycine en une quantité cor- respondant à 1 g de Streptomycine base libre et on agite le mélan- ; ge convenablement pendant quelque temps. On obtient ainsi une solution limpide qui peut être utilisée telle quelle en thérapeuti- que éventuellement après filtration stérile.
Pour rendre la solution obtenue propre à la conser- vation, on peut y dissoudre,par exemple.0,125 g de bisulfite de sodium et/ou 0,1 g de formaldéhyde sulfoxylate de sodium.
Comme stabilisant, on peut également utiliser,par exemple,du thioglycérol et des composés analogues.
Au lieu d'hydrate de terpine,, on peut aussi uti- liser 2,5 mg des produits d'oxydation ou un mélange de 2,5 mg des produits d'oxydation et de 7,5 mg d'hydrate de terpine.
EXEMPLE2 -
On réalise les opérations comme dans l'exemple 1, mais en utilisant 1 g de Dihydrostreptomycine sous forme de sulfa- te au lieu de la Streptomycine.

On ajoute 20 mg d'hydrate de terpine et 5 mg des

produits d'oxydation à 5 ml d'eau et on ajoute à ce mélange du pantothénate de Streptomycine en quantité correspondant à 500 mg de Streptomycine base libre. Après une agitation convenable, on obtient une solution dans laquelle on dissout du sulfate de-Streptomycine en quantité correspondant à 750 mg de Streptomycine base libre.- On peut stériliser et éventuellement stabiliser la solution obtenue ainsi comme la solution obtenue dans 1 -exemple 1.
Au lieu de la quantité indiquée du mélange des composés terpéniques, on peut prendre également des quantités correspondantes d'hydrate de terpine ou des produits d'oxydation uniquement.
EXEMPLE 4-
On réalise les opérations comme dans les exemples précédents, mais en prenant au lieu des antibiotiques ci-dessus des sulfates et/ou pantothénates de Dihydrostreptomycine, de Kanamycine, de Néomycine ou de Viomycine ou des mélanges de ces sels.
