La présente invention concerne un produit contenant de la bacitracine et le procédé pour le préparer.
On sait qu'on peut préparer la bacitracine par culture de micro-organismes et en particulier de Bacillus subtilis, Depuis quelque temps., cet antibiotique est utilisé à grande échelle pour la préparation d'aliments pour la volaille et le bétail. Entre autres, l'incorporation de la bacitracine aux aliments stimule beaucoup la croissance de la volaille et du bétail.
Toutefois, l'instabilité de la bacitracine dans les con- ditions habituelles d'entreposage crée des difficultés. De plus,

la préparation de la bacitracine par fermentation peut entraîner une perte considérable de l'antibiotique qui se produit au cours de l'isolement par les procédés classiques qui comportent habituelle- ment une évaporation et un séchage au tambour du bouillon de fer- mentation.
Cela étant l'invention a pour but de procurer une composi- tion de bacitracine qui ait un-degré élevé de stabilité à l'entre- posage.
Un autre but de l'invention est de procurer un supplé- ment diététique pour animaux qui contient de la bacitracine stabilisée contre la détérioration en cours d'entreposage,même dans des conditions d'humidité élevée.
L'invention a également pour but de procurer un procédé pour recueillir la bacitracine qui permette un pourcentage, élevé d'isolement et qui n'entraîne que de faibles pertes par comparaison avec les procédés antérieurs.
D'autres buts de l'invention ressortiront de la descrip- 'tion qui en est faite ci-après. 1.
Suivant l'invention un produit amélioré contenant de la ba- citracine peut être préparé en y ajoutant une petite quantité d'un composé du manganèse soluble dans l'eau. Le manganèse sert à stabiliser la bacitracine contre la détérioration en cours d'entreposage. L'invention est particulièrement utile pour la préparation de suppléments diététiques pour animaux qui contiennent ! une base nutritive et de la bacitracine. Dans ce cas, la bacitra- cine est également traitée par un sel de manganèse soluble dans . l'eau afin de la stabiliser dans le produit alimentaire.
L'invention a une utilité particulière pour le traite- ment de bouillons de fermentation en vue d'en isoler la bacitra- çine. Ainsi, suivant l'invention, un bouillon de fermentation

contenant de la bacitracine est traité par une petite quantité d'un sel de manganèse soluble dans l'eau avant d'être soumis à une évaporation et séché au tambour. On a découvert qu'en incorporant ainsi un sel de manganèse, on ne perd qu'une quantité relativement faible de la bacitracine lors de la concentration et du séchage.
L'invention est davantage illustrée, sans être limitée, par l'exemple suivant.
On prépare un milieu de fermentation aqueux contenant '7,5% de farine de soya, 2,0% d'amidon de mais,1,0% de carbonate de calcium et 0,33% de sulfate de magnésium. On stérilise ce milieu par des procédés habituels, puis on inocule avec 0,5% en volume d'une culture liquide de Bacillus subtilis vieille de 24 h.
Le milieu inoculé est alors incubé à environ 30 C sous aération constante pendant 30 heures. A la fin de la période de fermentation, le pH est d'environ 8,1.
Au terme de la fermentation, on ajoute 1,0% en poids de sulfate de manganèse au bouillon de fermentation liquide et le produit de fermentation peut alors être vendu tel quel aux producteurs d'aliments ou il peut être amené à une concentra- tion prédéterminée en bacitracine à l'aide de farine de mais, de farine de soya ou d'autres diluants nutritifs. Ainsi, un supplé- ment diététique approprié est constitué par un mélange de parties à peu près égales du produit séché au tambour et de farine de mais et contient environ 22,0 g de bacitracine par kg.
Le produit séché au tambour et le supplément diététique se distinguent par le fait que la bacitracine qu'ils contiennent est très stable à l'entreposage. Ceci st particulièrement impor- tant lorsque la bacitracine est utilisée dans des aliments qui peuvent être conservés pendant quelque temps avant la consommation,

Par exemple, on ne sait si le manganèse forme un sel ou un complexe avec les produits contenant de ia bacitracine. On ne peut actuellement qu'affirmer que le manganèse agit comme agent de stabilisation.
Dans l'exemple ci-dessus, on utilise du sulfate manganeux ; ' comme véhicule pour incorporer le manganèse à la bacitracine.Toute- fois, il va de soi que tout sel soluble dans l'eau et non toxique du manganèse peut être utilisé, notamment le chlorure manganeux, le nitrate manganeux, le bromure manganeux,etc. 'En général,la ' quantité de sulfate de manganèse utilisée doit être comprise entre environ 0,01 et 5,0% du poids du bouillon de fermentation traité. De préférence cette quantité est toutefois comprise entre
0,3 et 2,0%. En termes d'activité de la bacitracine, la concentra- .. tion du manganèse doit être comprise entre environ 0.6,eu et 300% du poids de la bacitracine.
Dans l'exemple ci-dessus, on se réfère à un milieu de fermentation particulier. Il va cependant de soi que des modifica- , tions peuvent être apportées sous ce rapport et que tout milieu de fermentation approprié pour la production de bacitracine par
Bacillus subtilis peut être utilisé. Les constituants importants sont évidemment l'eau, des hydrates de carbone susceptibles d'être transformés par métabolisme et une source d'azote telle quune protéine assimilable.
Bien que divers modes et détails de réalisation ont été décrits pour illustrer l'invention, il va dè soi que celle-ci est susceptible de nombreuses variantes et modifications sans sortir de son cadre.

The object of the invention is also to provide a process for collecting bacitracin which allows a high percentage of isolation and which causes only low losses compared with the prior processes.

According to the invention an improved product containing bacitracin can be prepared by adding thereto a small amount of a water soluble manganese compound. Manganese is used to stabilize bacitracin against spoilage during storage. The invention is particularly useful for the preparation of dietary supplements for animals which contain! a nutritional base and bacitracin. In this case, the bactracin is also treated with a soluble manganese salt. water to stabilize it in the food product.
The invention has particular utility for the treatment of fermentation broths for the isolation of bactracin therefrom. Thus, according to the invention, a fermentation broth

containing bacitracin is treated with a small amount of a water-soluble manganese salt before being subjected to evaporation and tumble dried. It has been found that by incorporating a manganese salt in this way, only a relatively small amount of the bacitracin is lost during concentration and drying.
The invention is further illustrated, without being limited, by the following example.
An aqueous fermentation medium was prepared containing 7.5% soy flour, 2.0% corn starch, 1.0% calcium carbonate and 0.33% magnesium sulfate. This medium is sterilized by the usual methods, then inoculated with 0.5% by volume of a liquid culture of Bacillus subtilis 24 hours old.
The inoculated medium is then incubated at approximately 30 ° C. under constant aeration for 30 hours. At the end of the fermentation period, the pH is approximately 8.1.
