Les composés de l'invention, du fait de leur mécanisme d'action, doivent être classés parmi les agents antitussifs agis- sant par voie centrale, ce qui a été montré par des tests pharma- ils cologiques. De plus, manquent d'action (narcotique, en se montrant ainsi,sous ce rapport, supérieur à beaucoup de remèdes naturels ou synthétiques couramment employés dans le traitement de la toux.
Depuis les dernièr2s années, on a beaucoup recherché des substances antitussives non narcotiqu, agissant par voie centrale, ce qui a amené l'utilisation de certains agents thé- rapeutiques qui sont capables d'augmenter la résistance du cen- tre médullaire aux impulsions tussives cafférentes sans les effets indésirables des narcotiques. On a maintenant trouvé que les composés de la présente invention assurent un effet antitus- sif qui est comparable à celui de la codéine et est exempt des effets secondaires bien connus de ce remède qui est généralement utilisé.
Le tableau suivant donne la dose protectrice moyenne, en mgr/kg, de certains des composés de l'invention contre la toux provoquée par l'inhalation d'acroléine sous forme d'aéros@ol chez les cobayes.

La toxicité chronique des composés de l'invention est également d'un ordre très faible.
Des doses allant jusqu'à 300 mg/kg étaient administrées journellement à des chiens pandant des périodes aussi longues que 3 mois, sans qu'on observe un effet défavorable quelconque.
Pour des besoins thérapeutiques, les composés de l'in- vention peuvent être utilisés comme tels ou associés avec un vé- hicule qui peut être une matière solide, un liquide parentéral stérile ou un sirop. Lorsqu'on choisit une administration par voie buccale, ce qui est couramment utilisé avec ce type de remèdes, l'administration thérapeutique peut être prévue sous forme de poudes, capsules, tablettes ou autres formes de dosages solides. Pour une utilisation par voie parentérale, on peut. employer des diluants liquides, tels que de l'eau distillée,

stérile, dans lesquels certains des composés sont librement solu- bles.
Bien que des effets appréciables du point de vue clinique soient observés avec des doses aussi faibles que 5 à 20 mgr, le dosage unitaire est habituellement un peu supérieur et peut en toute sécurité atteindre 500 mg et plus lorsque les dosages élevés sont requis pour des cas rebelles particuliers. Pour pré- parer les composés thérapeutiques réels,la dose choisie d'un remède quelconque peut être directement mise en capsules, soit solides seule, soit en association avec des diluants inertes'et/ou autres ingrédients actifs. On peut préparer des tablettes en mélangeant le remède choisi avec les matières couramment utilisées pour la formation de tablettes, telles que lactose,talc, amidon de mats, acide stéarique, stéarate de magnésium, etc.
On prépare des ampoules en dissolvant le remède choisi dans de l'eau distillée stérile et en mettant la solution dans des ampoules contenant un volume suffisant de liquide pour assurer le dosage requis du remède. Les composés peuvent également prendre avantageusement la forme d'un sirop, le remède étant dissous dans un sirop cou- rant en une concentration calculée pour administrer la dose convenable avec ou sans addition d'agents de mise en suspension et/ou d'aromatisation et/ou d'autres remèdes.

Un mélange de 10 gr de 6-carboxy-4,5-dihydro-3-pyri- dazinone et de 100 ml d'une solution à 2% d'acide chlorhydrique dans du méthanol est soumis à reflux pendant 6 heures. Lors d'un refroidissement, la 6-carbométhoxy-4,5-dihydro-3-pyridazinone précipite et est recueillie et séchée. Production : 9 gr (85%); point de fusion : 136-137 C.
L'analogue carbéthoxy peut être préparé de la même ma- nière en utilisant de l'éthanol au lieu de méthanol. Ren- dement : 85%; point de fusion de 135-136 C.
10 gr de 6-carbométhoxy-4,5-dihydro-3-pyridazinone sont dissous dans une solution de méthylate de sodium préparés en par- tant de 1,5 @r de sodium et de 100 ml de méthanol. A une solution de 13,9 gr de 3(2H)-pyridazinone-6- carboxamide, on ajoute 230 ml d'une solution dans du méthanol de méthylate de sodium (correspondant à 2,3 gr de sodium) et 800 ml d'une solution de 14,2 gr d'iodure de méthyle dans du mé- thanol. Le mélange est laissé au repos pendant 1 heure, ensuite il est soumis à reflux pendant 1 heure. Le solvant est enlevé sous le vide, séché avec soin et recristallisé avec de l'éthanol.
Rendement, 85% de 2-méthyl-3-pyridazinone-6-carboxamide, d'un point de fusion de 198-200 . Le composé a été préparé également comme décrit pour la 2-méthyl-4,5-dihydro-3-pyridazinone dans l'exemple précédent en faisant réagir du 2-méthyl-3-pyridazinone- 6-carboxylate de méthyle avec de l'ammoniaque concentré: Rende- ment : 90%.
EXEMPLE 3
Un mélange de 40 gr d'acide 2-méthyl-3-pyridazinone-6- carboxylique et de 160 ml de chlorure de thionyleest soumis à reflux pendant 3 heures; ensuite la solution est évaporée jusqu'à siccité et le résidu est recristallisé à partir d'éther éthylique.

Après une demi-heure, un précipité cristallisé se formant, le chlorhydrate de diéthylamine est enlevé par filtra- tion et le filtrat est évaporé jusqu'à siccité, ce qui donne une huile qui se cristallise, au repos. Ce produit est recristallisé dans de l'isopropanol. Rendement : 84% de 2-méthyl-3-pyridazino- ne-6-carboxy-diéthylamide d'un point de fusion de 73-75 C.
EXEMPLE 5
Un mélange de 20 gr de 2-butyl-4,5-dihydro-3-pyridazi- none-6-carboxylate de méthyle et de 50 ml d'ammoniaque concentrée- est laissé au repos pendant 1 heure. Après cette période le pré- cipité formé est recueilli et recristailisé avec du méthanol.
Rendement: 92% de 2-butyl-4,5-dihydro-3-pyridazinone-6-carboxami- de d'un point de fusion de 172 -174 .
