<Desc/Clms Page number 2>
<Desc/Clms Page number 3>
<Desc/Clms Page number 4>
<Desc/Clms Page number 5>

The invention relates to a process for the preparation of novel heterocyclic derivatives of hydrazine and their salts, characterized in that the product of the general formula
R1-NH-NH-R2 I in which R2 represents the picolinyl or nicotinoyl radical, and R an aralkyl radical, as well as its salts, is obtained
<Desc / Clms Page number 2>
either by condensation of picolinic or nicotinic acid with a substituted hydrazine of the general formula
H2N-NH-R2 II in which R2 has the same meaning as above in the presence of a carbodiimide and, where appropriate, by conversion into a salt, or according to known methods.
In the above general formula I, the aralkyl groups represented by R2 may be monoaryl and diaryl groups, in particular: phenyl-lower alkyl, diphenyl-lower alkyl, lower alkyl-phenyl-lower alkyl ,. such as benzyl, phenyl-isopropyl, tolyl-isopropyl, etc., P-phenyl-isopropyl and benzyl being preferred.
The invention relates, on the one hand, to a new process for the preparation of heterocyclic derivatives of hydrazine. According to this new process, picolinic or nicotinic acid is condensed in the presence of a carbodiimide, with a hydrazine substituted by an aralkyl group.
For condensation, acids can be used directly
<Desc / Clms Page number 3>
or their salts. Conversion into more reactive compounds, such as esters, halides, amides, etc., is unnecessary. The N, N'-disubstituted carbodiimides used for the condensation can be obtained, for example, by treating disubstituted urea with p-toluene- -sulfonic acid chloride in pyridine. The urea derivatives used as a starting material can be recovered after the reaction. By using the appropriate substituted carbodiimides, urea derivatives are obtained as side products which can easily be separated. reaction products.
The reaction can be carried out, for example, at a temperature between 0 and 50, preferably at room temperature or at a slightly elevated temperature. It is advantageous to use a floor covering. It is possible to use both an organic solvent, such as, for example, methylene chloride, chloroform, dioxane, tetrahydrofuran, dimethylformamide or acetonitrile, as water.
On the one hand, the invention relates to the preparation of heterocyclic derivatives of hydrazine according to methods known per se. For example reactive derivatives of nicotinic acid or picolinic acid, such as for example lower alcoholic esters, halides or amides, can be reacted with a hydrazine
<Desc / Clms Page number 4>
substituted by an aralkyl group.
@ It is also possible to react a hydrazine of nicotinic acid or of picolinic acid with a carbonyl compound substituted by an aralkyl group and simultaneously or successively hydrogenate the hydrazones of the carbonyl compound. The hydrogenation is preferably carried out in the presence of catalysts, such as, for example, platinum oxide, palladium on carbon, etc., and in an inert solvent. According to a variant of the latter reaction, it is also possible to react the hydrazone formed with a Grignard compound and to hydrolyze the addition compound thus obtained.
According to the process of the invention, the following derivatives can be obtained, for example: 1-nicotinoyl-2-benzyl-hydrazine, 1-nicotinoyl-2- (2-phenyl-isopropyl) -hydrazine, 1-nicotinoyl-2 -diphenylmethyl-hydrazine, 1-picolinoyl-2-benzyl-hydrazine, -picolinoyl-2- (2-phenyl-isopropyl) -hydrazine, 1-picolinoyl-2-diphenylmethyl-hydrazine, etc.
The heterocyclic derivatives of hydrazine obtained according to the invention form well-defined salts with inorganic and organic acids, for example with halogen-hydric acids, such as hydrochloric acid, hydrobromic acid, hydriodic acid; with others
<Desc / Clms Page number 5>
mineral acids, such as sulfuric acid, phosphoric acid, nitric acid; and with organic acids, such as tartaric acid, citric acid, camphorsulfonic acid, ethanesulfonic acid, salicylic acid, ascorbic acid, maleic acid, acid mandelic, etc. The preferred salts are the hydrohalides, in particular the hydrochlorides.
The acid addition salts are advantageously prepared in an inert solvent by treating the hydrazine derivative with the corresponding acid in excess.
The products obtained according to the process of the invention and their salts exert an inhibitory activity on the monoaminoxidase; some of them are distinguished by their clear antidepressant activity and cause an increase in weight during cachéxia. They therefore represent precious drugs.

platinum, hydrogenated under a pressure of approx. 3-4 atmospheres and at an initial temperature of 70 which is reduced to 25 once the reaction is started. After absorption of the calculated amount of hydrogen, the catalyst is removed. The 1-picolinoyl-2-benzyl-hydrazine thus obtained is recrystallized from ethanol. Melting point 193-194.
Example 6
942 g of 1-picolinoyl-2-benzylidene-hydrazine obtained according to Example 5 are dissolved in 5000 cc of ethanol and, after addition of 150 g of 10% palladium on carbon, hydrogenated under a pressure of 35 atmospheres and at a temperature of 25 The temperature rises to 50 during the reaction. The hydrogenation is continued until a 20% excess of hydrogen is absorbed.
After the hydrogenation, the catalyst is removed by filtration and the remaining mother liquors are concentrated to
2000 cc. The hydrogenation product partially crystallizes on cooling; it is separated by filtration.
On concentrating the filtrate a little more substance is obtained. The combined substances are recrystallized in
1800 cc of boiling alcohol. We thus obtain 1-picolinoyl-
