EAEXTLE 1 ;

The present invention relates to novel mucic acid derivatives and their preparation process.
The Applicant has found that compounds endowed with a marked anti-phlogistic action are obtained by reacting, in the presence of an acid-binding agent, a molecule of tetraacetyl-mucic acid dichloride or dibromide. with two molecules of a mono-alkyl or dial-alkyl ester of an aliphatic amino-monocarboxylic or amino-dicarboxylic acid with a primary or secondary amino group.

The same compounds are also obtained by causing acetyl chloride or acetyl bromide to act.

EMI2.4
The reaction between the tetraacetyl-muoic acid dibllloride and the esters of the specified amino acids can be carried out, for example, in an inert organic solvent or diluent, in the presence of a tertiary organic base such as acid binding agent.

the acid binding agent. Examples of di-amino acid esters are the alkyl esters, in particular methyl and ethyl esters of glycine, sarcosine, alanine, Ó-amino-butyric acid. valine, norvaline, leucine, norleucine and methionine, as well as the dialkyl esters, in particular diethyl esters of aspartic acid and glutamic acid.
Tetra-acetyl-mucyl dichloride can be obtained from tetra-acetyl-mucic acid (Skraup, M. 14, 488). for example using phosphorus pentachloride (Diels, Loflund, B.47, 2352 (1914) or thionyl chloride (J. Huiler, B. 47, 2655).
The dialkyl or tetraalkyl esters of mucyl-N, N'-bis-amino-monoca.rboxylic or bis-amino-dicarboxylic acids, respectively, to be reacted with an acetylating agent by reaction of dialkyl esters, by example diethyl or dimethyl, mucic acid, with the esters of the corresponding amino-carboyxlic acids.
The new compounds can be used. as intermediate products for the preparation of remedies. In addition, they themselves have an interesting pharmacological action. Their anti-phlogiston action can be shown, for example, by the reduction of swelling in the formalin edema of rat paws. This action can be used therapeutically for the treatment of inflammatory disease processes, for example thrombo-phlebitis or rheumatic diseases.

In these examples the parts are parts by weight, unless otherwise indicated.

14 parts of amino-ethyl acetate hydrochloride are added to 21 parts of tetra-acetyl-mucic acid dichloride dissolved in 500 parts by volume of chloroform and then the mixture is poured dropwise into the mixture, with stirring and cooling, 22 parts of triethylamine in 75 parts by volume of chloroform, between 0 and 5. When the dropwise addition is complete, the mixture is stirred for a further 3 hours at room temperature, the chloroform solution is washed with dilute hydrochloric acid then with a solution of sodium bicarbonate and finally with water and dried over sodium bicarbonate. sodium sulfate.
After removal of the solvent by distillation, a residue remains which is recrystallized from methanol. Diethyl tetra-acetyl-mucyl-N, N'-bis-amino-acetate and obtained melts at 216-218.
It is similarly possible, starting with the diethyl ester of 1-glutamic acid or 1-aspartic acid, to the ethyl ester of 1-methionine, sarcosine or 1- alanine, prepare the derivatives of tetra-acetyl-mucic acid corresponding to the formula
