La présente invention se rapporte à des suspensions aqueuses de pénicilline perfectionnées, et, plus particulièrement, des solutions aqueuses de pénicilline contenant des acides inositolphosphoriques ou des sels non toxiques de ces acides.
L'utilisation en médecine de suspensions aqueuses de sels insolubles de pénicilline par voie buccale ou en injection est bien connue. Pendant la période où de telles préparations ont pu être obtenues, on a rencontra des .difficultés procurer des préparations qui, à la conservation, ne précipitent. ni ne forment de flocons et qui soient aisément rendues à l'état de dispersion complète, sans grumeaux. Le seul moyen convenable , à la portée d'un médecin au chevet du malade pour atteindre ce résultat est l'agitation à la main du produit.
Le but de l'in- - vention est de procurer un procédé non toxique'et peu coûteux tel qu'une simple et courte agitation manuelle rétablisse un

état de dispersion complète et uniforme de ces suspensions aqueuses utiles de sels insolubles de pénicilline, même après une conservation prolongée. Un autre but de l'invention est de procurer des suspensions aqueuses de pénicilline de présentation meilleure que celles obtenues actuellement.
Les buts de l'invention ont été atteints grâce à la découverte, suivant la présente invention, d'une suspension aqueuse comportant un sel thérapeutiaue de pénicilline insoluble dans l'eau et un composé choisi dans le groupe comportant les acides inosi- tolphosphoriques et leurs sels non toxiques.
L'inositol, cyclohexane-hexol, C6H6(OH)6, peut'se présenter théoriquement sous sept formes inactives, deux formes optiquement actives et une forme racémique, oui diffèrent par la position relative des groupes hydroxyles dont l'un est lié à chaque atome de carbone. Les deux formes optiquement actives et la forme racémique sont connues ainsi que plusieurs formes inactives.
L'acide inositolhexaphosphorique (C6H18O24P6) est connu sous le nom d'acide phytique; ainsi que ses sels, l'acide phytiaue a été isolé de sources naturelles, par. exemple la liqueur de macération du mais, transformé en divers sels, par exemple les sels de fer, cuivre, calcium, magnésium, manganèse, sodium, potassium et ammonium, et transformé en inositol et en d'autres acides inositolphosphoriques comme les acides'inositolphosphorique- et inôsitoltétraphosphorique. Parmi ces inositols phosphorylés et leurs sels non ,toxiques qui sont utiles pour la présente invention, on préfère utiliser environ 0,1 à 2,0% de phytate de sodium. Les mélanges s'avèrent également utiles.
On -préfère les sels non toxiques solubles dans l'eau. L'acide phytique et ses sels ont été utilisés pour séquestrer le cuivre, le manga- nèse et le fer, pour rendre les jus de fruits déshydratés non- hygroscopiques, pour inhiber l'oxydation des mercaptans en présence de certains métaux, pour empêcher la formation de tartre par l'eau ,,dure, dans l'industrie du papier et pour -procurer une source de

Si on le désire, on peut ajouter un aent tampon comme 0,25 à 5,0% en poids, et, de préférence 0,5% de citrate de sodium ou 1,0 à 7,5% de phosphate de sodium et de préférence 2,6% de phosphate monosodiaue et 2,4% de phosphate bisodique pour faire un total de 5% en poids de la composition totale.Quand cela s'avère utile, comme dans le cas de la présence d'un excès de base libre de procaïne on peut tamponner la suspension en ajoutant des acides libres, par exemple l'acide citrique ou l'a- cide phosphorique. L'emploi d'un tampon n'est cependant pas essentiel à la stabilité ou à l'efficacité thérapeutique des composés de la présente invention. ,
Si on le désire, on peut ajouter des agents de suspen- sion ou de dispersion pour améliorer la présentation pharmaceu- tique.
Comme agent de suspension ou de dispersion, la carboxy- méthylcellulose sodique s'est avérée extrêmement satisfaisante, mais la carboxyméthylcellulose, l'alcool polyvinylique, la poly- vinylpyrrolidone, la gomme adragante, la gélatine, la pectine, les alginates de sodium, le dextrare,, la gomme Karaya etc. sont également utiles. La quantité de l'agent de suspension varie dans une certaine mesure,' mais on en emploie habituellement de 0,2 à 5,0% environ, de préférence de 0,5 à 2,5 environ, et tout chimiste ou pharmacien expérimenté peut apporter des modifications entre ces limites, selon l'usage envisagé du composé. Ainsi, la concentration de la polyvinylpyrrolidone peut varier de 0,1% à 25, mais on préfère en employer 10% environ.
La concentration de dextrane peut varier de 0,1% à 20%, mais on préfère en employer 10% environ. La concentration de pectine peut varier entre 0.,le, et 0,5%, mais on préfère en employer 0,2% environ. La concentration de gomme.adragante peut varier de 0,5% à 2%, mais on préfère en employer 1% environ, on peut y ajouter 5% de chlorure de sodium.

sont employés de manière interchangeable pour décrire les addi- tifs comme le carboxymthylcellulose sodique, la lécithine., les Spans et Tweens qui améliorent le comportement pharmaceutique de ces préparations,notamment en rendant plus aisées l'injection et la resuspension après précipitation.
D'autres agents de sus- pension et de dispersion comprennent la lécithine, le Falba, le cholestérol,le Span 20, le Span 40, le Span 60, le Span 80, les Tweens, les Amerchols, l'alginate de sodium, l'alginate de po- tassium, l'alginate d'ammonium, l'alginate de calcium, l'acide alginique, l'alginate de propylène glycol, les dérivés poly- oxyalkylènes des esters d'acides gras du sorbitol, l'urée et le p-aminobenzoate de sodium.
La suspension de la pénicilline-procaine peut avoir n'importe quelle puissance de 10,000 à 600,000 unités environ par millilitres, de préférence de 100,000 à 400,000 unités environ, par millilitre. Ordinairement, une suspension de pnicilline-G procaïne ayant une puissance d'environ 300,000 unités par Millilitre est optimum et s'avère entièrement satisfaisante. La puissance de la pénicilline-procaïne ne doit pas être interprétée comme un facteur de limitation, et les diverses activités sont simplement mentionnées pour indiquer que de la pénicilline-G procaïne d'activités diverses convient pour être incorporée dans la composition 'de la présente invention, tenant compte à nouveau de l'application envisagée de la suspension aqueuse.
La composition n'est pas limitée aux seuls ingrédients mentionnés et à l'exclusion de tout autre, puisque divers autres ingrédients peuvent être éventuellement ajoutés. Par exemple, on peut employer une petite quantité d'agent de conservation, comme le Phénol USP, le Crsol USP, le Méthyl Paraben (ester méthylique de l'acide p-oxybenzoïque), l'Ethyl Paraben (ester éthylique de l'acide p-oxybenzoïque), le Bentyl Paraben (ester butylique de l'acide p-oxybenzoloue) ou le Propyl Paraben (ester propylique de l'acide p-oxybenzoïque).

The present invention relates to improved aqueous suspensions of penicillin, and more particularly to aqueous solutions of penicillin containing inositolphosphoric acids or non-toxic salts thereof.
The use in medicine of aqueous suspensions of insoluble salts of penicillin by mouth or by injection is well known. During the period when such preparations could be obtained, difficulties were encountered in obtaining preparations which, on storage, do not precipitate. neither form flakes and which are easily rendered completely dispersed without lumps. The only suitable means within the reach of a bedside physician to achieve this result is by hand stirring the product.
The object of the invention is to provide a non-toxic and inexpensive process such that a simple and short manual agitation will restore a

The objects of the invention have been achieved thanks to the discovery, according to the present invention, of an aqueous suspension comprising a therapeutic salt of penicillin insoluble in water and a compound selected from the group comprising inositolphosphoric acids and their non-toxic salts.
