INSECTICIDE D'ORIGINE ANIMALE.
Pour mieux mettre en évidence l'importance de la découverte que des nouvelles substances insecticides peuvent etre obtenues du règne animal, il convient d'exposer d'abord quelques considérations générales.
A présent, on dispose d'insecticides de grande valeur, tant dans le domaine agricole que dans le domaine civil et domestique, mais en général on est loin de l'antiparasite idéal; en effet, il y a beaucoup de produits de haute efficacité, mais leur sélectivité d'action est faible, c'est-à-dire qu'ils exercent une action toxique non seulement sur les insectes à combat- tre, mais aussi sur d'autres insectes qui sont leurs ennemis naturels, ou rivaux dans la question de la nourriture, quelquefois même sur les plantes hôtes et, ce qui est encore plus grave, sur l'homme et sur les animaux supé- rieurs.
Par conséquent, bien que le nombre des substances insecticides soit grand, celles que l'on peut employer effectivement dans la pratique - même avec de nombreuses précautions hygiéniques - sont peu-nombreuses, et il y en a très peu qui ne donnent pas lieu à des préoccupations.
Elles dérivent des plantes et les plus connues sont les pyréthri- nes, mais à cause de l'action des agents atmosphériques, leur efficacité est si brève que l'emploi, à cause de leur labilité, est limité à un champ d'ap- plication très-restreint.
La découverte qu'il est possible d'obtenir-des substances insec- ticides nouvelles à partir des organismes animaux, eu égard à la,situation esquissée ci-dessus, est de nature à ouvrir une voie extraordinairement pro- metteuse et de réalisation pratique pour les raisons suivantes : - pour la première fois on a obtenu des substances insecticides du règne animal et différentes de l'acide formique ; priorité a une

grande importance parce qu'elle ouvre un champ très vaste;
- ces insecticides d'origine animale possèdent une sélectivité marquée, au moins en ordre général,, entre les animaux à sang chaud'et les insectes, et aussi dans la classe même des insectes:En. particulier l'iri- domyrmecine, découverte par la demanderesse dans l'iridomyrmex humilis Mayr, et obtenue pour la première fois suivant les nouveaux procédés à partir de cet insecte, si elle est administrée par la voie gastrique à la "souris blanche" (Mus musculus albinus) s'avère être 6-7 fois moins toxique que le DDT-pp' et l'isomère gamma de l'hexachlorocyclohexane, et 120 fois moins toxique que le parathion.
D'autre part, sur des espèces différentes d'in- sectes d'intérêt agraire ou sanitaire, l'iridomyrmécine a une action toxi- que supérieure à celle du DDT-pp'; - les insecticides que la demanderesse a extraits jusqu'ici du règne animal possèdent une stabilité élevée par rapport aux agents chimiques et physiques, qui au contraire altèrent rapidement les pyréthrines : c'est là une caractéristique importante pour les buts des possibilités d'emploi et de l'économie des traitements; - en outre, les insecticides auxquels la présente découverte se réfère, possèdent une très grande rapidité d'action insecticide.
Par exemple l'iridomyrmécine agit sur beaucoup d'espèces d'insectes économiquement impor- tantes dans un temps beaucoup plus bref que le DDT et dans certains cas mê- me plus rapidement que le Lindane. Cette caractéristique la fait ressembler aux pyréthrines, tandis que les insecticides organiques de synthèse les plus importants sont plutôt lents ; -l'iridomyrmécine a une constitution chimique qui ne correspond à celle d'aucune substance insecticide connue ; elle constitue donc une orien- tation nouvelle dans le domaine des insecticides;
- l'iridomyrméoine (une substance dépourvue de chlore) s'est aussi avérée efficace contre des souches d'insectes "résistantisés" (c'est- à-dire rendus résistants) contre le DDT, dans lesquels se développe aussi toujours une résistance plus manifeste contre les autres insecticides orga- niques contenant le chlore dans la molécule.
Il était connu que beaucoup d'organismes animaux sont doués de substances défensives et offensives chimiques, tel que l'acide formique des fourmis, mais jusqu'ici on ne savait presque rien de.ces substances et, en particulier, on ne connaissait pas la possibilité de les utiliser en qualité d'insecticides.
Ayant soumis à des recherches systématiques un premier lot de plus de 50 espèces animales, la demanderesse a découvert que dans un grand nombre d'espèces il existe des principes insecticides, jusqu'ici inconnus, ayant des caractéristiques d'un haut intérêt du point de vue des applica- tions, et que des substances peuvent être obtenues industriellement par des procédés qui ont été mis au point par la demanderesse.
Par exemple, la présence de ces principes à action insecticide a été mise en évidence dans les extraits d'espèces différentes des insectes Dolychoderinae des genres Tapinoma, Liometopum,Iridomvrmex.
Les substances extraites jusqu'ici sont des composés organiques d'un poids moléculaire bas, qui agissent sur les insectes comme un puissant poison de contact et d'asphyxie avec des manifestations très marquées de nature toxique, tout en étant de toxicité assez faible pour les animaux à sang chaud.
Par exemple sur l'hémiptère Pyrrochoris apterus L. à des doses de 10 par cm2, le DDT-pp? dans les 6 heures, ne provoque pas d'appari- tion de symptômes d'empoisonnement, tandis que des doses égales d'iridomyr- mécine provoquent une supination générale avec des contractions cloniques en 4 heures.

Pour donner un autre exemple de la toxicité sur un insecte d'in- térêt agraire, on peut signaler que l'iridomyrmécine en vitro à une dose de la 6 par cm2 produit, par contact, des effets mortels sur l'Iridomyrmex humilis Mayr ("fourmi argentine") même, avec une rapidité'beaucoup plus' grande que des concentrations égales de DDT-pp' et plus rapidement aussi qu'une dose égale de l'isomère de l'hexachlorocyclohexane.- --- - -- --
Les substances actives découvertes par la demanderesse agissent aussi sur les espèces ou souches d'insectes'résistantes contre les insecti- cides chlorurés de synthèse employés à présent, par exemple 'sur:'les mouches domestiques résistantes au DDT.
Par exemple une souche de cet insecte résis- tant pendant 24 heures de contact à des doses de 10.000 - de DDT-pp' par cm2, présentait après deux heures de permanence sur une surface traitée avec l'iridomyrmécine à la dose de 6,5 par cm2, 93 % d'individus en supina- tion et, après 4 heures, 90 % d'individus en akinèse irréversible, 62 %'en supination avec des contractions cloniques, 4 % en supination temporaire, tandis qu'après 2 heures, à des doses de 3 [alpha] par cm2, elle accusait la supination de 76 % des individus et après 4 heures l'akinèse irréversible de 80 % des individus.
Les substances actives insecticides sont contenues dans-les or- ganes internes et partant, pour pouvoir les utiliser, il faut d'abord soumet- tre les animaux à une désintégration très fine, ou à l'extraction avec des solvants convenables ou à la sublimation ou à un autre procédé qui s'y prê- te.
A titre illustratif et non limitatif, quelques exemples des pro- cédés pour obtenir ces substances actives seront décrits ci-après.
EXEMPLE 1.
Extraction avec un solvant et purification de l'iridomvrmécine.
Les individus d'Iridomyrmex humilis Mayr, recueillis dans la na- ture ou dans les élevages spéciaux, et mélangés soigneusement avec un exci- pient constitué par de la craie précipitée naturelle, sont broyés dans un moulin jusqu'à obtenir une masse homogène.
La charge du moulin est passée alors au séchoir sous vide ou en courant d'air, en tenant dans ce cas la température à une valeur ne dé- passant pas 50 , de préférence voisine de 30 .
La masse séchée dans les cuvettes en couche mince, est désinté- grée et passée subséquemment à l'appareil extracteur; l'extraction s'effec- tue au moyen de l'éther sulfurique, préchauffé à 30 et circulant en contre- courant sur une batterie de trois extracteurs. L'opération d'extraction est prolongée jusqu'à ce que le solvant ne véhicule plus de principes actifs.
Le résidu solide de l'extraction, après élimination de tout l'éther (que l'on récupère), est déchargé, reprenant le cycle d'extraction avec de la poudre nouvelle, toujours suivant le principe du contre-courant. L'éther, enrichi des produits solubles, est envoyé à une chaudière de distillation, avec chemise à vapeur, où il s'évapore pour retourner dans le cycle, tandis que la substance extraite reste dans la chaudière même.
La substance extraite est constituée par une huile dense, vis- queuse, foncée à cause de la présence de pigments liposolubles. Elle est .déchargée dans un appareil de distillation dont on obtient, en opérant entre 60 et 100 à la pression atmosphérique, un sublimé cristallin dont les ca- ractéristiques sont contrôlées par la détermination des constantes chimico- physiques.
De cette fagon on sépare une fraction de tête et une fraction de queue, qui sont transférées à des opérations successives de distillation ou de sublimation, tandis que la fraction moyenne est constituée par de l'iri- domyrmécine à haut degré de pureté. Le rendement industriel du procédé dé- crit est d'environ 1 % d'iridomyrmécine pure, par rapport au poids de la ma- tière organique de départ ; mille millions d'ouvrières ayant le gaster non replet, dans la saison d'été, on obtient environ 3500 gr d'iridomyrmécine.

A titre de variante du procédé décrit ci-dessus, l'huile obte- nue par l'extraction peut être laissée à repos prolongé câpres, lequel un gros pourcentage de l'iridomyrmécine se sépare sous forme'cristalline bru- te; ce produit, filtré, peut être employé directement pour la préparation d'insecticides agricoles..
Le produit cristallin brut peut être purifié, si cela s'avère nécessaire, par sublimation directe,,
L'iridomyrmécine, c'est-à-dire le produit insecticide chimi- quement pur ainsi obtenu, est constituée par un composé chimique ayant' la formule brute C10H16O2, et caractérisé par les valeurs suivantes des pro- priétés chimico-physiques point de fusion F. 60-61 C; point de ramollis- sement = 58-59 C; pouvoir rotatoire ( #/65 )D20 = + 210 ( c = '4; alcool éthy- lique);
index de réfraction nD = 1.4607; caractéristiques chimiques d'un lacton bicyclique. -
Ce produit, traité par de la potasse méthyl-alcoolique avec une acidification subséquente, se transforme en un autre lacton isomère, ayant F. = 55 C, ( [alpha])D20 = -52,5 (c = 4 ; éthylique), ayant lui aussi des propriétés insecticides.
.EXEMPLE 2.
Sublimation directe de l'iridomyrmécine..' @
On recueille le matériel vivant, Iridomyrmex humilis Mayr, et on le réduit en pâte homogène en opérant dans un appareil mécanique convena- ble ; la pâte ainsi obtenue est desséchée d'une façon analogue à ce qui-est décrit dans l'exemple précédent, et est ensuite broyée dans un moulin, en la réduisant en poudre fine que l'on peut assujettir à la sublimation frac- tionnée directe :on obtient ainsi une fraction moyenne constituée par de l'iridomyrmécine pure, tandis que les produits de tête et de queue sont réutilisés comme indiqué dans l'exemple précédent.
EXEMPLE 3.
Utilisation de l'iridomyrmécine brute.
Le matériel vivant, transformé en poudre fine, est mélangé avec des excipients convenables pour préparer des poudres à employer dans des buts insecticides, soit sèches, soit en suspension aqueuse ; suivant le rap- port de dilution obtenu avec l'addition des excipients on a des produits 'qui se prêtent aux différentes applications et pour les différents parasi- tes.
EXEMPLE 4.
Extraction du principe insecticide de Tapinoma nigerrimum Nvl.
Les corps des ouvrières de Tapinoma nigerrimum Nyl. sont assu- jettis au broyage, après dilution avec un excipient minéral (carbonate de calcium précipité) et à l'extraction subséquente avec l'éther sulfurique, suivant des modalités analogues à celles de l'Exemple 1.
On obtient ainsi une huile brute ayant une activité insecticide marquée, mais ayant des caractéristiques d'efficacité, par rapport aux dif- férentes espèces d'insectes, ne correspondant pas à celles des extraits bruts obtenus de l'Iridomyrmex humilis et pas non plus à celles de l'iridomyrméci- ne, EXEMPLE 5.
Extraction du principe insecticide de Liometopum nigerrimum Panz.
Par extraction avec l'alcool éthylique, suivant un procédé ana- logue aux précédents,on obtient à partir des corps des ouvrières de Liome- topum microcephalum,une huile ayant une activité insecticide, mais avec

des caractéristiques d'efficacité, par rapport aux différentes espèces d'insectes, ne correspondant pas à celles des extraits bruts obtenus de Iridomyrmex ou de Tapinoma et pas non plus à celles de liridomyrmécine.
REVENDICATIONS. la Substances insecticides caractérisées par le fait d'être obtenables à partir des insectes, en particulier des hyménoptères Doly- choderinaetels que Liometopum microcephalum Panzo, Tapinoma nigerrimum Nylo,- Iridomyrmex humilis Mayr et analogues, et d'être différentes'de l'a- cide formique et de ses composés.

INSECTICIDE OF ANIMAL ORIGIN.
To better demonstrate the importance of the discovery that new insecticidal substances can be obtained from the animal kingdom, it is appropriate to first set out some general considerations.
At present, we have insecticides of great value, as much in the agricultural field as in the civil and domestic fields, but in general we are far from the ideal antiparasite; indeed, there are many products of high efficiency, but their selectivity of action is low, that is to say they exert a toxic action not only on the insects to be combated, but also on d other insects which are their natural enemies, or rivals in the question of food, sometimes even on host plants and, what is even more serious, on man and higher animals.
Consequently, although the number of insecticidal substances is great, those which can be used effectively in practice - even with many hygienic precautions - are few, and there are very few which do not give rise to concerns.
They are derived from plants and the best known are the pyrethrins, but because of the action of atmospheric agents, their effectiveness is so short that the use, because of their lability, is limited to a field of application. very restricted plication.
The discovery that it is possible to obtain new insecticidal substances from animal organisms, in view of the situation outlined above, is likely to open an extraordinarily promising and practical way of achieving. the following reasons: - for the first time insecticidal substances have been obtained from the animal kingdom and different from formic acid; priority has a

great importance because it opens up a very wide field;
- these insecticides of animal origin have a marked selectivity, at least in general, between warm-blooded animals and insects, and also within the class of insects: En. in particular iridomyrmecine, discovered by the Applicant in iridomyrmex humilis Mayr, and obtained for the first time according to the new processes from this insect, if it is administered by the gastric route to the "white mouse" (Mus musculus albinus) is found to be 6-7 times less toxic than DDT-pp 'and the gamma isomer of hexachlorocyclohexane, and 120 times less toxic than parathion.

For example, iridomyrmecin acts on many economically important insect species in a much shorter time than DDT and in some cases even faster than Lindane. This characteristic makes it resemble pyrethrins, while the most important synthetic organic insecticides are rather slow; -iridomyrmecine has a chemical constitution which does not correspond to that of any known insecticidal substance; it therefore constitutes a new direction in the field of insecticides;
- Iridomyrmeoine (a substance devoid of chlorine) has also been shown to be effective against strains of insects "resistant" (that is to say made resistant) against DDT, in which also always develops more resistance. manifested against other organic insecticides containing chlorine in the molecule.
It was known that many animal organisms are endowed with chemical defensive and offensive substances, such as the formic acid of ants, but until now almost nothing was known about these substances and, in particular, we did not know the possibility of using them as insecticides.
Having subjected to systematic research a first batch of more than 50 animal species, the Applicant has discovered that in a large number of species there are insecticidal principles, hitherto unknown, having characteristics of great interest from the point of view of view of the applications, and that substances can be obtained industrially by processes which have been developed by the applicant.
For example, the presence of these principles with insecticidal action has been demonstrated in extracts of different species of the Dolychoderinae insects of the genera Tapinoma, Liometopum, Iridomvrmex.
The substances extracted so far are organic compounds of a low molecular weight, which act on insects as a powerful contact and asphyxiating poison with very marked manifestations of a toxic nature, while being of low enough toxicity for them. warm blooded animals.
For example on the hemiptera Pyrrochoris apterus L. at doses of 10 per cm2, DDT-pp? within 6 hours, does not cause symptoms of poisoning, while equal doses of iridomyrecine cause general supination with clonic contractions within 4 hours.

To give another example of toxicity on an insect of agrarian interest, it can be pointed out that iridomyrmecine in vitro at a dose of 6 per cm2 produces, by contact, lethal effects on Iridomyrmex humilis Mayr ( "Argentine ant") same, with a rapidity 'much greater' than equal concentrations of DDT-pp 'and also faster than an equal dose of the hexachlorocyclohexane isomer. - --- - - - -
The active substances discovered by the Applicant also act on species or strains of insects resistant to the synthetic chloride insecticides now employed, for example on: house flies resistant to DDT.
For example, a strain of this insect resistant for 24 hours of contact to doses of 10,000 - of DDT-pp 'per cm2, showed after two hours of permanence on a surface treated with iridomyrmecine at a dose of 6.5 per cm2, 93% of individuals in supination and, after 4 hours, 90% of individuals in irreversible akinesis, 62% in supination with clonic contractions, 4% in temporary supination, while after 2 hours, at doses of 3 [alpha] per cm2, it showed supination in 76% of individuals and after 4 hours irreversible akinesis in 80% of individuals.
The insecticidal active substances are contained in the internal organs and therefore, in order to be able to use them, the animals must first be subjected to very fine disintegration, or to extraction with suitable solvents or to sublimation. or some other suitable process.
By way of illustration and without limitation, a few examples of the processes for obtaining these active substances will be described below.
EXAMPLE 1.
Extraction with a solvent and purification of iridomvrmecin.
The individuals of Iridomyrmex humilis Mayr, collected in nature or in special farms, and carefully mixed with an excipient consisting of natural precipitated chalk, are ground in a mill until a homogeneous mass is obtained.
The load from the mill is then passed through the dryer under vacuum or in a draft, in this case keeping the temperature at a value not exceeding 50, preferably close to 30.
The mass dried in the cuvettes in a thin layer is disintegrated and subsequently passed to the extractor apparatus; the extraction is carried out by means of sulfuric ether, preheated to 30 and circulating in countercurrent on a battery of three extractors. The extraction operation is prolonged until the solvent no longer conveys any active ingredients.
The solid residue of the extraction, after elimination of all the ether (which is recovered), is discharged, resuming the extraction cycle with new powder, always following the principle of the counter-current. Ether, enriched with soluble products, is sent to a distillation boiler, with steam jacket, where it evaporates to return to the cycle, while the extracted substance remains in the boiler itself.
The extracted substance is a dense, viscous, dark oil due to the presence of fat-soluble pigments. It is discharged into a distillation apparatus from which, operating between 60 and 100 at atmospheric pressure, a crystalline sublimate is obtained, the characteristics of which are checked by determining the chemical-physical constants.
In this way, a top fraction and a bottom fraction are separated, which are transferred to successive distillation or sublimation operations, while the middle fraction consists of iridomyrmecin with a high degree of purity. The industrial yield of the described process is about 1% pure iridomyrmecine, based on the weight of the starting organic material; a thousand million workers having the non-plump gaster, in the summer season, about 3500 gr of iridomyrmecine are obtained.

As an alternative to the process described above, the oil obtained by the extraction can be left to stand for a long time, capers, whereby a large percentage of the iridomyrmecin separates out as crude crystalline; this product, filtered, can be used directly for the preparation of agricultural insecticides.
The crude crystalline product can be purified, if necessary, by direct sublimation.
Iridomyrmecin, that is to say the chemically pure insecticide product thus obtained, consists of a chemical compound having the crude formula C10H16O2, and characterized by the following values of the chemical-physical properties melting point Mp 60-61 C; softening point = 58-59 C; optical rotation (# / 65) D20 = + 210 (c = '4; ethyl alcohol);
refractive index nD = 1.4607; chemical characteristics of a bicyclic lacton. -
This product, treated with methyl alcoholic potash with subsequent acidification, turns into another isomeric lacton, having F. = 55 C, ([alpha]) D20 = -52.5 (c = 4; ethyl), also having insecticidal properties.
EXAMPLE 2.
Direct sublimation of iridomyrmecin .. '@
The living material, Iridomyrmex humilis Mayr, is collected and reduced to a homogeneous paste by operating in a suitable mechanical apparatus; the paste thus obtained is dried in a manner analogous to what is described in the preceding example, and is then ground in a mill, reducing it to a fine powder which can be subjected to direct fractional sublimation : an average fraction is thus obtained consisting of pure iridomyrmecine, while the head and tail products are reused as indicated in the previous example.
EXAMPLE 3.
Use of crude iridomyrmecin.
The living material, transformed into a fine powder, is mixed with suitable excipients to prepare powders to be used for insecticidal purposes, either dry or in aqueous suspension; depending on the dilution ratio obtained with the addition of the excipients, there are products which are suitable for the different applications and for the different parasites.
EXAMPLE 4.
Extraction of the insecticidal principle from Tapinoma nigerrimum Nvl.
The bodies of workers of Tapinoma nigerrimum Nyl. are subjected to grinding, after dilution with an inorganic excipient (precipitated calcium carbonate) and to the subsequent extraction with sulfuric ether, according to modalities similar to those of Example 1.
A crude oil is thus obtained having a marked insecticidal activity, but having characteristics of efficacy, compared to the various species of insects, not corresponding to those of the crude extracts obtained from Iridomyrmex humilis and not to those of iridomyrmecine, EXAMPLE 5.
Extraction of the insecticidal principle from Liometopum nigerrimum Panz.
By extraction with ethyl alcohol, following a process analogous to the preceding ones, from the bodies of workers of Liometopum microcephalum, an oil is obtained having insecticidal activity, but with

efficacy characteristics, relative to the various insect species, not corresponding to those of the crude extracts obtained from Iridomyrmex or from Tapinoma and not to those of liridomyrmecine either.
CLAIMS. Insecticidal substances characterized by being obtainable from insects, in particular Hymenoptera Doly- choderina, such as Liometopum microcephalum Panzo, Tapinoma nigerrimum Nylo, - Iridomyrmex humilis Mayr and the like, and being different from a- formic acid and its compounds.
