Die Erfindung bezieht sich auf ein Verfahren zum Herstellen neuer, Diphenylsulfidderivate darstellender Essigsäurethiolester, welche in der Lage sind, im Blut von Säugetieren die Menge an Lipiden wie sämtliche Cholesterine und Triglyceride zu verringern.
Im allgemeinen wird die Abscheidung von überschüssigen Mengen der im Blut vorhandenen Lipide als Folge von Arterosclerose wie Atherosclerose angesehen. Dementsprechend wird die Verringerung der Lipidkonzentration im Blut für die Therapie von Arterosclerose und damit verknüpften Krankheiten als wünschenswert erachtet.
Im Zuge ausgedehnter Untersuchungen wurde gefunden, dass gewisse derartige Diphenylsulfidderivate die Menge an im Blut von Säugetieren insgesamt vorhandenen Cholesterinen und Triglyceriden bei nur geringen Nebenwirkungen äusserst stark verringern.
Dementsprechend ist es Ziel der Erfindung ein Verfahren zum Herstellen neuer derartiger Diphenylsulfidderivate zu schaffen, welche in der Lage sind, im Blut von Säugetieren die Menge an den gesamten Cholesterinen und Triglyceriden zu verringern.
Weitere Ziele und Vorteile der vorliegenden Erfindung ergeben sich aus der folgenden Beschreibung, wo die Erfindung im einzelnen erläutert ist.

Die Verbindungen der allgemeinen Formel (I) zeigen im Blut von Säugetieren wie Ratten, Kaninchen und Menschen gute hypolipidämische Wirkung und besitzen niedrige Toxizität und sind deshalb als Heilmittel verwendbar. Zu diesem Zweck kann eine erfindungsgemäss hergestellte Verbindung oral oder parenteral in Form von nach in der Pharmazie üblichen Methoden hergestellten üblichen Verabreichungsformen wie Tabletten, Kapseln, Pulvern, Granulaten, Sirupen und Injektionspräparaten verabreicht werden.
Die wirksame Dosis einer erfindungsgemäss hergestellten Verbindung hängt vom Alter, vom Gewicht und vom Zustand des Patienten ab, jedoch liegt im allgemeinen die an Erwachsene in Form einer Einzeldosis oder mehreren Dosen zu verabreichende Tagesdosis zwischen 0, 1 und 3 g, vorzugsweise zwischen 0, 3 und 1, 5 g.
Aus der EP-A1-88 585 sind bereits lipidsenkende Verbindungen bekannt, welche sich jedoch von den erfindungsgemäss herstellbaren Verbindungen dadurch unterscheiden, dass sie sich vom Diphenyläther ableiten und eine andere Thiolsäurekomponente aufweisen können ; ihre Herstellung erfolgt analog zur Herstellung der erfindungsgemäss herstellbaren Verbindungen. Diese Verbindungen haben eine ausgezeichnete triglyceridsenkende Wirkung auf die Leber, jedoch ist ihre cholesterinsenkende Wirkung für manche Fälle der Hypercholesterinämie unzureichend, wie der nachfolgende Vergleichsversuch zeigt.
Vergleichsversuch :
Acht Wochen alte männliche Wistar-Ratten mit einem Körpergewicht von 170 g wurden für jede Gruppe verwendet und mit einer cholesterinreichen Diät, enthaltend 1% Cholesterin und 0, 5% Natriumcholeat, 7 Tage lang gefüttert. Während der Fütterung mit cholesterinreicher Diät wurden Suspensionen der Testverbindungen in einer 0,2%gen wässerigen CMC-Na-Lösung den Ratten jeder Gruppe in einer Menge von 100 mg/kg/Tag 7 Tage lang oral verabreicht. Eine 0, 2% ige CMC-Na-Lösung wurde Ratten oral als Kontrolle verabreicht. Nach Verabreichung wurde jede Ratte 24 h lang fasten gelassen und dann mit Diäthyläther anästhesiert. Durch die Leistenarterie und Leistenvene wurde Blut abgenommen und das Lipidvolumen im Serum unter Verwendung eines Analyseautomaten bestimmt.
Gleichzeitig wurde die Leber entfernt und ihr relatives Gewicht (bezogen auf das Körpergewicht) wurde bestimmt.

Die Erfindung, die weitere Pharmakologie der Endprodukte und die Herstellung der Ausgangsstoffe werden im folgenden durch Versuche und Beispiele konkret erläutert.
Versuch la)
5 Wochen alte männliche Wistar-Ratten mit einem Körpergewicht von 120 : 6 g wurden eine Woche bei konstanter Temperatur und konstanter Luftfeuchtigkeit ernährt. Für jede Versuchsgruppe wurden 5 Ratten verwendet. Als Vergleichsverbindung dient Clofibrate {die chemische Bezeichnung ist Äthyl- [2- (4-chlorphenoxy)-2-methylpropionat]}. Die Verbindungen der allgemeinen Formel (I) und die Vergleichsverbindung wurden für die Versuche in Form einer Suspension in 5% iger Gummi arabicum-Lösung verwendet. Jede der zu prüfenden Verbindungen wurde an die Ratten jeder Gruppe von Versuchstieren während drei aufeinanderfolgender Tage in einer Menge von 100 mg/kg verwendet. Zu Vergleichszwecken wurde an Ratten oral lediglich eine 5%ige Gummi arabicum-Lösung verabreicht.
Nach Verabreichung der zu prüfenden Verbindung wurden die Ratten während 18 h nicht mit Nahrung versorgt, worauf die Ratten mit Diäthyläther anästhesiert wurden. Das Blut der Ratten wurde über die Leistenarterie und über die Leistenvene abgezogen, worauf das Volumen des im Serum enthaltenen Lipids mittels eines Analysenautomaten bestimmt wurde.
Gleichzeitig wurde die Leber entfernt und das Verhältnis ihres Gewichtes zum Körpergewicht bestimmt.
In der folgenden Tabelle ist die Absinkgeschwindigkeit der Blutlipide in % der Differenz der Lipidkonzentration zwischen der mit Prüfverbindung versorgten Gruppe und der Vergleichsgruppe und die Geschwindigkeit der Zunahme des Gewichts der Leber in % der Differenz des Lebergewichts zwischen der mit Prüfverbindung versorgten Gruppe und der Vergleichsgruppe für den Fall der Verabreichung einer Verbindung der Formel (I) und der Vergleichsverbindung angegeben.

Versuch 2
Akute Toxizität
8 Wochen alte männliche ICR-Mäuse mit einem Körpergewicht von 28 bis 32 g wurden zu je 8 Mäuse umfassenden Gruppen zusammengefasst, oral mit einer Suspension der gemäss Beispiel 1 hergestellten Verbindung in 5%iger Gummi arabicum-Lösung versorgt und dann 7 Tage beobachtet, worauf der LD -Wert berechnet wurde.
Der LD--Wert der gemäss Beispiel 1 hergestellten Verbindung überstieg 1000 mg/kg.
Beispiel 1 :
1. In 100 ml Dichlormethan wurden 5, 58 g Diphenylsulfid und 2, 99 g Maleinsäureanhydrid gelöst, worauf die erhaltene Lösung allmählich mit 5, 97 g wasserfreiem Aluminiumchlorid versetzt und das erhaltene Gemisch bei Raumtemperatur 5 h gerührt wurde. Das Reaktions- gemisch wurde sodann unter verringertem Druck eingeengt, worauf das erhaltene Konzen- trat in ein Gemisch aus 5 ml konzentrierter Salzsäure und 100 g Eis gegossen und das erhaltene Gemisch mit Äthylacetat extrahiert wurde.
Der erhaltene Extrakt wurde mit
Wasser gewaschen, über Magnesiumsulfat getrocknet und durch Eindampfen unter verrin- gertem Druck vom Äthylacetat befreit, worauf der erhaltene Rückstand aus einem Gemisch aus Hexan und Äthylacetat umkristallisiert wurde, womit 6, 56 g 3- (4-Phenylthiobenzoyl)- - acrylsäure mit Fp. = 157 bis 159 C erhalten wurden.
2. Eine Lösung von 2, 84 g'3- (4-Phenylthiobenzoyl) -acrylsäure in 30 ml Chloroform wurde mit 0, 8 ml Thioessigsäure versetzt, worauf das erhaltene Gemisch 5 h bei Raumtemperatur gerührt und durch Eindampfen unter verringertem Druck vom Chloroform befreit, der erhaltene Rückstand durch Säulenchromatographie an einer mit Silikagel gefüllten Kolonne unter Verwendung eines Gemisches aus Hexan und Äthylacetat als Eluiermittel gereinigt und die aus dem Eluat erhaltene Verbindung aus einem Gemisch aus Hexan und Äthylace- tat umkristallisiert wurde, womit 3, 24 g 2-Acetylthio-3-(4-phenylthiobenzyl)-propion- säure mit Fp. = 116, 5 bis 118, 5 C erhalten wurden.
