Dicarbonsäuren führen zu Diimidazoliden. Das Nebenprodukt, Imidazol, fällt aus ; es kann abgetrennt werden, wonach das Imidazolid isoliert wird, wobei dies jedoch nicht wesentlich ist. Diese Reaktionen sind bekannt, (vgl. z. B. USA-Patentschriften Nr. 3, 079, 314, Nr. 3, 117, 126 und Nr. 3, 129, 224 undbrit. Pa- tentschriften Nr. 932, 644, Nr. 957,570 und Nr. 959, 054).
Bei der Herstellung von Kanamycin mitiii6'-StellungabgeschirmterAminogruppewahrendderVerfah- rensstufe A wird es vorgezogen, die molaren Anteile der Komponenten so zu regeln, dass ein Verhältnis von 1 Mol oder weniger als 1 Mol Acylierungsmittel pro Mol 61-abgeschirmten Amino-Kanamycin verwendet wird.
Ein grösserer Anteil an Acylierungsmittel wird auf Grund der erhöhten Anzahl von Nebenreaktionen, bei denen als Verunreinigungen Polyacylderivate entstehen, zu geringeren Ausbeuten an dem gewünschten Zwischenprodukt führen.
Wie bei den meisten chemischen Reaktionen können Temperaturen, die höher oder niedriger sind als die angegebenen, verwendet werden. Jedoch wird man bei Temperaturen, die wesentlich höher liegen als die angeführten (50 C), im allgemeinen auf Grund einer erhöhten Anzahl von Nebenreaktionen geringere Ausbeuten erhalten.
Die Verbindung (IVa), l- [Lr (-)-y-Amino-cx-hydroxybutyryl)-kanamycinA, besitzt eineausgezeictmete antibakterielle Wirksamkeit, welche derjenigen von Kanamycin A selbst überlegen ist. In nachstehenden zwei Tabellen sind die geringsten Minimalhemmkonzentrationen (MHK) von Kanamycin A und der Verbindung (IVa) (BB-K8) mit Bezug auf verschiedene grampositive und gramnegative Bakterien gezeigt, die nach dem Steers Agar-Verdünnungsverfahren (Tabelle 1) bzw. nach dem zweifachen Verdünnungsverfahren (Tabelle 2) erhalten worden waren. Bei den Versuchen gemäss Tabelle 1 wurde ein Mueller-Hinton-Agar-Medium und bei denVersuchen gemäss Tabelle 2 eine Herzmuskelbrühe verwendet.

Die obigen MHK-Resultate zeigen, dass die Verbindung (IVa) (BB-K8) Kanamycin A, insbesondere mit Bezug auf Kanamycin A gegenüber resistenten Organismen überlegen ist.
Diese Ergebnisse stimmen auch gut mit in vivo Resultaten für die drei Organismen überein, mit Bezug auf welche Kanamycin A und die Verbindung (IVa) geprüft wurden.
Die Verbindung (IVa) und Kanamycin A waren gleichfalls wirksam gegenüber Mäuseinfektionen, die durch mit Bezug auf Kanamycin A sensible Stämme von E. coli A-15119 and Staph. aureus A-9537 bewirkt waren.
Obgleich die CD-Werte (kurativeDosisbei50% vontodlictiinfiziertenMausen) furStaph. auieus A-9537 nahelegen. dass dieVerbindung (IVa) etwas weniger wirksam ist als Kanamycin A, ist dieser geringe Unterschied wahrscheinlich nicht von Bedeutung, da die Dosishöhen weit auseinander lagen (5fache Verdünnungen).
Mit Bezug auf den Kanamycin resistenten Stamm E. coli A-20520 war Kanamycin A, wie erwartet, in vivo nicht wirksam, wogegen die Verbindung (IVa) eine deutliche Schutzwirkung zeigt. DieVerbindung (IV a) war ungefähr zehnmal aktiver gegenüber diesem E. coli-Stamm, wenn sie je viermal und nicht je zweimal verabreicht wurde.

Die Verbindungen (IV) sind wertvoll als antibakterielle Mittel, Zusätze zu Futtermittel, therapeutische Mittel bei Geflügel und Säugetieren, einschliesslich Menschen, und sind insbesondere wertvoll zur Behandlung von durch grampositive und gramnegative Bakterien verursachte Infektionskrankheiten.
Bei oraler Verabreichung sind die Verbindungen (IV) als zusätzliche Behandlung bei der preoperativen Darmdesinfektion verwendbar. Auf diese Arzneimittel ansprechende aerobe und anaerobe Flora wird im Dickdarm vermindert. Bei entsprechender mechanischer Reinigung sind diese Verbindungen auch in der Dickdarmchirurgie verwendbar.
Die Verbindungen (IV) sind auch wirksam zur Behandlung von bakteriellen Infektionen, wenn sie im Bereich von ungefähr 250 mg bis ungefähr 3000 mg/Tag, verteilt in drei oder vier Dosen, verabreicht werden. Im allgemeinen sind die Verbindungen wirksam, wenn sie in einer Dosierung von ungefähr 5,0 bis 7,5 mg/kg Körpergewicht alle 12 h verabreicht werden.
DieVerbindung (IVb), l- [L- (-)-y-Amino-c < -hydroxybutyryl]-kanamycinB, besitzt eine hervorragende antibakterielle Wirksamkeit, welche derjenigen von Kanamycin A überlegen ist. In nachstehender Tabelle 4 sind die Minimalhemmkonzentrationen (MHK) von Kanamycin A und der Verbindung (IVb) (BB-K26) mit Bezug auf verschiedene grampositive und gramnegative Bakterien gezeigt. Die MHK-Werte wurden nach dem Steer- Agar-Verdünnungsverfahren (Tabelle 4) auf Agar-Medium erhalten.

Die Verbindung (IVb) (BB-K26) und Kanamycin B waren in gleicher Weise wirksam gegenüber E. coli Juhl A-15119 bei einer Dosis von 6, 3 mg/kg in Mäusen. Die Verbindung (IVb) war besonders wirksam gegenüber Kanamycin B resistenten E. coli ML-1630 bei einer Dosis von 6,3 mg/kg vs. 100 bis 400 mg/kg für Kanamycin B.
Die Verbindung (IVb) zeigte eine verglichen mit Kanamycin B etwas bessere Wirksamkeit gegenübermässig resistenten Ps. aeruginosa D15 und eine wesentlich bessere Wirksamkeit gegeniiber sehrresistenten Ps. aeruginosa D-113 (vgl. nachstehende Tabelle 5).

Die Erfindung soll an Hand von Beispielen näher erläutert werden.
Vorschrift 1 : Herstellung von L-(-)-γ-Benzyloxycarbonylamino-α-hydroxybuttersäure (VI).
L-(-)-γ-Amino-α-hydroxybuttersäure (7,4 g, 0,062 Mol) wurde einer Lösung von 5, 2 g (0, 13 Mol) Natriumhydroxyd in 50 ml Wasser zugesetzt. Zu der Lösung wurden unter Rühren tropfenweise während 1/2 h bei 0 bis 50C 11,7 g (0,068 Mol) Carbobenzoxychlorid hinzugefügt, wonach die Mischung eine weitere Stunde bei der gleichen Temperatur gerührt wurde. Das Reaktionsgemisch wurde mit 50 ml Äther gewaschen, mit verdünnter Salzsäure auf einen pH-Wert von 2 eingestellt und viermal mit je 80 ml Anteilen Äther extrahiert. Die

Der ölige Rückstand wurde in einer Mischung von 500 ml Wasser und 500 ml Butanol gelöst, wonach das Gemisch zwecks Abscheidung von unlöslichem Material abfiltriert wurde.

Einer Lösung von 1,5 g (0,014 Mol) Natriumcarbonat in 40 ml Wasser wurden 5,3 g (0,01 Mol) Dehydro- abietylammonium-L-α-hydroxy-γphthalimidobutyrat und 60 ml Äther zugesetzt. Die Mischung wurde heftig geschüttelt, bis die ganzenFeststoffe aufgelöst waren. Die Ätherschicht wurde abgetrennt. Die wässerige Lösung wurde zweimal mit 20ml Anteilen Äther gewaschen und unter vermindertem Druck auf 15 ml eingeengt. Dem Konzentrat wurden 10 ml konz. Salzsäure zugesetzt, wonach die Mischung 10 hunter Rückfluss erhitzt wurde.
Nach dem Abkühlen wurde die abgeschiedene Phthalsäure abfiltriert. Das Filtrat wurde unter vermindertem Druck eingedampft. Der Rückstand wurde in 10 ml Wasser aufgelöst und die Lösung zur Trockne eingedampft. Dieser Vorgang wurde zur Entfernung von überschüssiger Salzsäure zweimal wiederholt. Der zurückbleibende Sirup wurde in 10 ml Wasser aufgelöst und zur Entfernung einer kleinen Menge von unlöslicher Phthalsäure abfiltriert. Das Filtrat wurde auf einer Polystyrol-Ionenaustauschersäule IR-120 Rohm & Haas Comp. U. S. A.

Schwefelsäure auf 7 bis 7,5 herabgesetzt wurde.
8, 5 g Darco G-60 der Firma Atlas Powder Company, U. S. A. (Aktivkohle) wurden zugesetzt, wonach die Mischung bei Raumtemperatur 0, 5 h aufgeschlämmt wurde. Die Kohle wurde durch Filtrieren abgetrennt und mit 40 ml Wasser gewaschen. Das Waschwasser wurde dem Filtrat zugesetzt.
Das mit dem Waschwasser kombinierte Filtrat wurde mit 50 Vol. /Vol. -% Schwefelsäure auf einen PH-Wert von 2 bis 2, 6 gebracht, wobei eine grosse Menge von Kohlendioxyd abgegeben wurde. Die Lösung wurde zur
Entfernung von weiterem Kohlendioxyd unter Rühren 20 min mit dem Hausvakuum in Verbindung gelassen.
Zu der so erhaltenen Lösung wurden 8,5 g Darco G-60 hinzugefügt. Die Mischung wurde bei Raumtempe- ratur 0, 5 h aufgeschlämmt. Die Kohle wurde abfiltriert und mit 35 ml entionisiertem Wasser gewaschen. Das
Wasser wurde dem Filtrat zugesetzt, wonach es mit 50 Vol. /Vol. -% Schwefelsäure auf einen PH-Wert von 1 bis
1, 3 gebracht wurde. Dieser Lösung wurde unter starkem Rühren innerhalb von 10 min 600 bis 800 ml Methanol (3 bis 4Volumina Methanol) zugesetzt. Die Mischung wurde bei einem PH-Wert von 1 bis 1, 3 5 min gerührt, durch ein Sieb mit lichter Maschenweite von 149 u durchgeführt, 2 min gerührt und 5 min sich absetzen ge- lassen. Der grösste Teil der überstehenden Flüssigkeit wurde abdekantiert.

In the preparation of kanamycin with a 6'-position shielded amino group during process step A, it is preferred to regulate the molar proportions of the components so that a ratio of 1 mol or less than 1 mol of acylating agent per mol of 61-shielded amino-kanamycin is used.
A larger proportion of acylating agent will lead to lower yields of the desired intermediate due to the increased number of side reactions in which polyacyl derivatives are formed as impurities.
As with most chemical reactions, temperatures higher or lower than those indicated can be used. However, at temperatures which are substantially higher than those mentioned (50 ° C.), lower yields will generally be obtained because of an increased number of side reactions.

The above MIC results show that the compound (IVa) (BB-K8) is superior to kanamycin A, particularly with regard to kanamycin A, over resistant organisms.
These results also agree well with in vivo results for the three organisms for which kanamycin A and compound (IVa) were tested.
Compound (IVa) and kanamycin A were also effective against mouse infections caused by kanamycin A sensitive strains of E. coli A-15119 and Staph. aureus A-9537.
Although the CD values (curative dose in 50% of deadly infected mice) for Staph. Auieus A-9537 suggest. that compound (IVa) is slightly less effective than kanamycin A, this small difference is probably not important as the dose levels were widely spaced (5-fold dilutions).
With respect to the kanamycin-resistant strain E. coli A-20520, as expected, kanamycin A was not effective in vivo, whereas the compound (IVa) shows a marked protective effect. Compound (IV a) was approximately ten times more active against this E. coli strain when administered four times rather than twice.

The compounds (IV) are useful as antibacterial agents, feed additives, therapeutic agents in poultry and mammals including humans, and are particularly useful for the treatment of infectious diseases caused by gram-positive and gram-negative bacteria.
When administered orally, the compounds (IV) can be used as additional treatment in preoperative intestinal disinfection. Aerobic and anaerobic flora responsive to these drugs is diminished in the colon. With appropriate mechanical cleaning, these compounds can also be used in colon surgery.
The compounds (IV) are also effective for treating bacterial infections when administered in the range of about 250 mg to about 3000 mg / day divided into three or four doses. In general, the compounds are effective when administered at a dosage of about 5.0 to 7.5 mg / kg body weight every 12 hours.
The compound (IVb), 1- [L- (-) - γ-amino-c <-hydroxybutyryl] -kanamycin B, has an excellent antibacterial activity which is superior to that of kanamycin A. Table 4 below shows the minimum inhibitory concentrations (MIC) of kanamycin A and the compound (IVb) (BB-K26) with respect to various gram-positive and gram-negative bacteria. The MIC values were obtained on agar medium by the Steer agar dilution method (Table 4).

The compound (IVb) (BB-K26) and kanamycin B were equally effective against E. coli Juhl A-15119 at a dose of 6.3 mg / kg in mice. Compound (IVb) was particularly effective against kanamycin B resistant E. coli ML-1630 at a dose of 6.3 mg / kg vs. 100 to 400 mg / kg for kanamycin B.
Compared with kanamycin B, compound (IVb) showed somewhat better activity against moderately resistant Ps. Aeruginosa D15 and a significantly better activity against very resistant Ps. Aeruginosa D-113 (cf. Table 5 below).

To a solution of 1.5 g (0.014 mol) of sodium carbonate in 40 ml of water were added 5.3 g (0.01 mol) of dehydroabietylammonium L-α-hydroxy-γ phthalimidobutyrate and 60 ml of ether. The mixture was shaken vigorously until all of the solids were dissolved. The ether layer was separated. The aqueous solution was washed twice with 20 ml portions of ether and concentrated to 15 ml under reduced pressure. The concentrate was 10 ml of conc. Hydrochloric acid was added and the mixture was refluxed for 10 hours.
After cooling, the separated phthalic acid was filtered off. The filtrate was evaporated under reduced pressure. The residue was dissolved in 10 ml of water and the solution was evaporated to dryness. This process was repeated twice to remove excess hydrochloric acid. The remaining syrup was dissolved in 10 ml of water and filtered to remove a small amount of insoluble phthalic acid. The filtrate was on a polystyrene ion exchange column IR-120 Rohm & Haas Comp. USA.
